      Das Depot hat gegen Ende des Jahres noch einmal kräftig "Federn gelassen", so dass 2010 mit einer Jahresperformance von 26.7% abgeschlossen wurde - das ist dennoch ok, zumal natürlich auch in diesem Jahr einige nicht erwarteten Negativmeldungen wie z.B. bei Immunogen, der von der FDA nicht angenommene Zulassunbgsantrag von TDM1 zur Drittlinienbehandlung von metastatischem Brustkrebs, hinzunehmen waren...weshalb als Fazit für 2010 durchaus Miss Sophie zitiert werden könnte ... "The same procedure as every year".

      Und wegen der Anfrage damals -hast Du Dir inzwischen eigentlich schonmal SBS.V angeschaut?? Wenn nicht, ist kein Ding, aber Deine Meinung zur Firma, wie Insulin/Apo AI Milano hätte mich schon interessiert.

      Wieviel Recherchezeit steckst Du dann eigentlich in Deine Börsenaktivitäten?

Sollten die demnächst veröffentlichten Daten einen zulassungsantrag in 2011 rechtfertigen, dann hat die Curis-Aktie enormes Potenzial (5-6 USD)

      die gute Performance ist hauptsächlich der Dendreon-Aktie geschuldet, die zum Start des Depots 2007 als erste und für längere Zeit aus Überzeugung einzige Aktie ins Depot aufgenommen wurde --> siehe Beitrag#4 aus der allerersten Depot-Diskussion:

      In den amerikanischen Foren wird spekuliert, dass der aktuelle Kursanstieg mit einer Indiskretion bei den erwarteten "Hedgehog Pathway Inhibitor"-Daten zur Behandlung des Basalzellenkarzinoms in Verbindung steht. Da die Daten bei Roche evaluiert werden, müsste das Leck jedoch beim Basler Pharmakonzern sein - kann ich mir eigentlich nicht vorstellen, aber dies muss nicht unbedingt etwas heissen.

      ich hatte bei spectral capital so ziemlich riesen summe verloren und habe jetzt deswegen ernsthafte schwierigkeiten. kennst du biotech oder hightech aktien wo man auch auf etwas risiko gehen kann wo möglichkeiten bestehen short was dazu zugewinnen??

      Die in diesem Forum dokumentierten Aktivitäten sind das Ergebnis meiner Recherchen - darüber hinaus habe ich keine Expertise.
      Wenn du mittlerweile "ernsthafte Schwierigkeiten" hast, dann würde ich dir raten, deine Anlagestrategie zu überdenken bzw. deine Kaufentscheidungen zukünftig nur von Fakten und nicht von Absichtserklärungen wie bei spectral capital abhängig zu machen.

      Das liest sich doch nicht schlecht: Die 5 befragten Analysten haben zu Delcath ein Kursziel von USD 18 für die nächsten 12 Monate, was einem Zuwachs von derzeit 92% entspricht. Not too bad!

      ...die Komplikationen bei der Anwendung der Chemosaturation von Bedeutung. Da sowohl Melphalan als auch die Anwendung der Prozedur nicht unkritisch sind, ist die Anzahl der Todesfälle (3 von 40 Patienten) auf den ersten Blick erschreckend:

      a) Treatment related Deaths:
      --> 3/40 patients (7.5%) 3/116 procedures (2.6%)
      --> Neutropenic Sepsis (n=2) 5%, Hepatic Failure (n=1) 2.5% (95% tumor burden)

      In Anbetracht dessen, dass es sich jedoch um Patienten ohne wirkliche Alternative handelt, wird dieses Mortalitätsrisiko (3-10%) bei Melphalan von der FDA toleriert!

      zum besseren Verständnis sei noch einmal auf die klinischen Ergebnisse näher eingegangen:

      PFS ist die Abkürzung für Progression Free Survival, was soviel bedeutet wie Überleben ohne Fortschreiten der Erkrankung --> mit Delcath Chemosättigung/Melphalan 186 Tage gegenüber der Standardbehandlung mit 46 Tagen!

      52% stable disease bedeutet, dass ein Behandlungserfolg erzielt wurde, der über längere Zeit das Fortschreiten (die Progression) der Krankheit aufhalten kann; im Vergleich dazu, war dies bei der bestmöglichen zugelassenen Therapie (BAC) nur bei 27% der Studienteilnehmer der Fall!!

      Antwort auf Beitrag Nr.: 41.040.137 von Magnetfeldfredy am 15.02.11 09:20:18Die FDA hat auf alle Fälle den Status "Orphan Drug" (--> siehe unter 1.) zuerkannt, was so viel bedeutet wie die Behandluing von seletenen Erkrankungen, so dass bei den zur Zulassung eingereichten Indikationen (Augentumor und Hautmelanom) auch keine riesigen Studien erwartet werden. Wichtig erscheint mir, dass trotz der geringen Mächtigkeit der Studie, statistisch signifikante Ergebnisse (--> siehe unter 2.) erzielt wurden, die den Vorteil einer solchen Behandlung eindeutig unter Beweis stellen. Und dennoch sind deine Bedenken nicht ganz unbegründet, weil die FDA in der Vergangenheit diesbezüüglich schon mehrfach nicht den allgemeinen Erwartungen entsprochen hat.

      ...zu p=0.001
      unter wissenschaftlichen Aspekten ist ein Ereignis dann signifikant, wenn die Irrtumswahrscheinlichkeit kleiner ist als 5%, also der p-Wert kleiner ist als 0.05 (p<0.05); da die Irrtumswahrscheinlichkeit beim primären Endpunkt 0.1% (p=0.001) beträgt, ist davon ausgehen, dass die FDA die Wirksamkeit der Behandlung trotz der geringen Studiengrösse nicht beanstandet.

      ...zu Hazard Ratio = 0.301

      Was bedeutet eine derartige Hazard Ratio?

      Hierzu ein Beispiel:
      Während in der Kontrollgruppe B 50 Prozent der Patienten sterben, sind es in der Gruppe A 40 Prozent. Das Hazard Ratio (HR) beträgt in diesem Fall 0,8. Es bedeutet, dass die Patienten der Gruppe A ein um 20 Prozent geringeres Sterblichkeitsrisiko haben als die Patienten der Gruppe B. Wäre HR=1, bestünde kein Unterschied zwischen den Gruppen. Je kleiner der Wert ist, desto größer ist die Risikoabnahme durch die Behandlung A.

      Beim primären Endpunkt in der Zulassungsstudie beträgt die Hazard Ratio 0.301
      Das heisst, dass das Sterblichkeitsrisiko in der mit Chemosättigung/Melphalan behandelten Gruppe weitaus geringer.




      übrigens Jim Cramer scheint von Delcath überzeugt zu sein:

      Hut ab, Du bist ein wahrer Biotechkenner mit absloutem Fachwissen, danke für die ausführlichen Erklärungen, super!;)

      Ich halte Delcath auch für absolut spannend, ja sogar für einen Game-Changer in Sachen "seperate Chemobehandlung" und habe mir 1000 Stück zugelegt, schon vor einem Jahr als die Aktie bei EUR 12,.. stand.
      Ich habe damals schnelle 1.000 EUR gemacht und bin jetzt wieder eingestiegen!

      Jedoch kann man der FDA überhaupt nicht "trauen" und habe in letzter Zeit viel gelitten mit Arena und Mannkind!

      Ich bin noch in SPPI eingestiegen und halte nach wie vor eine kleine Position Dendreon!

      Was hälts Du von SPPI?

      Es ist jedoch ein fundamentaler Unterschied, ob ein Medikament zur Diätunterstützung, also zur Gewichtsreduktion, oder aber zur Behandlung einer finalen Krebserkrankung eingesetzt wird. Im Falle der Diätpillen war und bin ich eher skeptisch.


      Ich bin noch in SPPI eingestiegen und halte nach wie vor eine kleine Position Dendreon!

      Zu Zeiten von Satraplatin wusste ich über Spectrum Pharmaceuticals (SPPI) etwas Bescheid, weil dies im Dendreon-Forum sehr kontrovers diskutiert wurde. Satraplatin als systemisch wirkendes Krebstherapeutikum "nach altem Rezept" konnte mich damals nicht überzeugen. Wie es allerdings um die Pipeline bestellt war und ist und somit um die Aussichten der Firma, weiss ich nicht.

      Bei Dendreon halte ich ebenfalls noch eine kleine Position von 10% des ursprünglichen Bestandes. Werde demnächst auch hier im Musterdepot reduzieren. Habe bereits einige neue Kandidaten auf der watch list.

      Eine Frage hätte ich noch bzgl. Curis an Dich: Wie hoch schätzt Du die Chancen auf gute Phase II Ergebnisse für deren Hautkrebsprogramm mit Roche nachdem ja im Juli 2010 Phase II für Darmkrebs scheiterte?

      Zugegebenermaßen wurde ich vom Ruf eines Börsenbriefs drauf aufmerksam, trotzdem scheint die Story sehr interessant zu sein.

Delcath’s innovative device involves the use of melphalan hydrochloride to treat metastatic melanoma of the liver (melanoma that has spread to the liver). Melphalan is typically administered orally or intravenously for multiple myeloma, ovarian cancer and occasionally malignant melanoma. However, administered levels required to treat melanoma were not effective, and earlier investigations found other applications for the drug.

      Their novel device is in the form of their device/drug combination termed a “chemosaturation system”. Their proprietary system, as best described in this video, basically involves isolating the organ’s (liver for this application) return blood flow to the body by inflatable balloons placed in the vena cava.

      The mephalan hydrochloride solution is directly injected into the artery supplying blood to the liver (the hepatic artery) to deliver a dose up to 100 times more concentrated than traditional chemotherapy would have. Then the return blood is rerouted after treatment through a filter device to remove the mephalan hydrochloride, and then it is returned to the patient’s body via a catheter in the jugular.

      After treatment, the catheters are simply removed, balloons deflated and the patient is prevented from having his entire biologic system have to deal with the side effects often associated with chemotherapy. The targeted organ containing the malignant melanoma meanwhile received a substantial dose thereby increasing efficacy while maintaining a safety profile more palatable to the patient (and the FDA).       Delcath Systems applied for an NDA for their device on December 22, 2010 with a request for priority drug review. The data in it exceeded their primary endpoints as noted here: "The FDA told us what result they would want to see" to approve PHP – an average duration of hepatic progression-free survival of 7.7 months in PHP patients and an average 4.0-month survival in control patients, Dr. Nutting said in an interview. “The results exceeded that," he noted, with an average hepatic progression-free survival of 245 days (8.2 months) in the patients randomized to initial PHP and an average of 49 days (1.6 months) in control patients who received best alternative care. The hazard ratio for the PHP patients compared with the controls for this primary end point was 0.301 (P =.0001).”

      The typical FDA NDA acceptance deadline for this type of application is 60 days, though it may take a little longer with their deadlines being somewhat later this previous year. This puts the NDA acceptance with priority review at sometime from February 22nd to the end of the month. If given priority review, we should hear back from the FDA for its decision on whether to approve the novel device sometime in June.

      Not to be forgotten in all the attention Delcath is getting for its submission to the FDA here in the US, on December 6 they submitted their CE Mark Technical File for marketing in the European Union. Per their CEO, Eamonn Hobbs, they expect a decision for CE Mark approval sometime in mid 2011 also. June and July could be an exciting time for Delcath Systems!

      2010 marked a year of substantial volatility of the DCTH stock with the stock trading from 5.1 to well over 16 dollars per share. Current market cap of the company per Google Finance is about $469 million, which could be considered low for a company with a device that if approved for its current application request for metastatic melanoma of the liver could be generating good revenue over the next few years.

      However, many other trials are likely being planned for other application such as other liver cancers as well as cancers of other organs that may be isolated from the rest of the human body and treated with the Delcath system. This sort of current application as well as potential applications could very well be drawing attention from big Pharma as a merger or acquisition target. Actually, there were rumors back in October 2010 pertaining to Delcath being a takeover target for Bristol-Meyers (BMY).

      While I don’t give much credit to these types of rumors, it is interesting to note that the stock rose over 12% on these rumors. Additionally, Jim Cramer mentioned on his February 3rd program’s “Lightning Round” pertaining to Delcath that “This is in the sweet spot. Companies like these are getting bought every day. I'm going to say that I like your stock.”


      2011 looks to be a promising and exciting year for a company on the verge of greatness or on the verge of defeat. I need to review the phase III data a little more closely to give an opinion for FDA approval in June of this year. However, I have viewed it enough to give an opinion on the NDA acceptance with priority review. I believe Delcath Systems will have their NDA accepted sometime the last week of February of 2011 with the request for priority review. This will cast DCTH into the spotlight as opinions and rumors will circulate at a feverish pace pertaining to buyout, FDA approval and company worth.

      Regardless of the FDA’s NDA acceptance, there is still the Mark CE decision also coming at about the same time as the FDA decision so there are two chances for this company to succeed and start marketing its novel device. Essentially, DCTH should see catalyst after catalyst after catalyst this year and most of it in the first half of the year. Investors in for the “long haul” have the potential for real gains meanwhile traders riding the dips and spikes will have an exciting time to say the least.

      This brings me to one big question, why with all the potential here is the short interest in the stock so high?! With a short interest of almost 21% and over 13 days to cover, per this article, somebody’s going to be getting concerned fairly soon now. Will it be the “longs” or the “shorts” that will come out on top with DCTH? Hopefully, the ones coming out on top will be the patients and healthcare providers that need another weapon in their arsenal against an unforgivable enemy, cancer.

      die Nachricht heute von Delcath über die Zertifizierung sehe ich als sehr sehr gut denn in Europa wird Delcath als Medizingerät zugelassen und nicht als Medikament, sehe ich das richtig?
      Die Zulassung als Medizingerät hat keine so hohen Hürden wie die Medikamentenzulassung, das weis ich da ich selber Medizinprodukteberater für Magnetfeldsysteme bin!

      die FDA verlangt bisher keine neuen Studiendaten, vielmehr geht es um den Validierungsprozess von Produktion, Produkt und Sterilisation (eher kleinere Peanuts) sowie um zusätzliche Daten zur Sicherheit - damit dürfte die Produktsicherheit, also die Anwendung der Sättigungstechnologie unter Melphalan, gemeint sein --> Mortalitätsrate in der Zulassungsstudie 7.5% (das ist wohl die Knacknuss!)

      Here is how they have done:

      Cepheid (CPHD): Up 168% from $8.83 to $23.65

      Cerus (CERS): Up 261% from $.86 to $3.11

      Curis (CRIS): Up 127% from $1.33 to $3.02

      Illumina (ILMN): Up 122% from $31.10 to $69.04

      Questcor (QCOR): Up 202% from $5.06 to $15.29

      Do I have your attention? All five of these stocks have legs but the run has been so great and grand for some of them I have modified the list for 2011. And, since the market has been kind to you the past twenty seven months, these are all very high risk, stop your heart stocks facing major binary events that could make or break the stock

      Cerus (CERS): Cerus developed and markets the INTERCEPT system. It's a blood pathogen inactivation system to purify blood platelets. It is the only game in town and approved in most large European countries, it has been held up in the U.S. by an incredibly ridiculous FDA staff person even though it hit the primary end points in its pivotal phase III trial and gets grants from the U.S. military. INTERCEPT kills HIV, XMRV and a zillion other pathogens in the blood, making the blood platelet supply donor independent.

      Cerus is working on developing an INTERCEPT system for whole blood and the program is on schedule and funded with a recent stock offering. Management has bumbled working with the FDA – they don’t fight back - but this will be fixed over time. There is no scientific or product risk in the stock and it is the number one medical products company pick for 2011 by Stephens & Company. The target price in 1-3 years is $14. Downside risk is market risk. I own the stock.

      Curis (CRIS): This little gem has developed a series of cancer treatments in various stages of clinical trials based on a technology that disrupts intercellular signaling in the Hedgehog pathway. Disrupt communications and you disrupt cell duplication, the foundation of tumor growth. More than twenty trials with Genentech/Roche and the National Cancer Institute are underway. This year, Genentech will probably report out results of a basal cell carcinoma trial for skin cancer. The company has said it will go from this mid-Phase trial to an application for approval if the results are strong enough. One success means a volcanic eruption in the stock as it will prove their core technology is viable as a platform for cancer treatments. A failure could put their entire program – and the company – in jeopardy. I believe they will be. Upside is $40-$44, downside $.50. I own the stock.

      ...mit dem Risiko (Kurs 50 Cent) bin ich einverstanden, die Chance ist jedoch mehr als optimistisch, um nicht zu sagen mittelfristig unrealistisch. Bei ca. 80 Mio Aktien würde ein Kurs von USD40 einer Marktkapitalisierung von über USD3 Mrd. entsprechen. Da Curis lediglich mit Meilensteinzahlungen und einstelligen Lizenzgebühren am Erfolg von Vismodegib beteiligt sein wird und Roche in der Spitze Umsatzerwartungen unter 2 Milliarden bekanntgibt, wird Vismodegib alleine sicherlich nicht für derartige Kursphantasien ausreichen, und da die Pipeline nicht "aus allen Nähten platzt" ...

      Von Roche/Genentec sind demnächst die Ergebnisse einer Pivotalstudie zu Vismodegib (ursprünglich von Curis, jedoch an Genentech auslizenziert) zur Behandlung des Basalzellkarzinom zu erwarten und es deutet einiges daraufhin, dass diese eine Marktzulassung auf Anfang 2012 ermöglichen.

Neu ist jedoch, dass bei Roche angeblich eine Markteinführungg gezielt vorbereitet wird. Zulassungsanträge (FDA, EMA, Swiss Medic) sollen möglicherweise noch 2011 eingereicht werden!!

      We selected 33 patients with metastatic or locally advanced basal-cell carcinoma to receive oral GDC-0449 at one of three doses; 17 patients received 150 mg per day, 15 patients received 270 mg per day, and 1 patient received 540 mg per day. We assessed tumor responses with the use of Response Evaluation Criteria in Solid Tumors (RECIST), physical examination, or both. Molecular aspects of the tumors were examined.

      Results
      The median duration of the study treatment was 9.8 months. Of the 33 patients, 18 had an objective response to GDC-0449, according to assessment on imaging (7 patients), physical examination (10 patients), or both (1 patient). Of the patients who had a response, 2 had a complete response and 16 had a partial response. The other 15 patients had either stable disease (11 patients) or progressive disease (4 patients). Eight grade 3 adverse events that were deemed to be possibly related to the study drug were reported in six patients, including four with fatigue, two with hyponatremia, one with muscle spasm, and one with atrial fibrillation. One grade 4 event, asymptomatic hyponatremia, was judged to be unrelated to GDC-0449. One patient withdrew from the study because of adverse events. We found evidence of hedgehog signaling in tumors that responded to the treatment.

      Gegenüber konventionellen Chemotherapeutika ist Vismodegib fast schon harmlos bspw. ist die akute Toxizität im Tierversuch äusserst gering (LD50, oral, Ratte > 2000 mg/kg) und dennoch ist durch die hohe Spezifität der therapeutische Effekt anscheinend enorm. Aufgrund der geringen Toxizität ist Vismodegib natürlich auch als potentielles Medikament für Kombinationstherapien mit anderen Wirkstoffen von Interesse.

      Because of its low toxicity and specificity for the Hh pathway, this drug has potential advantages compared with conventional chemotherapy, and may also be used in combination treatments.

      Obwohl die Ergebnisse der Pivotalstudie (Pivotal heisst, dass man zuvor eine These aufgestellt hat, welche es zu beweisen gilt und die Studie im Beweisfall somit für eine mögliche Zulassung Verwendung findet) weder von Roche und Genentech noch auf einem wissenschaftlichen Kongress veröffentlicht wurden, kann man diese überaus positiven Ergebnisse nereits im web aufspüren:

      Eine 25-fache (!) Reduktion der Rate bei den Basalzellkarzinomen, wobei die doppelt-verblindete Studie aus ethischen Gründen bereits vorzeitig beendet wurde - bei einer deratig eindeutigen Wirksamkeit ist eine Plazebo-Verabreichung an Studienteilnehmer nicht gerechtfertigt! Das müsste der Curis-Aktie doch eigentlich Flügel verleihen --> kurzfristiges Ziel/Erwartung bis Mitte 2011 ca. 4 bis 5 USD.

      NEW ORLEANS - An oral inhibitor of the hedgehog pathway proved effective for the treatment and prevention of basal cell carcinomas in patients with basal cell nevus syndrome in a phase II study.

      The randomized double-blind trial was halted early for ethical reasons because patients assigned to vismodegib, formerly known as GDC-0449, demonstrated a 25-fold reduction in the rate of new basal cell carcinomas (BCCs), compared with placebo-treated controls, Dr. Jean Y. Tang reported at the annual meeting of the American Academy of Dermatology.

      "We're really excited by these results as dermatologists because we're sick of just chopping up these poor patients' skin, and we're excited to offer something better," said Dr. Tang of Stanford (Calif.) University.

      Basal cell carcinomas are most commonly treated by surgical excision. That's a traumatic therapy for patients with the genetic disease known as basal cell nevus syndrome (BCNS) because they experience many hundreds of BCCs during their lifetime.

      The phase II study involved 41 patients at three medical centers who were randomized 2:1 to vismodegib at 150 mg once daily or placebo for a planned 18 months.

      At baseline, the participants collectively had 2,000 existing BCCs. During a median 8 months of follow-up before halt of the study, the patients developed 694 new BCCs. The incidence of new BCCs was 0.07 cancers per month in the vismodegib arm, compared with 1.74 per month in controls.

      In addition to preventing new skin cancers, treatment with vismodegib typically began shrinking existing tumors within the first month or two. Sixty percent of biopsied tumors that appeared clinically clear showed histologic clearance.

      A side benefit seen after several months of vismodegib therapy was the "remarkable" clearance of the palmar and plantar pits which are a characteristic part of BCNS, Dr. Tang observed.

      "Our patients are really struck by this. They've lived with this condition all their lives and now they can comfortably shake the hands of strangers," she explained.

      Side effects were mild to moderate but sufficiently frequent that the investigators believe 18 months of daily therapy is unlikely to be tolerable. As patients from the placebo arm of the halted phase II study are crossed over to vismodegib, they are being dosed on an intermittent regimen consisting of repeated cycles of 3 months on daily therapy, then 2 months off, with the aim of minimizing side effects.

      Eighty-three percent of patients on vismodegib experienced taste loss, typically starting in the first month. Seventy percent reported muscle cramping, again beginning in the first month or two. Half of treated patients experienced significant hair loss which occurred months later than the other side effects. Thirty percent of vismodegib-treated subjects dropped out of the study, because of adverse events. All side effects were completely reversed 1 month after stopping therapy.

      Thus far, no tumor resistance has been seen. Patients who stopped the drug after a few months have not experienced a return of tumors to baseline size or number during the next 6 months.

      Vismodegib is an oral small molecule antagonist of the smoothened receptor. In binding to smoothened, the drug turns off the hedgehog-signaling pathway, which is inappropriately activated in patients with BCNS, leading to tumor cell growth and proliferation.

      Dr. Tang and coworkers became interested in evaluating vismodegib in patients with BCNS after other investigators reported the drug to be effective in a study involving 33 non-BCNS patients with locally aggressive or metastatic BCCs (N. Engl. J. Med. 2009;361:1164-72).

      Remaining questions involving treatment of BCNS patients include the best vismodegib dosing regimen, and whether the tumors will eventually come back after the drug is stopped, she noted.

      Dr. Tang is considering conducting a study of vismodegib in non-BCNS patients with a large BCC burden. One audience member suggested including patients with post–radiation therapy BCCs. "I have postradiation patients with 400 BCCs," he said.

      Another audience member, a dermatologist involved in the BCNS Network, commented that the buzz circulating among members of the patient support group is focused mainly on the improvement in quality of life resulting from vismodegib therapy.

      Dr. Tang agreed. "The patients say that this medication has completely changed their lives," she said. "These patients have terrible phobias after undergoing tons and tons of biopsies and surgeries, so they're really incredibly grateful for this drug. Obviously there are some side effects, but I think the positive effects of this drug on quality of life cannot be adequately quantitated."

      The investigator-initiated phase II study was sponsored by Genentech. Dr. Tang reporting having no commercial financial relationships.

Study participants received 150 mg vismodegib once daily until disease progression. The primary endpoint of the study was overall response rate (tumor shrinkage) as assessed by independent reviewers.       ZURICH, March 21 (Reuters) - Roche's (ROG.VX) new cancer drug vismodegib could help patients with a potentially fatal skin cancer type, positive mid-stage trial results showed, going some way to lift confidence in the drugmaker after a rough 2010.

      Roche is struggling to find its feet again after a series of product setbacks last year unsettled investors and lopped nearly one fifth from the value of the company.

      But the group has had a better run of late and this latest success, albeit a fairly small one, should help to boost sentiment and remind investors of Roche's strength in oncology.

      At 1257 GMT, Roche stock was trading 1.4 percent higher, outperforming a 0.8 percent rise in the European healthcare sector index .SXDP and cross-town rival Novartis (NOVN.VX).

      Roche, the world's largest maker of cancer drugs, said vismodegib shrank tumours in patients with advanced basal cell carcinoma, meeting the primary endpoint of a Phase II trial.

      The group, which is developing the drug with Curis (CRIS.O), said the most common adverse side effects included muscle spasms, hair loss, weight loss, fatigue and nausea.

      Analysts at Deutsche Bank estimate the total market is unlikely to exceed $500 million and see peak sales of 300 million Swiss francs ($331.7 million).

      Basal cell carcinoma (BCC) is the most common type of skin cancer in Europe, Australia and the United States and there are about 2 million cases worldwide. It is generally thought to be curable when the cancer is restricted to a small area of the skin.

      But in a small group of people, the cancer can advance further into the skin, bones or other tissues if it is left untreated or does not respond to treatment.

      BCC can advance or spread to other parts of the body in a small proportion of patients, estimated at less than 1 percent, and become difficult to treat or life-threatening, Roche said.

      The drug is a so-called Hedgehog Pathway Inhibitor. The Hedgehog signalling pathway is important in regulating proper growth and development in the early stages of life and then becomes less active in adults.

      Roche hat bekanntgegeben, dass der primäre Endpunkt in der Pivotalstudie der Phase 2 zur Behandlung des Basalzellkarzinoms mit Vismodegib erreicht wurde. Detailliertere Ergebnisse würden allerdings erst auf einem medizinischen Kongress veröffentlicht - man darf gespannt sein. Jedenfalls geht man davon aus, in diesem Jahr noch mindestens einen Zulassungsantrag für Vismodegib einzureichen (vermutlich zuerst bei der FDA für die US), wobei durchaus auch vorstellbar ist, dass dies fast zeitgleich mit einem Zulassungsantrag bei der EMA für ganz Europa erfolgen wird.

      Bis Mitte Jahr erwarte ich Kurse oberhalb 4 USD - Good hunting!

      Curis scheint nun nach unten bestens abgesichert zu sein, weil noch knapp 100 Mio USD von insgesamt 115 Mio USD (115 - 18 = 97 Mio USD) an Meilensteinzahlungen für GDC-0449 bzw. Vismodegib von Roche/Genentech möglich sind sowie 5 bis schätzungsweise 8% (mid-to-high single digit royalty) an Lizenzgebühren auf den Umsatz fällig werden.
      Es ist damit zu rechnen, dass Roche noch in 2011 den Zulassungsantrag zur Behandlung des Basalzellkarzinoms in den US bei der FDA und in Europa bei der EMA einreichen wird, so dass für Curis reichlich Geld fliessen sollte.

      Roche, Curis Drug Prevented Lesions, Reduced Tumor Size in Trial

      Business Exchange Buzz up! Digg Print Email Roche Holding AG (ROG) and Curis Inc. (CRIS)’s experimental drug shrank tumors and prevented new lesions in a clinical trial of skin-cancer patients with a rare inheritable disease.

      Patients given the medicine, known as vismodegib or GDC- 0449, had fewer basal cell carcinomas than those who received a placebo, according to a statement from the American Association for Cancer Research conference in Orlando, Florida. The size of existing tumors “decreased significantly” for those getting the drug, while carcinomas were “essentially unchanged” for patients who got placebo, researchers said.

      The 41 patients in the trial had basal cell nevus syndrome, a disease that leads to a higher risk of skin cancers. Side effects, including loss of taste, muscle cramps and weight loss, caused 20 percent of patients to discontinue the drug.

      Curis Announces Investigator-Initiated Phase II Study Results Presented at 102nd AACR Annual Meeting 2011
      Interim data show effect in prevention and treatment of BCC in Basal Cell Nevus Syndrome Patients

      A Data Safety Monitoring Board (DSMB), tasked with reviewing the unblinded results from an interim analysis of 29 patients who completed an average of 6 months of drug treatment, recently recommended to end the placebo arm of the trial due to statistically significant differences between the two groups, in order for all of the patients enrolled in the trial to receive vismodegib treatment.

      The DSMB's analysis revealed that vismodegib reduced the rate of new BCCs from an average of 1.74 BCCs per month in the placebo group to 0.07 in the vismodegib group (p=<0.0001). Vismodegib also reduced the size of existing BCCs (-24cm vs. -3cm placebo, cumulative diameter, p=0.006). Some patients achieved near complete remission with no BCC developing resistance during this period of time on trial.

GDC-0449 zur Behandlung des Basalzellkarzinoms einreichen wird - erfahrungsgemäss zuerst bei der FDA für die US und anschliessend wohl auch bei der EMA für Europa. Dann dürften Meilensteinzhahlungen fällig werden, welche der Curis-Aktie Auftrieb geben sollten.

      es wurden folgende Nebenwirkungen festgestellt: Haarausfall, Verlust des Geschmackssinnes und Muskelkrämpfe - dies lässt ihn später zu der Schlussfolgerung kommen, dass die Risisken die Anwendung das Therapeutikum im Anfangsstadium wohl nicht rechtfertigen, jedoch auf alle Fälle zu einem späteren Zeitpunkt.


      Zur Studie selbst sagt Epstein Folgendes:
      "...we randomized 41 patients in a 2:1 ratio to drug to placebo over the course of about 15 months. This was th number calculated to give a statisticly significant result as it turned out it was far more than we needed ...but the truth is that we could see at 1 month in most patients on the drug a significant decrease of the tumors..."

      wie allgemein üblich wurde die Studiengrösse im Voraus so berechnet, dass ein statistisch signifikantes Ergebnis möglich ist. Es hat sich jedoch gezeigt , dass diese Grösse nicht einmal nötig gewesen wäre, da sich bei den meisten Patienten, die das Medikament (Verum) erhielten, bereits nach einem Monat eine signifikante Wirkung (Tumorverkleinerung, geringe Zuwachsrate) eingestellt hat.



      Die interessante Komponente ergibt sich daraus, dass Vismodegib noch bei vielen anderen Indikationen, teilweise sogar fremdfinanziert vom National Cancer Institute (NCI) oder anderen Sponsoren, in der Klinik getestet wird:

Dendreon ist wohl kurz- bis mittelfristig bei CURIS nicht möglich, weil Vismodegib von Genentech (also Roche) einlizenziert wurde und somit Curis lediglich mit Meilensteinzahlungen (noch 97 Mio USD sind möglich) sowie Lizenzgebühren auf den Umsatz (mid to high single digit = würde ich mit 5 bis 8% auf den Umsatz interpretieren) am Erfolg von Vismodegib partizipieren wird. Bei Dendreon ist die Phantasie weitaus grösser, da Provenge nicht auslizenziert wurde und dessen weltweite Rechte sich im Besitz von Dendreon befinden.

      Curis wäre dann eine 2. Dendreon, wenn CUDC-101 - "CUDC-101 is the lead drug candidate from Curis’ network-targeted cancer programs and is designed to inhibit histone deacetylase (HDAC), epidermal growth factor receptor (EGFR) and epidermal growth factor 2 (Her2)" - sich als wirkungsvolles Krebstherapeutikum erweisen sollte. Dann allerdings wird es beim Kurs kein Halten geben. Momentan denke ich, dass die Aktie nach unten durch die erwiesene Wirksamkeit von Vismodegib gut abgesichert ist und Kurse auch oberhalb 5 USD möglich sein sollten. Die ganz grosse Phantasie ergibt sich jedoch wie bereits erwähnt durch andere Kandidaten. Dass die Curis-Forscher ihr Handwerk verstehehn, haben sie allerdings bereits unter Beweis gestellt.

      Roche gibt positive Phase-II-Ergebnisse für trastuzumab emtansine (T-DM1) bei fortgeschrittenem HER2-positivem Brustkrebs bekannt
      Roche (SIX: RO, ROG; OTCQX: RHHBY) gab heute Topline Ergebnisse der ersten randomisierten Studie mit trastuzumab emtansine (T-DM1) bei HER-2-positivem metastasierendem Brustkrebs bekannt.

      Die Phase-II-Studie TDM4450g verglich die Gabe von trastuzumab emtansine (T-DM1) mit der Kombination von Herceptin (trastuzumab) und Chemotherapie (docetaxel) bei zuvor noch nicht behandelten Patientinnen. Die Resultate zeigten, dass bei den mit trastuzumab emtansine (T-DM1) behandelten Patientinnen in dieser Studie die Erkrankung signifikant länger unter Kontrolle war (progressionsfreies Überleben, PFS) und weniger Chemotherapie-typische Nebenwirkungen auftraten.

      „Diese ermutigenden Daten bestätigen unser derzeit laufendes Entwicklungsprogramm für trastuzumab emtansine (T-DM1) bei zuvor unbehandeltem HER2-positivem metastasierendem Brustkrebs,“ so Hal Barron, Chief Medical Officer und Leiter Global Development bei Roche. „Die Behandlung mit trastuzumab emtansine (T-DM1) ist neuartig und hat aufgrund ihrer Wirksamkeit und ihres günstigen Sicherheitsprofils das Potenzial, die Therapieergebnisse bei Patientinnen mit HER2-positivem Brustkrebs zu verbessern.“

      Die Daten der Studie TDM4450g werden zur Präsentation auf einem medizinischen Fachkongress eingereicht.

      Eine frühere Analyse dieser Studie, die 2010 auf dem 35. Kongress der Europäischen Gesellschaft für medizinische Onkologie (ESMO) vorgestellt wurde1, zeigte bei Patientinnen mit einer Nachbeobachtungszeit von mindestens 4 Monaten vielversprechende Ergebnisse die Tumorschrumpfung (Gesamtansprechrate, ORR) betreffend. Darüber hinaus zeigte die Studie, dass trastuzumab emtansine (T-DM1) die typischen Nebenwirkungen, die mit der herkömmlichen Chemotherapie verbunden sind, signifikant verringerte.

      Zu Vismodegib sind Umsatzprognosen nur sehr schwer möglich, da die Anzahl der möglichen Indikationen gross ist. Ich hab jedoch eine Roche-Grafik im Hinterkopf, in welcher potenzielle Aktivsubstanzen nach "Wahrscheinlichkeit auf Zulassung" und "Umsatzpotenzial" klassiert werden.

      Vismodegib --> "gross" "< 2 Milliarden CHF/a" - also es wird mit einer Zulassung gerechnet bei einem Umsazpotenzial pro Jahr kleiner 2 Milliarden CHF - die Unschärfe beim Umsatz ist eindeutig. Nebst Meilensteinzahlungen (noch USD 97 Mio sind möglich) erhält Curis Lizenzgebühren gestaffelt von 5 bis 8% ("mid to high single digit") auf den Umsatz.

      Vielen Dank für eure Infos. Das Umsatzpotentzial scheint bei Curis/Vismodegib schwer abschätzbar zu sein. Aber ich denke, in Anbetracht der noch immer niedrigen Bewertung und wie von euch schon geschrieben, der relativ guten Absicherung nach unten bzgl. der noch offenen Meilensteinzahlungen, kann man trotz Kursanstieg hier eine Position riskieren.

      Was haltet Ihr von Vical? Wurde letztes Jahr etwas gepuscht (mit dem typischen Ausgang :laugh: ) Aber die Pipeline sieht mit Allovectin-7 ganz interessant aus.

      Kommentar:
      Der Depot-Neuling "Seatlle Genetics" ist neben Immunogen der Marktführer in der "Linker-Technologie" - mit Linker-technologie werden ADCs (steht für Antibody-Drug-Conjugate) hauptsächlich für die Therapie von unterschiedlichsten Krebsarten entwickelt: Der Antikörper (antibody) findet über Rezeptoren die Krebszellen (die Krebszellen unterscheiden sich von anderen Zellen über Rezeptoren auf deren Oberflächen) und schleust ein Zellgift in die Krfebszelle ein, welches über einen sogenannten "Linker" an den Antikörper gebunden ist. Der Clou der Geschichte ist, dass das Zellgift systemisch (also in der Blutbahn) nicht freigegeben wird sondern fest an den Antikörper gebunden ist. Erst wenn der Antikörper über die den Krebszellen charakteristischen Rezeptoren in die Zelle eingeschleust wurde, wird die Brücke oder der Verknüpfer (Linker) enzymatisch gelöst und das Toxin freigegeben, welches die Zelle zerstört.
      Seattle Genetiocs hat in letzter Zeit sehr viele Abkommen mit den Pharmagrössen abgeschlossen, weshalb deren Linkertechnologie im Allgemeinen mehr Vorschusslorbeeren als derjenigen von Immunogen eingeräumt werden. Allerdings ist auch zu lesen, dass Immunogen wesentlich höhere Gegenleistungen einfordert und möglicherweise deswegen nicht immer erste Wahl sein soll. Ein grosses Plus für Immunogen ist zudem, dass mit TDM-1 bereits ein potenzieller Blockbuster bereit steht, der das zukünftige Geschäft von Immunogen finanzieren könnte. Für Seattle Genetics spricht die eigene Pipeline, wobei ein potenzieller Wirkstoff zur Behandlung verschiedenster Krebsarten (Brentuximab Vedotin) in der Entwicklung sehr weit fortgeschritten ist und zur Zulassung ansteht. Sehr wahrscheinlich wird der Markt beiden Linker-Experten die Möglichkeit geben, sich erfolgreich am Markt zu platzieren.

      ich habe wirklich viel Respekt vor Deiner Fähigkeit mit den Biotech-Trades. Ich habe jedoch festgestellt, dass der enorme Zuwachs von Deinem Depot in der Vergangenheit im Großen und Ganzen Dendreon zu verdanken ist. D.h., wenn Dendreon nicht so wäre, wie es wirklich ist, dann wäre dieser Zuwachs nicht denkbar. Wenn ich mir Dein Depot anschaue, macht Dendreon fast die Hälfte des Depots aus, was auch bedeutet, ein potentieller Zuwachs hängt größtenteils von Dendreon ab. Nun stellt sich die Frage, siehst Du wirklich in Dendreon noch Potenzial nach oben?

      ONLINE FIRST: ‘Life-Changing’ Hedgehog Pathway Disrupter for Advanced Basal-Cell Carcinomas Continues to Show Promise

      Orlando, FL--Clinicians all too familiar with disfiguring basal-cell carcinomas were very encouraged by the results of the targeted drug GDC-0449 reported here at the American Association for Cancer Research Annual Meeting.



      "This is a real step forward," said Daniel D. Von Hoff, MD, Physician in Chief for TGen, the Translational Genomics Research Institute. "It's very important for doctors to know there's something out there for patients with advanced basal-cell carcinoma."



      Dr. Von Hoff moderated a news conference designed to showcase AACR’s increased focus on clinical trials expected to have an impact on patient care. Ervin Epstein Jr., MD, presented an abstract of a Phase II study he led of GDC-0449, also known as Vismodegib, which Dr. Von Hoff has also been studying.



      Basal-cell carcinoma affects approximately one million Americans every year, the vast majority of whom have early-stage local tumors that are best treated surgically. Yet for a small number of patients--0.1% to 0.5% of the US population--basal-cell carcinomas metastasize or become so advanced locally that excision is nearly impossible.



      These tumors have had no effective treatment, and the median survival time for patients with metastatic disease is approximately eight months. GDC-0449 is the first targeted drug for advanced basal-cell disease, and works by inhibiting the hedgehog pathway, which is implicated in nearly all cases of basal-cell carcinoma.



      The drug also has shown promise in clinical trials for the rare genetic disorder basal-cell nevus syndrome, or Gorlin syndrome, which is characterized by continual development of hundreds and sometimes thousands of lesions over the body. The standard of care has been numerous resections, often leading to widespread scarring.



      Surgery Averted in Treated Patients

      Gorlin syndrome was the focus of the study led by Dr. Epstein, a senior scientist at Children's Hospital of Oakland Research Institute in California. Between late 2009 and early 2011, he and his colleagues treated 24 subjects with 150 mg a day of oral GDC-0449 and 12 with a placebo in a randomized, double-blind trial.



      The patients in the treatment arm developed 0.07 new carcinomas per month compared with 1.74 in the placebo arm, and the drug also shrank existing carcinomas.



      The aggregate tumor size decreased by 24 cm in treated patients and 3 cm in those taking the placebo. None of the treated patients required resections during the study, which some normally would have had several times a month, Dr. Epstein noted.



      The results were so promising that the data safety monitoring board stopped the placebo arm midway through the study.



      "[GDC-0449] works in large tumors; it works in smaller tumors,” Dr. Epstein said. “It works both by shrinking basal-cell carcinomas and by stopping the development of new ones."



      Slides he displayed of pictures taken of patients over a period of five to nine months showed dramatic improvements in the appearance of individual lesions and in the number of lesions.



      "We usually can see shrinkage in one to two months," said Jean Tang, MD, PhD, the first author of the study, a dermatologist at Stanford University School of Medicine, contacted after the meeting for this article. "The maximum benefit is close to nine months to a year.



      “This drug is absolutely life-changing for basal-cell nevus patients."



      Also contacted after the meeting, Vernon Sondak, MD, Chair of the Department of Cutaneous Oncology at H. Lee Moffitt Cancer Center, who has also treated Gorlin-syndrome patients involved in GDC-0449 trials, said, “We've seen a lot of dramatic results. There's going to be a lot of interest in whether patients could have cancers prevented."



      Adverse Effects Prevalent, But Minor

      Dr. Von Hoff's Phase I trial, the results of which were published in 2009 in the New England Journal of Medicine centered on the somewhat more common cases of locally advanced basal-cell carcinoma. Of the 33 patients in the study, 18 responded; 11 had stable disease and four had carcinomas that progressed. An impressive 50% of patients with metastatic disease also responded. The study was published Sept. 17, 2009, in The New England Journal of Medicine (http://www.nejm.org/doi/full/10.1056/NEJMoa0905360), the abstract of which was reported at the 2008 AACR Annual Meeting (OT, 6/25/08 issue).



      The patients in that trial, as with the ones in the trial by Drs. Tang and Epstein, had mild side effects, 90% of which were Grades 1 or 2. The most common was loss of taste, which 83% of treated subjects reported. That was associated with weight loss, which affected half the patients.



      Another prevalent adverse effect was muscle cramps, reported by 67% of patients. Some patients also had minor hair loss.



      None had Grade 5 events or dose-limiting toxic effects, and only one patient dropped out of the trial. Some of Von Hoff's patients have been taking the drug for more than three years, he noted.



      Still, some were bothered enough by side effects to take a brief hiatus from the drug--"If you stop it, the problems go away," Dr. Von Hoff said.



      "Treatment results are so dramatic that I doubt the side effects will be a real hindrance. Most people didn't want to stop, especially if [the carcinoma] was in a cosmetically sensitive area."



      Dr. Epstein's Gorlin-syndrome patients seemed more averse to the side effects. Twenty percent dropped out within a year of enrolling in the trial.



      Although basal-cell carcinoma is the only cancer so far in clinical trials with GDC-0449, the hope is that it could also be used with other cancers because of its mechanism of action as a disrupter of the hedgehog pathway, which is not only associated with nearly all basal-cell carcinomas, but also some medulloblastomas, leukemias, and breast, lung, and pancreatic cancers.



      Early evidence of its efficacy beyond basal-cell carcinoma comes from a case study, published last September in the New England Journal, that showed quick regression of metastatic medulloblastoma in a 26-year-old man treated with GDC-0449, although he later developed resistance and died from the disease.



      The hedgehog pathway plays a crucial role in embryonic development, and then typically shuts down in adult tissue. In 1996, Dr. Epstein identified the site of mutation that triggers reactivation of the pathway in cancer, the PTCH gene. GDC-0449 is the first drug that blocks the hedgehog pathway, and others are in the pipeline.



      A spokeswoman for the manufacturer, Genentech, Krysta Pellegrino, said the company expects to seek approval for Phase III trials soon. :rolleyes:

      auch wenn bereits Pivotalstudien aus ph2 vorliegen und mit diesen eine Zulassuung zur Behandlung des Basalzellkarzinoms beantragt wird, werden ph3-Studien initialisiert werden. Es wird angenommen, dass der Hedgehog-Signalweg bei verschiedensten Krebsarten eine Rolle spielt und somit die Hemmung dieser interzellulären Informationsübertragung zumindest als Begleittherapie sinnvoll sein könnte.

      Delcath ist ja nun etwas gefallen, trotz der CE-Kennzeichnung für Europa.
      Wie seht ihr hier die kurzfristigen Chancen bis Jahresende?

      In den US-Foren liest man, dass trotz einer CE für Europa, es noch keine baldige FDA-Zulassung bedeutet und dies sich über Jahre ziehen könnte. Man führt dafür etliche Beispiele anderer Unternehmen an.

      Und die große Frage für Europa ist, gelingt hier erfolgreiche Vermarktung und die Erstattung der Behandlung bei den verschiedenen, staatlichen Gesundheitssystemen und jeweiligen Besonderheiten der EU-Mitgliedsländer. Zweifellos scheint ja der Bedarf vorhanden zu sein.
      Delcath selbst schreibt ja von ca. 100.000 die in der EU dafür in Frage kämmen, das wäre ja einiges an Umsatzpotenzial würde man sich einen Teilbereich davon erschließen.

      Roche könnte in diesem Quartal noch den Zulassungsantrag für Vismodegib zur Behandlung des Basalzellkarzinoms einreichen, so dass einerseits Meilensteinzahlungen an Curis sowie die Phantasie auf zukünftige Lizenzeinnahmen andererseits den Kurs der Curis-Aktie beflügeln sollten.

      Seattle Genetics erwartet auf Ende August eine Zulassungsentscheidung zu Brentuximab zur Behandlung von 2 unterschiedlichen Indikationen (Hodgkin-Lymphom sowie eine spezielle Form eines Lymphoms). Sollte hierbei etwas schief gehen, dürfte der Kurs arg unter die Räder kommen, da mE eine positive Zulassungsentscheidung weitestgehend in den derzeitigen Kurs eingepreist ist. Die zukünftige bzw. mittelfristige Entwicklung ist jedoch durchaus positiv zu beurteilen.


      BOTHELL, Wash. (AP) - Seattle Genetics Inc. said Monday the Food and Drug Administration is now reviewing its experimental cancer drug as a treatment for Hodgkin's disease and a type of lymphoma.

      The company said the FDA will make separate decisions on the two applications by Aug. 30. Seattle Genetics filed for marketing approval of the drug, called brentuximab, as a treatment for patients whose cancer did not respond to other drugs or has returned after previous treatment.

      Aber so weit man den offiziellen Hinweisen des Roche-Managements vertrauen kann, soll Vismodegib noch in diesem Jahr bzw. noch in diesem Quartal zue Zulassung bei der FDA eingereicht werden.

      In Europa bei der EMEA werden jedoch normalerweise Erbgebnisse aus ph3-Studien zur Zulassung vorausgesetzt.

      Allerdings wird sich eine Zulassung von Vismodegib relativ bescheiden im Kurs auswirken (ich hoffe auf Kurse > USD 5), da Curis nur mit Meilenstein- sowie Lizenzzahlungen am Erfolg von Vismodegib partizipieren wird.

      NEW YORK (AP) -- A Jefferies and Co. analyst started covering shares of Immunogen Inc. with a "Buy" rating Wednesday, saying the company's drug candidate T-DM1 will become a standard treatment for breast cancer

      Analyst Thomas Wei said the drug could become the standard treatment for advanced hormone-resistant breast cancer, replacing Roche's drug Herceptin. He said T-DM1 is safer than Herceptin, and said he thinks the drug will be approved as a secondary treatment for breast cancer in 2013, as a primary treatment in 2015, and as a supplemental treatment by 2018.

      Immunogen is developing T-DM1 through a partnership with Roche. T-DM1 is a combination of Herceptin and a cell-killing agent discovered by Immunogen. Wei said T-DM1 will eventually take the majority of Herceptin's $5 billion in annual sales.

      He set a price target of $19 on shares of the Waltham, Mass., company. Immunogen shares closed at $12.21 Tuesday, meaning Wei expects the stock to rise 55.6 percent over the next year.

      nächsten Donnerstag diskutiert eine Expertenrunde auf Einladung der FDA den Zulassungsantrag = BLA (Biologics License Application) von ADCETRIS (brentuximab vedotin) von Seattle Genetics zur Behandlung von Hodgkin’s lymphoma (HL) am Vormittag sowie am Nachmittag zur Behandlung von "relapsed or refractory systemic anaplastic large cell lymphoma (ALCL)".

      Dann dürfte es wieder einmal spannend werden, da es sich um eine binäre Angelegenheit handelt, wobei eine Ablehnung sicherlich eine Kurshalbierung zur Folge hätte. Ich rechne jedoch fest mit einer Zulassungsempfehlung. Ein Kursfeuerwerk wird jedoch ausbleiben, da mE eine positive Entscheidung weitestgehend in den Kurs eingepriesen ist --> Halteposition; risikoscheue Anleger dürften vor dem AC den Verkaufsdruck erhöhen, so dass der Kurs bis dahin möglicherweise leicht nachgibt.

      There is no ongoing trial designed to confirm the clinical benefit of brentuximab vedotin in patients with ALCL.
      The Agency would like the ODAC to consider whether or not accelerated approval should be granted without an ongoing confirmatory trial.

      Per 30.06.2011 betrug die Shortquote 21,3 Millionen Stücke - das ist enorm bei ca. 110 Millionen ausstehenden Aktien.

      Also ist klar, dass die Shorter auf dem falschen Fuß erwischt wurden und nun wird alles versucht , es zu richten ... mit Cramer, Feuerstein oder eben Canaccord und fiesesten Tricks. Denk mal an Dendreon, als es diesen unerklärlichen Spike nach UNTEN gab - das waren auch die Shorter, die ihre Haut retten wollten.

      Bei SGEN scheinen sie nicht mit der Brechstange anzusetzen und deshalb vermute ich, dass bei Erreichen der 200er Schluss sein könnte, die aktuell bei ca. 16,60 USD steht. Wenn diese Marke hält, dann aufzustocken macht bestimmt Sinn.

      Roche Holding AG (ROG)’s experimental vismodegib treatment helped patients with a form of skin cancer known as advanced basal cell carcinoma, according to a study presented today at the European Multidisciplinary Cancer Congress in Stockholm.

      The drug shrunk tumors and healed lesions in patients whose cancer had progressed too far to be removed with surgery, Roche said in a statement Sept. 12.

      Eine Frage an Cyberhexe zu Delcath. So weit ich hier mitlesen konnte, hast du Delcath noch nicht aus deinem Depot geräumt. Die sind ohne negative Nachrichtenlage ganz schön unter die Räder gekommen... trotz des anstehenden Meetings Mitte Januar mit der FDA.
      Wie ist deine Einschätzung der Lage bei Delcath? Eine kleine Spekulation zu diesem Preis, müsste die Aktie doch Wert sein, oder? :cool:

      NEW YORK (AP) -- Federal regulators on Monday approved a pill that treats the most common type of skin cancer, basal cell carcinoma.

      The pill is called Erivedge and is made by Genentech, a unit of Swiss drugmaker Roche. Erivedge is intended to treat locally advanced cancer for patients who are not candidates for surgery or radiation, and for patients whose cancer has spread to other parts of the body. The capsule is taken once per day.

      Genentech said Erivedge is the first drug approved to treat advanced basal cell carcinoma. It said the drug will be available within one to two weeks.

      The drug's label will warn that it is linked to fetal death and severe birth defects when it is used by pregnant women. The most common side effects of Erivedge include muscle spasms, hair loss, weight loss, diarrhea, fatigue, changes or loss in sense of taste, decreased appetite, constipation, and vomiting.

      Curis Inc. of Lexington, Mass., which collaborated with Genentech on the drug, is getting a $10 million payment from Genentech now that the drug has been approved.

      A skin cancer drug aimed at stopping the disease from using the so-called hedgehog pathway was approved by the Food and Drug Administration more than a month ahead of schedule.

      Vismodegib, a once-a-day capsule that will be marketed by South San Francisco-based Genentech Inc. as Erivedge, was approved for use by patients with basal cell carcinoma that has spread to other parts of the body, has returned after surgery or cannot be treated with surgery or radiation.

      The drug was worked on by Genentech and Lexington, Mass.-based Curis Inc. (NASDAQ: CRIS), but its Bay Area roots run deep. Labs at the University of California, San Francisco , Children’s Hospital Oakland Research Institute and Stanford University worked on the drug.

      The drug, the first FDA-approved medicine for people with advanced forms of the most common form of skin cancer, will cost a reported $7,500 a month over 10 months of treatment. It is expected to be available in the United States in one to two weeks.

      Erivedge inhibits the hedgehog signaling pathway, named for the prickly appearance of the targeted gene. The gene normally is involved in development but sometimes goes awry, with abnormal signaling of the gene implicated in more than 90 percent of basal cell carcinoma cases.

      Genentech, which also is studying Erivedge in a Phase II trial of patients with operable forms of basal cell carcinoma, said its owner, Roche of Switzerland, has submitted Erivedge for approval in the European Union.

      “We continue to be pleased with Genentech and Roche’s global Erivedge development efforts,” Curis President and CEO Dan Passeri said in a press release.

      Approval translates into a $10 million milestone payment for Curis. The company also can receive royalties on sales of Erivedge.

      The FDA was scheduled to move on the drug by March 8.

      NEW YORK, Feb. 2, 2012 /PRNewswire/ -- Delcath Systems, Inc. (NASDAQ: DCTH - News) announced today that the first patients in Europe have been treated with the Delcath Hepatic CHEMOSAT® Delivery System at the European Institute of Oncology (Instituto Europeo di Oncologia – IEO), a premier cancer treatment and research center in Milan. The cases were treated as part of the initial launch and training agreement the Company announced with the IEO in November 2011.

      Two patients were treated for inoperable liver-dominant metastases from ocular melanoma and gastric cancer. All CHEMOSAT procedures were successfully completed without procedure-related complications. Delcath and the IEO will host a joint-press conference, on February 15, 2012 at the IEO, at which time an update on patient status will be presented.

      The procedures were conducted with the Generation One version of the CHEMOSAT system, while the Generation Two version is under review for CE Mark approval by the Notified Body.

      CHEMOSAT is a proprietary product that utilizes chemosaturation technology, a minimally invasive, repeatable procedure that delivers high doses of chemotherapeutic drugs directly to the liver while minimizing systemic exposure of such drugs. CHEMOSAT received CE Mark in April 2011 as a Class III medical device with an indication for the percutaneous intra-arterial administration of a chemotherapeutic agent (melphalan hydrochloride) to the liver.

(NASDAQ: DCTH - News) today announced that the Company has received CE Mark approval for the second generation hemofiltration cartridge of the Company's proprietary Hepatic CHEMOSAT® Delivery System. With the new hemofiltration cartridge, the CHEMOSAT system carries the same broad indication as the previous generation system, permitting physicians to use the product for the percutaneous intra-arterial administration of a chemotherapeutic agent (melphalan hydrochloride) to the liver to any patient who in their opinion may benefit.

(DCTH) announced today that doctors from the European Institute of Oncology (Instituto Europeo di Oncologia – IEO) presented preliminary data showing reduced side effects in the first treatment using the Company's second generation Hepatic CHEMOSAT® Delivery System as compared to a prior treatment with the first generation system.

      The comparison was presented at a sponsored symposium entitled Chemosaturation Therapy – Initial European Experiences hosted by Delcath at the Third European Conference on Interventional Oncology (ECIO, April 25-28) held in Florence, Italy. The session was moderated by Dr. Ricardo Lencioni, Director of Imaging and Diagnostic Intervention at the University of Pisa, and featured presentations by Dr. Pier Francesco Ferrucci, Director of Translational Research on Melanoma and medical oncologist at the IEO, Dr. Franco Orsi, Director of Interventional Radiology at the IEO, and Dr. Thomas J. Vogl, Director of the Institute of Diagnostic and Interventional Radiology at the J.W. Goethe University Hospital in Frankfurt, Germany.

      Dr. Ferrucci presented the comparison of side-effect profiles in two CHEMOSAT treatments performed in a single patient. Treatment with the second-generation CHEMOSAT system showed brief mild bone marrow suppression and no significant related toxicities. In the prior treatment of the same patient, which was conducted with the first generation CHEMOSAT system, moderate to severe bone marrow suppression and related toxicities were noted.

      ...die Komplikationen bei der Anwendung der Chemosaturation von Bedeutung. Da sowohl Melphalan als auch die Anwendung der Prozedur nicht unkritisch sind, ist die Anzahl der Todesfälle (3 von 40 Patienten) auf den ersten Blick erschreckend:

      a) Treatment related Deaths:
      --> 3/40 patients (7.5%) 3/116 procedures (2.6%)
      --> Neutropenic Sepsis (n=2) 5%, Hepatic Failure (n=1) 2.5% (95% tumor burden)



      In Anbetracht dessen, dass es sich jedoch um Patienten ohne wirkliche Alternative handelt, wird dieses Mortalitätsrisiko (3-10%) bei Melphalan von der FDA toleriert!

      --> Current approved labeling for Melphalan - 3% to 10% mortality rate.

      Delcath erhielt seitdem einen CRL in den USA und die Zulassung in der EU, wo schon behandelt wird!
      Der neue Gen2 Filter der das hochdosierte Chemoblut filtert wird meiner Meinung nach der Schlüssel zur positiven Abstimmung, außerdem hat die FDA eine EPA für Delcath gewährt:

      Hallo Cyberhexe,

      Kann man mal fragen was Du davon hältst? Möchte die nicht kaufen, hab mich nur, als Laie, gefragt ob dieser Ansatz 'einfach grundsätzlich erstmal Sinn macht'? Oder ob der dann auch "automatisch" bessere, oder schechtere, Chancen verspricht? Mach Spass sich einfach mal ein bisschen in was 'themenfremdes' reinzudenken. :)

      Meinem Eindruck nach: Sinn machen dürfte es schon einigen, bessere oder schlechtere Chancen aber eigentlich nicht versprechen -da die Entwicklung der Anwendung ja eigentlich nicht, direkt, betroffen ist. Oder?? Aber mal rein aus Sicht der (sonst immer vorkommenden??)Beschädigung der Zellen scheint mir das schon 'ziemlich genial'? Ist im Pdf auf Seite 14:

      "BIND’s nanomedicine platform has a game-changing ability to target therapeutics to the site of disease, which could transform the treatment of cancer and other diseases.

      Drugs that kill cancer cells are generally toxic to healthy cells as well, and to get these drugs to the diseased cells, you have to treat the whole body.
