Speichel von Parasiten beinhaltet wertvolle chemische Substanzen

Blutsaugende Tiere wie Zecken, Moskitos und Läuse haben in ihrem Speichel Substanzen, die in der modernen Medizin Verwendung finden könnten, meinen Wissenschaftler. Dort konnten Forscher die Substanz Hidurin aus dem Speichel von Läusen gewinnen. Diese wird zur Blutverdünnung verwendet, berichtet die Straits Times in ihrer jüngsten Ausgabe.

Der einzige Nachteil an Hirudin ist die Tatsache, dass dieser Stoff nur ein paar Mal verabreicht werden kann, da das menschliche Immunsystem ihn erkennt und dann die Wirkung ausschalten kann. Daher suchen die Wissenschaftler neue Substanzen in den rund 40.000 blutsaugenden Lebewesen, berichtet das Wissenschaftsmagazin New Scientist. So sind zum Beispiel Hakenwürmer, die sich von menschlichem Blut ernähren und sich in den Organen festsetzen, Kandidaten für weitere Untersuchungen. Das amerikanische Pharma-Unternehmen Corvas hat entdeckt, dass die Hakenwürmer ein gerinnungshemmendes Protein erzeugen, das für einige operative Eingriffe verwendet werden könnte. Im vergangenen Jahr konnte in einem Versuch an Knieoperierten die Substanz gegen die Bildung von Blutklumpen verabreicht werden.

Das britische Pharmaunternehmen Evolutec arbeitet intensiv daran, das Geheimnis der Speichelsekrete von Zecken zu erforschen: Mehr als 300 Chemikalien sind darin verborgen. Sie sorgen dafür, dass die Zecken unentdeckt und unbemerkt so lange am Wirt bleiben, bis sie vollgepumpt mit deren Blut abfallen. "Bis zu 14 Tage kann das dauern", so Wynee Weston-Davies, Development Director von Evolutec. "Wir wissen, dass dahinter ein kluger Schachzug steckt", meint der Wissenschaftler. Normalerweise würde ein Biss zu einer Schwellung und Entzündung führen, die dann zu einem Juckreiz führt. Aber der Zeckenspeichel verfügt über histaminbindende Proteine, die das Histamin neutralisieren und so eine Entzündung verhindern.

Da Histamine auch zu allergischen Reaktionen wie Heuschnupfen oder Asthma führen, verspricht sich das Unternehmen die Entwicklung von neuartigen Arzneimitteln. Evolutec sucht gezielt nach der Substanz EV 131, die im Speichel von Zecken vorhanden ist. Bereits in wenigen Monaten soll mit den ersten Tests von neuen Präparaten gegen Bindehautentzündung begonnen werden. Wenn die Tests erfolgreich verlaufen, könnte schon in zwei Jahren das neue Präparat aus Zeckenspeichel in den Regalen stehen. Das Medikament wird das Immunsystem des Menschen nicht beeinflussen. "Zecken gibt es länger als Dinosaurier, daher haben sie während dieser Zeit genug Möglichkeiten gefunden, eine optimale Strategie zu entwickeln", so Weston-Davies.

Evolutec Group plc (AIM: EVC), the biopharmaceutical company developing novel products for the treatment of allergic, inflammatory and autoimmune diseases, is pleased to announce, ahead of schedule, a positive clinical trial result for its lead molecule, rEV131, in a 112 patient Phase II study in allergic rhinitis (hay fever).

rEV131 is a recombinant version of a naturally occurring molecule identified in ticks, parasites whose saliva contains immuno-suppressive and immuno-modulatory molecules over which Evolutec has strong patent protection.

This Phase II dose-ranging nasal allergen challenge study of rEV131 administered as a nasal spray was undertaken at two centres in San Antonio, Texas, under the leadership of Dr Paul Ratner. The trial was conducted in accordance with the Food & Drug Administration`s Guidance for Industry recommendations.

The trial comprised four cohorts of 20 patients (16 active, 4 placebo), with the active patients on ascending single doses of rEV131, followed by a fifth cohort of 32 patients (16 active, 16 placebo) at the optimum dose. The ragweed pollen extract was administered 30 minutes after dosing.

The trial met its primary endpoint, a statistically significant difference (p< 0.05) in the mean sum of symptom scores at 15 minutes post allergen challenge in the 62 patients who completed the trial according to the protocol. rEV131 showed a dose dependent drug effect enabling Evolutec to select the optimum dose for further work to determine the onset of action and the duration of effect. There were no significant adverse events and rEV131 was comfortable and well-tolerated.

This result shows that rEV131 has an onset of action of 45 minutes or less - quicker than steroid nasal sprays which have an onset of action of approximately 8 hours. The main effects of rEV131 were against congestion and mucus production, symptoms that patients find the most troublesome and which are not well addressed by oral antihistamines. The rapid onset of action and efficacy against congestion and mucus underpin the potential commercial advantages of rEV131.

Mark Carnegie Brown, Evolutec`s Chief Executive Officer, commented: "This result highlights the potential of rEV131 in the $6 billion allergic rhinitis market. We now intend to undertake a multi-dose Phase II study to define further the onset and duration of action which will guide commercial positioning. We will also continue our dialogue with potential partners."

Scientists tick off benefits of Evolutec hay fever drug

Evolutec, an anti-allergy drug development company, said yesterday its hayfever drug based on tick saliva appeared to have "significant advantages" over current treatments for the condition.

The company announced Phase II results for the drug, which is at present called rEV131. These showed that the drug worked much faster than nasal steroids.

"Many people with hayfever wake up and sniff the air before deciding whether or not to take their drugs," explained chief executive Mark Carnegie Brown.

"However, nasal steroids don`t start working for a period of up to eight hours."

He said that the drug also worked on congestion, which antihistamines do not.

Meanwhile, rEV131 works in 45 minutes or less. "The rapid onset of action against congestion and mucus underpin the potential commercial advantages of rEV131," the company said yesterday in a statement.

The company`s drugs are based on the saliva of ticks, which were originally crushed up so that scientists could extract their saliva glands.

The drug is now grown in petri-dishes, but Mr Carnegie Brown explained that the action is the same. "Ticks remain unnoticed on people and animals for a period of up to 14 days," he said.

"The original scientists working on this realised that they must be doing something to suppress the immune system."

The drug works by binding histamine, the substance that is made by the body which causes itching and swelling symptoms common in an allergic reaction.

Evolutec is working on a number of drugs using the same method, which it believes will treat everything from bronchitis to heart attack.

The company is now going to do a larger Phase II study to define the onset and duration of action of the drug.

Mr Carnegie-Brown said it would be unlikely to be on the market for another four years.
