      Hey Cyberhexe,

      hab vorhin mal angefangen Deinen2007 Thread zu lesen,
      fand ich wirklich interessant,
      zu der Performance -größten Respekt!

      Damit dürftest Du wohl mindestens 95% aller anderen Anleger
      hochhaus geschlagen haben.

      Biotechnologiewerte sind mir ziemlich suspekt,
      wills aber gern besser verstehn, werd bestimmt mindestens mitlesen.

      (nicht wirklich so viel auf Diversifizierung zu geben
      -den Charme kann ich schon verstehen,
      einzige Voraussetzung: man muss wissen was man macht
      und worauf man sich einlässt!)

      Am 4. Januar schloss der Dow Jones Industrial Index bei 12.800 Indexpunkten mit einem Minus von 1,96% auf dem Tief aufgrund schlechter Arbeitsmarktdaten, was nichts Gutes für die nächste Woche befürchten lässt. Die schlechten Konjunkturdaten und der Rekordölpreis von erstmals 100 USD/Barrel vermassselten den Anlegern damit weltweit die erhoffte Jahresanfangsrallye. Noch mehr kam zuvor aufgrund der Angst vor einer US-Rezession

      Bei Elan steht diese Woche noch eine wichtige Entscheiduing an und zwar hat die FDA über eine erweiterte Anwendung von Tysabri (Natalizumab) zu entscheiden.
      Sollte die Zulassung zur Behandlung von Morbus Crohn erteilt werden - allerdings lediglich für Patienten, die auf die bisherigen TNF-alpha-Wirkstoffe nicht ansprechen - dann wird das Potential für Tysabri weiter zumehmen.
      Heute wurde übrigens in einer Pressemitteilung bekanntgegeben, dass mittlerweile 20.400 MS-Patienten kommerziell und über 700 in klinischen Tests mit Tysabri behandelt werden und bisher keine PML aufgetreten sei:

      Auch die Zahl der verschreibenden Ärzte stegt kontinuierlich, so dass zukünftig ein progressives Wachstum durchaus wahrscheinlich ist.

      Von dieser Entwicklung profitiert auch PDL Biopharma, mit deren Technologie der humanisierte Antikörper Natalizumab entwickelt wurde und von dessen Umsätzen PDL Lizenzgebühren (wahrscheinlich 3.5% von den Umsatzerlösen) erhält.
      Da PDL derzeit massiv unterbewertet zu sein scheint, wurde die Anzahl an Call-Optionen im Depot auf 10.000 verdoppelt! Die Laufzeit bis 17.12.2008 erscheint ausreichend genug, um an einem potentiellen Verkauf der Gesellschaft zu profitieren.

      Um das Risiko etwas zu steuen, werden demnächst 200 Elan-Anteile veräussert...jedoch lediglich als Vorsichtsmassnahme - die Zukunftzsaussichten von Elan rechtfertigen nach wie vor eine überproportionale Gewichtung im Depot!

      Mit Spannung wird spätestens am Montag die Entscheidung der FDA über eine erweiterte Zulassung von Tysabri erwartet - da sehr viele MC-Patienten auf die TNF-Wirkstoffe nicht ansprechen, wäre Tysabri eine zusätzliche Opton, einen chirurgischen Eingriff zu vermeiden. Das Potential nur bei MC wird auf ca. 500 Mio USD/a geschätzt. Auch PDL würde an einer erweiterten Zulassung profitieren, da dieser humanisierte Antikörper mit PDLs Schlüsseltechnologie entwickelt wurde und für PDL Lizenzeinnahmen generiert.

      Wie erwartet wurde Tysabri nun auch zur Behandlung von MorbusCrohn mit mässigem bis schwerem Verlauf von der FDA zugelassen. Auch wenn die Anwedndung nur restriktiv für all diejenigen Patienten erfolgen wird, bei denen die am Markt bereits platzierten Medikamente wirkungslos sind (in den US immerhin 100.000 Patienten), dürfte das Wachstum im 2.Quartal von dieser Entscheidung positiv beeinflusst werden. Mit erwarteten 30.000 Patienten, die Mitte 2008 mit Tysabri behandelt wereden, dürfte Elan dann die Gewinnzone erreichen.

      Bei PDL sind die Kurse trotz positiver Analystenkommentare rückläufig. Und da vom Management derzeit wenig bis gar nichts zu den Verkaufsabsichten zu hören ist, ist die Situation alles andere als übersichtlich. Jedoch ist die Überzeugung nach wie vor gross, dass diese Gesellschaft derzeit weit unter Wert gehandelt wird.

      In Europa erhebliche Zulassungserweiterung für Avastin in der Behandlung des metastasierenden Kolorektalkarzinoms (Dickdarmkrebses)
      Jetzt können deutlich mehr Patienten vom nachgewiesenen Überlebensvorteil durch Avastin profitieren

      Roche gab heute bekannt, dass der Angiogenesehemmer Avastin (Bevacizumab) des Konzerns von der Europäischen Kommission eine erhebliche Zulassungserweiterung im Anwendungsgebiet metastasierendes Kolorektalkarzinom (Dickdarmkrebs) erhalten hat.

      Nach der Zulassungserweiterung kann Avastin bei Patienten mit metastasierenden Kolorektal¬karzinomen von nun an sowohl als Erstlinienbehandlung als auch in späteren Behandlungslinien in Kombination mit jeder beliebigen Chemotherapie einschliesslich des oral anzuwendenden Zytostatikums Xeloda (Capecitabin) von Roche eingesetzt werden. Dank dieser Zulassungserweiterung können in Zukunft so gut wie alle Patienten mit metastasierenden Kolorektalkarzinomen vom nachgewiesenen Überlebensvorteil von Avastin profitieren. Schätzungen zufolge wird 2008 bei über 400.000 Menschen in Europa die Diagnose metastasierender Dickdarmkrebs gestellt.

      Während bei Elan derzeit nichts Spektakuläres ansteht und der Kursverlauf den Erwartungen entspricht, stellt der Kursverlauf von PDL ein grosses Rätsel dar. Entweder sind die Lizenzverträge für die Antikörper derart degressiv abgeschlossen (will heissen, dass bei zunehmendem Umsatz die abzuführende Rate deutlich abnimmt) oder aber die Kleinanleger werden hier bewusst von institutionellen Anlegern "weich geklopft". Für dieses Szenario spricht die Tatsache, dass auch nach dem Ausstieg des bisher grössten Anteilseigner THIRD POINT LLC die instituionelle Quote immernoch über 100% liegen wird. Auch der etwas entäuschende Preis für den Rest des cardiovasculären Portfolio ändert nichts an der Tatsache, dass der Wert der vielen auslizenzierten humanisierten Antikörper die derzeitige Marktkapitalisierung bei Weitem übertreffen sollte. Wenn man bedenkt, dass für die Lizenzrechte von Blockbustern nahezu 1 Milliarde USD bezahlt werden ("...$700 million purchase of Humira royalties...a $650 million deal for Remicade royalties from New York University and a $500 million..."), dann sollte dies auch für die am Markt eingeführten Blockbuster Avastin und Herceptin sowie den potentiellen Blockbustern Natalizumab, Tocilizumab, Bapineuzumab etc. annähernd möglich sein. Die jetzige Marktkapitalisierung von PDL (1,7 Milliarden USD) scheint mir vor diesem Hintergrund nicht nachvollziehbar.

      PDL hat die Produktionsstätte für Antkörper in Brooklyn Park, Minnesota (USA) für 240 Mio$ an das dänische Unternehmen Genmab verkauft - der Verkaufspreis und für das Know how der Antikörperproduktion hatten die Experten jedoch im Minimum 3 USD/Aktie (dies wären ca. 350 Mio USD) erwartet. Bis auf die Entwicklungsprodukte der eigenen Pipeline hat PDL nun alles ausser der Antikörperplatform mit den daraus abgeleiteten Lizenzansprüchem verkauft. Interessant ist hierbei, dass die FDA Bevacizumab (brans name Avastin) nun auch zur Behandlung von Brustkrebs in den US zugelassen hat, wodurch das Avastin-Potential massiv beeinflusst wird (zusätzliches Potential wird auf über 1 Milliarde USD geschätzt). Da PDL für Avastin Lizenzgebühren erhält, wird die "cash cow" Avastin auch für PDL noch etwas fetter.

      Ich rechne mit einem Verkauf der Gesellschaft für ca. 24 - 27 USD. Möglicherweise werden jedoch nochz erste Daten aus den klinischen Studien von Bapineuzumab (viel versprechender Wirkstoff von Elan/Wyeth zur Behandlung von Alzheimer) abgewartet, weil hierdurch die zukünftigen Lizenzeinnahmen massiv beeinflusst werden könnten. 3.5 % der Einnahmen eines Umsatzgiganten könnten den Wert von PDL deutlich erhöhen.

      PDL BioPharma(PDLI - Cramer's Take - Stockpickr) couldn't find a buyer.

      The Redwood City, Calif.-based biopharmaceutical firm said Tuesday that it has ended plans for a potential company sale after no bids were submitted.

      As a result, PDL intends to remain an independent company but will put in place a corporate restructuring plan that includes cutting costs and firing nearly half of its employees. The company is also exploring ways to distribute past and future proceeds from product royalties and asset sales to shareholders.

      Shares were plunging $2.94, or 19%, to $12.90 in after-hours trading.

      PDL put itself up for sale in October, seeking buyers for all or parts of its operations. Since then, the company sold its antibody manufacturing facility and its marketed cardiovascular drugs.

      But PDL's most important assets -- its pipeline of experimental drugs and the royalties it receives on humanized antibody drugs like Genentech's(DNA - Cramer's Take - Stockpickr) Avastin and Biogen Idec's(BIIB - Cramer's Take - Stockpickr) Tysabri -- remained out there for the taking.

      No one was interested enough to put in a bid.

      "Although we garnered interest regarding certain of our pipeline programs, we did not receive a firm offer for the company as a whole or for our biotech R&D assets," said PDL Chairwoman Karen Dawes in a statement.

      To remain independent, PDL says it will fire about 260 employees, reducing its headcount to about 300. The company will also reduce operating expenses to about $150 million annually from its current run rate of $340 million.

      PDL says it will continue to develop its drug pipeline, which includes a cancer drug and a multiple sclerosis drug partnered with Biogen Idec. The company has a poor track record of developing its own drugs, which is one reason why a buyer was being sought.

      Separately, PDL said its fourth-quarter loss from continuing operations swelled to $24.5 million, or 21 cents a share, from $2.7 million, or 2 cents a share, a year earlier. Revenue tumbled to $49.8 million from $59.8 million.

      Genentech expects to submit the supplemental Biologics License Application (sBLA) to the U.S. Food and Drug Administration (FDA) for accelerated approval of Avastin® (bevacizumab) for the treatment of relapsed glioblastoma multiforme in the second half of 2008. Data will be presented at the 2008 annual meeting of the American Society of Clinical Oncology (ASCO).

      Genentech announced that the Phase III study sponsored by its collaborator Novartis evaluating Xolair® (Omalizumab) for subcutaneous use in patients 6 to 11 years of age with moderate-to-severe, persistent, inadequately controlled allergic asthma met its primary endpoint, demonstrating a statistically significant reduction in exacerbations in Xolair-treated patients compared with placebo-treated patients with no new safety signals reported. The companies will evaluate the complete study results and feedback from the FDA to determine appropriate next steps. The data will be presented at a medical conference later this year.

      Besonders hervorzuheben ist die Option mit Tysabri direkt in eine Ph3-Studie zur Behandlung von "Ulcerative Colitis" einzusteigen sowie Phase2-Studien in der Krebstherapie (Multiple myeloma) aufzunehmen. Ergenisse von Bapineuzumab werden Ende Juli erwartet.

      PDL hat nach dem Verkauf des cardiovaskulären Portfolio sowie der Antikörper-Produktionsstätte eine Sonderdividende in Höhe von 500 mUSD (4,25 USD/Aktie) ausgeschüttet. Wie in solchen Fällen üblich, wurden in Folge dessen auch die Ausübungspreise (Strikes) für Optionen angepasst. Die Bedingungen der im Depot befindlichen Option haben sich dementsprechend wie folgt verändert:
      Strike: 13,89 USD (vorher 20 USD - wie man allerdings auf diesen Wert kommt, ist mir schleierhaft!)
      Ausübungsverhältnis 0,1444 (vorher 0,1 - ebenfalls schleierhaft)

      Heute nach Börsenschluss wird das Quartalsergebnis bekanntgegegen. Es ist mit Lizenzeinnahmen auf Verkäufe des 4. Quartals der Lizenznehmer so um die 50 mUSD zu rechnen - das 2. Q wird jedoch mit geschätzten 102,6 mUSD auf die Verkäufe im 1. Quartal wesentlich ertragreicher.

      Naproxcinod

      NicOx SA's lead product is in Phase III clinical development for treating the signs and symptoms of osteoarthritis. According to NicOx management, naproxcinod has the potential to become the drug of choice for osteoporosis, with no detrimental effect on blood pressure and a solid gastrointestinal tolerability. Known by the product code HCT 3012, naproxcinod is a unique nitric oxide-donating anti-inflammatory. Naproxcinod represents the first of the CINOD class (Cox-Inhibiting Nitric Oxide Donators).

      Top-line results from the first Phase III study were successful, according to company executives. This performance represents a significant step toward biopharmaceutical company NicOx's (nicox.com) goal of making naproxcinod the drug of choice for osteoarthritis patients.

      Millions of people rely on non-steroidal anti-inflammatory medicines for the treatment of chronic pain and inflammation relating to a range of conditions, such as osteoarthritis. cox-2 selective non-steroidal antiinflammatory drugs and non-selective non-steroidal anti-inflammatory drugs have been linked to an increased risk of serious cardiovascular events. These events include heart attack and stroke, creating a necessity for new medicines with better safety profiles. This demand is particularly crucial for patients with coexisting cardiovascular risk factors, such as hypertension, who represent about 40% of osteoarthritis sufferers.

      NicOx announced in October 2006 top-line results from the first pivotal Phase III clinical trial for naproxcinod. This study is known as the 301 trial. The results confirmed that naproxcinod is superior to placebo in relieving the signs and symptoms of osteoarthritis. Naproxcinod demonstrated a clear and sustainable effect of lowering blood pressure and had good safety and tolerability.

      The following month NicOx announced top-line results of a non-
      pivotal study, using the Ambulatory Blood Pressure Monitoring technique in 131 hypertensive subjects. This Ambulatory Blood Pressure Monitoring trial showed a differentiated and favorable 24-hour blood-pressure profile for naproxcinod compared with naproxen after two weeks of treatment.

      NicOx management believes that naproxcinod will generate significant market potential if the drug's non-negative blood-pressure profile in contrast to existing non-steroidal anti-inflammatory drugs is confirmed. This is based on the fact that the blood-pressure increase evident with existing non-steroidal anti-inflammatory drugs is considered one of the potential factors responsible for the apparent heightened risk of serious cardiovascular events observed with these agents.

      The Phase III efficacy program for gaining regulatory approval for naproxcinod in the United States and Europe consists of three pivotal Phase III studies. The 301 study was the first clinical trial in the program. This study produced top-line results in October 2006. The 13-week, double-blind, placebo and naproxencontrolled trial for 918 patients with knee osteoarthritis was conducted in 120 U.S. clinical sites. Eligible patients were randomized to one of four treatment groups: naproxcinod 375 milligrams twice daily, naproxcinod 750 milligrams twice daily, naproxen 500 milligrams twice daily, or placebo twice daily. Naproxcinod demonstrated good overall safety. The number of serious adverse events was low and evenly spread among treatment groups. Bloodpressure data for both naproxcinod doses demonstrated a sustained reduction compared with baseline and naproxen at all time points. The number of adverse hypotensive events was low across all treatment groups. The 301 study has a longterm extension that will provide additional safety data.

      NicOx completed enrollment in the second pivotal Phase III study for naproxcinod in December 2007. Begun in April 2007, the 302 study is a 53-week, randomized, double-blind, efficacy and safety trial in which 1,020 patients with knee osteoarthritis have been enrolled at 150 U.S. clinical centers. Efficacy results are anticipated during thirdquarter 2008.

      Patients have been randomized to one of these treatment groups: naproxcinod 375 milligrams twice daily (52 weeks), naproxcinod 750 milligrams twice daily (52 weeks), naproxen 500 milligrams twice daily (52 weeks) and placebo twice daily during the first 13 weeks. Following 13 weeks, the placebo-treated patients are being randomized to either naproxcinod 375 milligrams twice daily or naproxcinod 750 milligrams twice daily for the trial remainder (39 weeks). The two doses of naproxcinod will be compared to placebo on the three co-primary efficacy endpoints that are the same as those used in the other pivotal Phase III studies. A secondary trial endpoint will compare the efficacy of naproxcinod and naproxen at 26 weeks. The general safety and tolerability of naproxcinod will be assessed until week 52, and the clinical trial has a one-week posttreatment safety period.

      NicOx launched the third and final pivotal Phase III trial for naproxcinod in June 2007. The 303 study is a 13-week, double-blind, placebo and naproxen controlled trial in patients with hip osteoarthritis. This efficacy and safety clinical trial is expected to enroll 800 patients at 100 clinical centers in the United States, Canada, and Europe. Efficacy results are anticipated by yearend 2008. Eligible patients will be randomized to three arms: naproxcinod 750 milligrams twice daily, placebo twice daily, and naproxen 500 milligrams twice daily.

      NicOx will conduct a statistical analysis according to a predefined plan on the pooled office bloodpressure measurements data from the three Phase III studies following completion of the 302 and 303 trials. Company management anticipates filing a new drug application with FDA for naproxcinod during first-quarter 2009.

      In October 2007, France-based NicOx opened U.S. headquarters in Warren, N.J. The 6,000-squarefoot facility serves as the base for NicOx's commercial affairs and U.S. clinical operations departments.

      "As NicOx transitions from a pure research and development organization to a fully integrated commercial company, we recognized the need to open NicOx' U.S. headquarters," says Damian Marron, VP of corporate development for NicOx. "As we move closer to our anticipated new drug application filing for naproxcinod, we will need to build our sales and marketing infrastructure."

      Most of NicOx' clinical trials are planned and will be conducted in the United States. This process will help implement the company's U.S. clinical strategy and provide strong support and management for the CROs working for NicOx.

      Da sich Naproxcinod so langsam der bevorstehenden Zulassungsphase nähert, erscheint es an der Zeit, eine erste Position ins Depot zu legen.
      Bei PDL sollte nach Abschluss des 2. Quartal mit Lizenzeinnahmen so um die 100 mUSD eine Erholung überfällig sein.
      Bei Elan werden im Juli mit Hochspannung die Daten der Phase2-Studie des Alzheimer-Antikörper Bapineuzumab erwartet , für den PDL im Falle einer Zulassung ebenfalls Lizenzeinnahmen generieren würde.

Und nun, im Borsorama-Artikel, wird dieser Termin mit Mitte 2009 angegeben. Derartige Verzögerungen werden im Normalfall von der Börse abgestraft. An den bisherigen Studienergebnissen ändert dies jedoch nichts.

      Acorda kann mit grosser Wahrscheinlichkeit die Zulassung von Fampridine-SR erwarten, mit dessen Unterstützung die Gehfähigkeit bei MS überaus positiv beeinlusst werden soll. Da dieser Wirkstoff mit Elans Technologie entwickelt wurde sowie bei Elan in die Produktion gehen wird, dürfte Elan zukünftig (ab Ende 2009) selbst massiv an diesem angehenden Blockbuster partizipieren.

      Bei PDL ist der Interims-CEO zurückgetreten. Anscheinend gab es innerhalb der Führungsmannschaft in diesen turbulenten, richtungsweisenden Zeiten unüberbrückbare Meinungsverschiedenheiten, die den Interims-CEO und eine Mitstreiterin zum Rücktritt veranlasst haben.       Elan hat ein sehr gutes p2-Studienergebnis für Bapineuzumab zur Behandlung von Alzheimer vorgelegt. Daraufhin hat der Kurs erwartungsgemäss angezogen und ein neues 5-Jahreshoch realisiert. Detaillierte Einsichten werden áuf der International Conference on Alzheimer's Disease 2008 Ende Juli publiziert.

      PDL hält sich derzeit in einem eher negativen Marktumfeld ausgezeichnet. Mit Bekanntgabe des Quartalsergebnis im August werden weitere Kursgewinne erwartet, wobei die Optionen überproprtional davon profitieren sollten. PDL profitiert selbstverständlich auch von den guiten Bapineuzumab-Daten, da PDL im Falle einer Zulassung auch für diesen humanisierten Antikörper Lizenzgebühren erfhält (3 bis 3.5% auf den Umsatz). Und in Anbetracht des riesigen Potentials in einer zunehmend älter werdenden Gesellschaft verleiht Bapineuzumab auch PDL neue Kursphantasie.

      Bei PDL wirken sich die guten p2-Daten zu Bapineuzumab bisher nicht aus. Das Manmagement scheint jedoch intensiv an einer Neustrukturierung des Konzerns zu arbeiten, weshalb für definierte Ziele (spin-off Wirkstoffentwicklung, Monetarisierung bzw. Verkauf von Lizenzansprüchen auf einzelne Wirkstoffe) für die Topmanager Boni für das Erreichen dieser Ziele ausgehandelt wurden - und dies noch vor Ablauf des Jahres. Falls diese Boni realisiert werden, dürften auch die PDL-Optionen (Ablauf 17.12.2008) noch interessant werden.

      Bei Elan läuft alles nach Plan. Die detaillierten Bap-Ergebnisse auf Ende Juli werden mit Hochspannung erwartet.

      Aufgrund von Nebenwirkungen (bei 6% der Studienteilnehmer wurden Entzündungen der Hirnhaut beobachtet) wurde diese ph2-Studie jedoch beendet. AAB-001 oder Bapineuzumab ist ein auf diesen Erkenntnissen modellierter Antikörper, der gegenüber dem Vaccin den Vorteil hat, nicht über eine aktive (Über)Stimulierung der Immunabwehr die Plaque anzugreifen, sondern passiv, also ohne körpereigene Immunreaktion, den degenerativen Prozess verhindern soll.

      NEW YORK (Dow Jones)--U.S. stock indexes looked to add to gains Wednesday,
      after an unexpected tick up in a private-sector employment report and as oil
      prices continued to fall.

      Oil prices, a key driver of stock action this year, were recently down 52
      cents at $121.65, and about 16% from their closing peak on July 3.

      Financial issues led both a 240-point selloff Monday and a 266-point rally
      Tuesday, as fears about brokers like Merrill Lynch (MER) and hope related to
      its asset firesale preoccupied markets. One market participant said the
      volatility in financial stocks will likely continue because it's driven by
      short-term traders like hedge funds.

      "I think people are still happy to miss some of this news to the upside before
      they back up the truck on some of these financials," said Steve Berte, head
      U.S. equity trader at Standard Life Investments, a Boston long-only money
      manager.

      "I think there's still some uncertainty out there, until we get some pretty
      good conviction behind the move your long-only funds are not going to pile in
      yet. People like us need some fundamental change to the group before we become
      convinced that they've turned around."

      The Standard & Poor's 500 futures rose 8 points to 1270, Dow Jones Industrial
      average futures added 68 points to 11441, and Nasdaq 100 futures were up 15
      points to 1857. A precursor to Friday's non-farm payrolls, the ADP national
      employment report showed growth of 9,000 in the private sector in July.

      The flattish showing follows two tumultuous days that left U.S. stocks roughly
      even at Tuesday's close with where they left off last week. Stocks were as
      strong Tuesday as they were weak Monday, with financials jumping after Merrill
      Lynch & Co. took a decisive step to unload billions of dollars in troubled
      assets, even at a high cost to shareholders.

      Data on U.S. oil inventories are due from the Department of Energy at 10:35
      a.m. EDT.

      The Securities and Exchange Commission late Tuesday extended a rule that makes
      it more burdensome to short-sell the stock of 17 big banks as well as housing
      giants Fannie Mae (FNM) and Freddie Mac (FRE).

      The dollar fell against the Japanese yen and other currencies.

      Earnings are due from Walt Disney Co. (DIS) and Starbucks Corp. (SBUX) after
      the close.

      Shares that could be active Thursday:

      -Fannie Mae and Freddie Mac appeared to benefit from the extension of the
      SEC's short-selling restrictions. Fannie Mae rose 6.9% while Freddie Mac gained
      8% in premarket trading.

      -Elan Corp. PLC (ELN) tumbled in Dublin trading and Wyeth (WYE) dropped 14.7%
      in premarket U.S. trading, after the drugmakers said a Phase II study of an
      Alzheimer's disease drug didn't generate statistically significant results.

      -Viacom Inc. (VIA) shares hadn't traded premarket, but were lower after hours
      Tuesday after the media company reported a 6% drop in second-quarter net income
      and signaled a sharp slowdown in cable-television advertising. The results were
      the first from a major media company and suggested the economic downturn is
      hurting national-television advertising more aggressively than previously
      thought.

      -Shares in Corning Inc. (GLW) were 5% lower premarket, after the company's
      revenue projection for the current quarter fell below analysts' expectations.
      The company reported a surge in second-quarter net income fueled by a $2.43
      billion tax gain.

      -Steel stocks could benefit after the world's top steelmaker, ArcelorMittal
      (MT), said its second-quarter profit more than doubled and issued an upbeat
      outlook for the third quarter. The news came a day after United State Steel
      Corp.'s (X) surprisingly strong results.

      -Moody's Corp. (MCO) said second-quarter net income fell 48%, as the credit
      ratings and research agency continues to struggle in the credit crunch. Revenue
      dropped 25%. The numbers, however, appeared to top analysts' forecasts.

      -Garmin Ltd. (GRMN) posted a 19% increase in second-quarter net income but the
      company lowered its guidance for full-year earnings. Shares fell 11.2% in
      premarket trading.

      PDL hat das erwartet starke Quartalsergebnis abgeliefert. Mit den Lizenzeinnahmen von knapp 105 mUSD auf steigende Umsätze der humanisierten Antikörper ist eine solide Einnahmequelle mit steigendem Potential gegeben. Und mit Actemra (Tocilizumab) steht ein weiterer potentieller Blockbuster kurz vor Markteintritt - Zulassung durch die FDA wird kommende Woche erwartet. Das 2. Quartal wird hinsichtlich der Lizenzeinnahmen allerdings schwächer, da Genentech auf kumulierte Jahresumsätze geringere Lizenzraten abzuführen hat. Die Umsätze des 2.Q, welche für die Lizenzzahlungen im 3. Quartal massgebend sind, wurden von den Lizenznehmern bereits veröffentlicht:

      Die Lizenzeinnahmen im 3q sollten ca. 75 mUSD betragen. Darüber hinaus hat PDL mit Bristol-Myers Squibb (BMY) eine Vereinbarung für eine gemeinsame Entwicklung eines Antikörpers (elotuzumab) zur Behandlung des Multiplen Myeloms (Krebserkrankung des Knochenmarks) abgeschlossen, der PDL eine Vorabzahlung in Höhe von 30 mUSD garantiert sowie weitere Meilensteinzahlungen und Lizenzeinnahmen. Wichtig für die Liquidität des zukünftigen Spin-offs ist die Tatsache, dass BMY 80% der Entwicklungskosten übernehmen wird. BMY erhält darüberhinaus die Option auf einen weiteren Antikörper (PDL241).

      Elan ist nach Veröffentlichung von 2 neuen PML-Fällen unter Tysabri und der Bekanntgabe des p2-Ergebnis von Bapineuzumab total unter die Räder gekommen. Obschon sich der Kurs mittlerweile etwas erholt hat, halte ich die Reaktion für arg übertrieben.
      Mit der erwarteten Zulassung von Johnson&Johnsons Paliperidone palmitate Ende Monat werden die potentiellen Lizenzeinnahmen masiv zunehmen. Dieser Wirkstoff wird der erste am Markt sein, der mit Elans Nanokristalltechnologie eine erhöhte Bioverfügbarkeit einer wasserunlöslichen Aktivsubstanz ermöglicht.

      auf S.7/128 ist die Überlebenszeitanalyse nach KM dargestellt, in welcher nach 24 Monaten eine zunehmende Begünstigung der Verum-Gruppe sichtbar wird. Nachdem die Überlebenszeitanalyse beim Zwischenergebnis von IMPACT bereits einen Überlebensvorteil von 20% erzielt hat, ist eine weitere Zunahme des Überlebensvorteils durchaus wahrscheinlich. Sollte dieser Vorteil beim Überleben 22% nach Abschluss der Studie erreichen, ist eine Zulassung obligatorisch.

      Gold: (9:43) …the study (IMPACT) was designed to be powered at 88% with 304 death events. As I said in my JPMorgan presentation in January, we have reached that number of events, and we anticipate the final analysis of these data by the end of April.

      So what do we know about the IMPACT study and how does it compare to our other studies? Well, there’s a tool we can use in prostate cancer called a Halabi model. You can plug in the demographic factors that the patient has, and Halabi will give you a predicted survival for the patient. All this is is a way for us to predict…are these patient populations similar based on their demographic factors? One of the concerns for IMPACT has been, “is the patient population from IMPACT similar to what we saw in 9901?”

      So what we see here at the top in Study 1 is 9901 (referencing Halibi slide) the median survival predicted by Halabi is 20.1 months. Next is Study 2, which was a smaller study and you can see it was a significantly sicker population, the predicted survival by Halabi there was 18.9 months. The next one is study 1 and 2 together, and Study 3 is the IMPACT study, and this is the final data for Halabi and the median survival is 20.7 months. This is very much in line with the 9901 study, so from a demographic perspective, these studies are similar in their patient populations. I’m going to share with you blinded survival curves from the IMPACT, and I caution you not to read too much into this, but what we see here is encouraging.

      When we talk about the demographics we are talking about these patient populations behaving similarly when we look at the actual Kaplan-Meier curves. The red curve is the combined placebo and treatment arms from 9901. The green curve are the combined blinded data, treatment and placebo arms from the IMPACT study and what we see here is the 9902b or IMPACT study essential overlaps the 9901 study Phase III study. The blue curve is 9902a represents a study where were know patients are sicker, and we see that that curve drops substantially below the other two curves.

      And finally looking at 36 month KM curves the estimates of overall survival again these are combined treatment and placebo estimates. I’ve already shown you the estimates broken out for 9901 when you separate treatment arm and placebo are, but combined…I just want to make sure we’re comparing apples to apples here…9901, 26% of patients were alive at 36 months, 9902A 28%, 9902B very much in line with what we saw in 9901 with a 26% estimate from KM for 36 month survival. So the take home message is that these patient populations appear to be behaving very similarly.

      Then we move on to an interim analysis that we announced in October. The company had as part of its SPA a planned interim analysis of overall survival that was conducted by an IDMC and when they conducted the interim analysis at the 2 year median follow up time timepoint, we saw a 20% reduction in risk of death from the IMPACT study with a HR of .80. So how does that compare to the survival we saw at a similar follow up time from 9901 and 9902a when they were integrated together? It’s very similar. For those two studies at the 2 year follow up point we saw a 22% reduction in the rate of death. So we’re seeing a similar patient population based on demographics and we’re seeing those curves behave similarly based on an integrated analysis of treatment arms and placebo arms, and we’re seeing an interim analysis of overall survival that’s consistent at the two year time point.

      Now for us to hit to our final analysis, which we’re going to announce in April of this year, we need to go from the 20% that we already have to 22%. So how achievable is that for us? How hard is it for us to get from 20% to 22%? Well let’s go back to our data--a potential proxy for what occurred on the previous two studies. When we look at the 9901 and 9902a studies integrated together, 225 patients, we saw that during that two year median follow up time there was a 22% reduction in the risk of death, and by the end of that study that 22% increased to 33%, so there’s a 50% increase of the…risk reduction of death. So in IMPACT we need to go from 20% to 22%, or a 10% improvement.

      Mich würde interessieren, wo du DNDN kursmäßig siehst, falls die Ergebnisse von IMPACT wider Erwarten doch nicht zur Zulassung von Provenge führen ?

      Und damit es rund wird, natürlich auch dein Kursziel für den Fall der Zulassung ..... die Zeiten haben sich geändert, aber mich würde nicht wundern, wenn du das Risiko mit 0,50 $ und die langfristige Chance mit ca. 50 $ bewertest ?

      Die zukünftigen Behandlungskosten für eine Behandlung mit Provenge (enthält 3 Sitzungen) dürften von folgenden Fakten beeinflusst werden:
      Kosten für eine 80mg Taxotere-Infusion €925; normalerweise werden in Deutschland 2 Infusionen pro Sitzung gegeben = €1850 und dies alle 3 Wochen --> kosten pro Jahr = 18 * 1850 = €33.300

      Falls Provenge Taxotere massiv überlegen ist (bei den Nebenwirkungen ist dies sowieso der Fall), dann erwarte ich einen Preis in der Grössenordnung von Taxotere + 10%.

      Bei einer Zulassung und entsprechendem Kapazitätsausbau ist zudem damit zu rechnen, dass allein in den USA bis in einigen Jahren jährlich mindestens 50.000 Patienten mit Provenge behandelt werden. Der Bruttoumsatz daraus würde sicherlich 2 Milliarden $ übersteigen. Mit einer möglichen Ausdehnung auf ROW könnte Provenge bei gutem Verlauf weltweit durchaus in der Spitze 5 Mrd. $ erwirtschaften. Bei einem Grossprofit von 80% ergeben sich daraus orbitante Kursphantasien.

      Als möglichen Vergleich aus der Peer-Gruppe habe ich bereits mehrfach Celgene erwähnt:

      ...nun, Elan ist tatsächlich, neben Dendreon, meine "Lieblingsaktie" (obwohl man zu Aktien keine sentimentale Beziehung haben sollte!), in welche ich sehr viel Herzblut investiert habe. Wieso? Weil Elan, bestückt mit hervorragenden Wissenschaftlern , das Potential hat, die Geschichte der Pharmaindustrie neu zu schreiben. Kein anderes Unternehmen ist meiner Ansicht nach in der Erforschung von neurodegenerativen Erkrankungen so weit wie dieses (nur) mittelständische Unternehmen - selbst mein (wesentlich grösserer) Arbeitgeber ist um Längen zurück. Jedoch, und dies zieht sich wie ein roter Faden durch die jüngere Geschichte, bin ich mir nicht sicher, in wie weit Elan in der Lage ist, diese wissenschaftlichen Leistungen von R&D in gewinnbringende Produkte umzusetzen. Dies soll jedoch nicht heissen, dass das Management nicht fähig ist, vielmehr habe ich mittlerweile gewisse Zweifel, ob Elans Atem lang genug ist - sprich dessen wirtschaftliche Potenz -, um diese beschwerliche Entwicklung, weil nicht frei von Rückschlägen, ohne finanzkräftigen Partner durchzustehen. Elans Wissenschaftler um Dale Schenk und Dennis Selkoe werden, da bin ich mir ziemlich sicher, irgendwann einmal für den Nobelpreis vorgeschlagen, ob diese dann allerdings noch bei Elan auf der Gehaltsliste stehen, kann man bezweifeln. Die finanziellen Mittel dieser Gesellschaft sind endlich und die durch PML gedämpften Zuwachsraten bei Tysabri, mit dem die Entwicklung der neurodegenerativen Pipeline finanziert werden sollte, geben Anlass zur Sorge. Allerdings habe ich die Hoffnung, dass Elans CEO, der ja bekanntlicherweise aus der Finanzbranche rekrutiert wurde, seine Herkunft beweisen kann und Elan die Mittel auf vernünftige Weise beschafft, die dem Unternehmen ermöglicht, die unbestritten grossartigen Leistungen aus R&D in gewinnbringende Produkte (welche der Menschheit grossen Nutzen bringen!) umzusetzen. Es bleibt überaus spannend. Deine Frage jedoch, wie Elan in 1 bis 2 Jahre aufgestellt sein wird, kann ich ehrlicherweise nicht beantworten.

      ...über Provenge und AIPC (androgen independent Prostate carcinom):

      1.) Bei AIPC gibt es ausser Docetaxel (brand name: Taxotere) keinen zugelassenen Wirkstoff, bei dem eine einen lebensverlängernde Wirkung nachgewiesen wurde (bei Docetaxel ca. 2 bis 3 Monate). Unter Provenge war der Überlebensvorteil in Studie 9901 doppelt so gross.


      2.) Da es sich bei AIPC um eine finale Erkrankung handelt, hatten die Studienteilnehmer in dieser doppelt verblindeten randomisierten Studie immer auch die Möglichkeit Docetaxel zu erhalten (Crossover). Diese Daten wurden selbstverständlich ausgewertet und brachten die Erkenntnis, dass Provenge dann die grösste Wirkung entfaltet, wenn vor der Chemotherapie appliziert wurde:

      The analysis was conducted by evaluating the integrated data from the Phase 3 Studies D9901 and D9902A to assess the influence of PROVENGE on clinical outcome in patients who received docetaxel chemotherapy after primary treatment with PROVENGE. Specifically, the analysis evaluated survival data from 82 patients who received docetaxel chemotherapy following initial treatment with either PROVENGE or placebo. The median survival observed in the PROVENGE treated patients who subsequently received docetaxel was 34.5 months compared to 25.4 months for patients randomized to receive placebo who went on to receive docetaxel, a difference of 9.1 months (HR = 1.90; p-value = 0.023).
      Approximately 68 percent of the patients randomized to receive placebo also subsequently participated in a cross-over salvage protocol that allowed them to receive active cellular immunotherapy with APC8015F, a version of PROVENGE generated from cryopreserved cells. The median survival was 25.7 months for patients who received APC8015F followed by docetaxel. In contrast, the median survival was 20.2 months for patients who received placebo only and subsequent treatment with docetaxel, a 14.3 month difference compared to 34.5 month median survival seen in the patients who received initial treatment with PROVENGE followed by docetaxel. These three groups appeared to be well balanced based on their baseline prognostic factors, using an independently validated predictive nomogram (Halabi, et al. Prognostic model for predicting survival in men with HRPC: Journal of Clinical Oncology, 2003; 21(7):1232-7).

      Dendreon hat im Blitzlichtverfahren eine KE durchgezogen und über 200 Mio USD "eingesammelt". Mit dieser Kapitalspritze ist der Ausbau der Produktionskapazitäten zumindest finanziell gewährleistet, wobei auch beim Personal Flagge gezeigt wird, da über 80 ausgeschriebene Stellen die zukünftigen Wachstumsraten vorwegnehmen. Spannend wird sein, ob eine ROW-Partnerschaft vor der Provenge-Zulassung in den USA verkündet wird.

      Mittelfristig werden mit Provenge auch dreistrellige Kurse nicht ausser Reichweite sein.

      ...ich bin mir ziemlich sicher, dass Dendreon noch sehr viel Potential hat und die Marktkapitalisierung mittelfristig durchaus um den Faktor 10 zulegen kann. Man bedenke, dass bisher zur Behandlung von AIPC nur eine Chemotherapie möglich ist, die einerseits einen minimalen lebensverlängernden Effekt hat und andereseits durch beträchtliche Nebenwirkungen die verbleibende Lebensqualität massiv beeinträchtigt. Ich bin deswegen sehr zuversichtlich, dass Provenge nach Zulassung sehr schnell den Markt penetriert und bei entsprechenden Produktionskapazitäten - und diese werden derzeit massiv ausgebaut - innerhalb kürzester Zeit Blockbusterstatus erreichen sollte.

      Aber dennoch sollte ich das Depot etwas diversifizieren und werde deswegen die bis Jahresende gültige Verkaufsorder ändern:

      Seeking approval solely on phase II data is usually foolish except when the FDA says it\'s OK, which is what the agency told Allos.

      The phase II study of Allos\' cancer drug pralatrexate, dubbed \"PROPEL,\" was conducted under a Special Protocol Assessment (SPA) with the FDA. An SPA is essentially an agreement between a drug company and the FDA stating that a clinical trial\'s design, including the size of the trial, the clinical endpoints used and the data analysis plan, are strong enough to form the basis of a drug\'s approval.

      An SPA in no way guarantees a drug approval, however. In this case, the response rate, duration of response and safety profile of pralatrexate needed for approval were not set by the SPA covering the PROPEL trial. All those things are subject to FDA review and will be discussed at the Sept. 1 advisory panel meeting. I happen to think the pralatrexate efficacy data, as we\'ve seen it, is strong enough for approval.

      · Good 2Q09, but focus remains on near-term approval of pralatrexate; reiterate Market Outperform rating and $20 price target on Allos. Allos reported financial results for 2Q09 and provided regulatory and clinical updates for pralatrexate. All eyes remain on the September 24th PDUFA date for pralatrexate in peripheral T-cell lymphoma (PTCL). The FDA has not yet posted an advisory panel to the Federal Register, but Allos is making preparations in anticipation that one will be announced. Our view continues to be that a panel should be assumed and even if the timing does result in a delay to pralatrexate’s approval it will likely be weeks rather than months, preserving our expectations for a 4Q09 launch. We continue to expect approval of pralatrexate on or shortly after the PDUFA date and would remain buyers of ALTH shares before this event. During 2Q08, enrollment was completed in the Phase IIb trial comparing pralatrexate to Tarceva in patients with previously treated nonsmall cell lung cancer. The trial exceeded its enrollment target of 160 patients and recruited 201 patients who were either former or current smokers. The open label trial will assess smoking status (current vs. former and light vs. heavy) as well as histology and is intended to inform the design of a pivotal Phase III trial. Results from this trial are expected to be available in 1H10. Additionally, patient enrollment is continuing in the Phase II single arm, open label trial assessing pralatrexate in patients with advanced or metastatic relapsed transitional cell carcinoma (TCC) of the bladder.



      Patients are continuing to be enrolled in the Phase I trial for pralatrexate in cutaneous T-cell lymphoma with an enrollment target of 56 patients. Finally, the Phase IIa portion of the open label trial for pralatrexate in combination with gemcitabine in relapsed or refractory non-Hodgkin’s lymphoma is ongoing, and results from the Phase I portion of the trial are expected to be presented at ASH.

      · 2Q09 financial update. Allos reported a 2Q09 EPS loss of ($0.19) slightly below our estimate and the Street consensus of ($0.17). Net loss was greater than our expectation of $15.3MM at $16.7MM due to higher than expected operating expenses. R&D expenses were $7.2MM, above our estimate of $5.9MM, G&A expenses were $8.0MM compared to our estimate of $6.8MM while clinical manufacturing spending was below our estimate of $2.7MM at $1.6MM. Allos ended the quarter with cash and equivalents of $105MM following a financing during the quarter that raised
      net proceeds of $47MM. Management updated financial guidance for FY09, increasing its expectations for operating cash burn from a range of $55-60MM to a range of $65-70MM. With $105MM in cash and equivalents at the end of 2Q09, this implies that the company will exit FY09 with approximately $65MM, sufficient to fund our currently projected cash needs in FY10, especially if an ex-US commercial partnership is signed in 1H10 as we anticipate. A summary of actual results for 2Q09 compared to our estimates and changes to our model for FY09 are shown in Figure 1.

      Wie kommst Du zu Deinem Know How in Sachen Biotechnologie -hat das irgendeinen beruflichen Hintergrund? Und schaffst Du es, tatsächlich, mit privaten eigenen Biotechinvestitionen über einen längeren Zeitraum positiv abzuschneiden, wie stark dann auch immer? Weil -unter dem Strich sind mir da würd ich denken noch annähernd keine Leute untergekommen, die das dann auch wirklich schaffen.

      Bei Allos Therapeutics wird es diese Woche sehr spannend, da die Entscheidung durch die FDA über die Zulassung des Krebstherapeutikum Pralatrexate bis zum kommenden Dommerstag erwartet wird (PDUFA 24.09.2009). Nachdem eine von der FDA einberufene Expertenrunde sich am 2.9.09 mit 10 zu 4 Stimmen für eine Zulassung ausgesprochen hat, bin ich vorsichtig optimistisch (Mai 2007, Dendreon lässt grüssen!).

      popeye...bin zwar in der Pharmabranche beschäftigt, jedoch nicht im Bereich klinische Entwicklung. Falls es hier ein Konzept gibt, dann heisst dieses recherchieren, in Frage stellen, recherchieren und nochmals recherchieren. Das Musterdepot ist hinsichtlich Gewichtung i.d.R. ein Abbild meiner realen Investitionen.

      What is new — ALTH announced commercial availability of Folotyn for the treatment of patients with relapsed or refractory peripheral T-cell lymphoma (PTCL). Folotyn pricing is at the high end of prior management guidance of $75k-145K.

      Folotyn Pricing — Average wholesale price for the 20 mg and 40 mg doses are $3,125 and $6,250, respectively. Given that the recommended dose is 30mg/m2/wk for 6 weeks, in 7 week cycles, and assuming that the average patients' (70 kg, 170 cm) surface body area is 1.8 m2, this translates to 54 mg/dose/patient/wk (~3 vials) or a weekly cost of therapy of $9,375. Therefore, Folotyn costs ~$56,250 per cycle (6 weeks on, 1 week off).

      Folotyn pricing represents upside to our estimates — We are assuming 3.7 months of average treatment at $24,000 per month. ALTH pricing is ~ $32,000 per month.

      Commercial efforts — ALTH expects to detail Folotyn with a 25 person specialty sales force initially, and to double the number of sales reps by January 2010. ALTH has also established a patient assistance program (called ASAP) to provide support for drug reimbursement.

      Conclusion(s) — While we anticipate a solid launch and believe the current valuation does not fully reflect the commercial potential of PTCL, we see the data from Phase 2 study of Folotyn in NSCLC (1H10) as the next near-term stock catalyst, which could be viewed as a free option on ALTH at these levels.


      Investment strategy

      We recommend Allos Therapeutics (ALTH) with a Buy/Speculative (1S) rating and a 12-month target price of $15 per share. Its lead drug product, Folotyn (PDX/pralatrexate), an antifolate agent for the treatment of refractory or relapsed Peripheral T Cell Lymphoma (PTCL) patients, received FDA approval September 24, 2009. Folotyn is the first FDA approved drug for the treatment of relapsed/refactory PTCL patients. We have conservatively modeled only Folotyn revenue contributions for PTCL. ALTH is also developing Folotyn in additional indications, such as CTCL, NSCLC, NHL and B cell lymphoma, which may provide upside to our estimates. We have not included revenue contribution from these potential additional indications as well as RH1 for solid
      tumors/NHL in our model.

      Valuation

      Our 12-month target price of $15 is derived from using a 35x P/E multiple on Allos’ 2012 fully-taxed EPS estimate of $0.85 discounted back at 25% per year. We believe using the current median P/E multiple of a range of profitable biotech companies (including large-, mid- and small-cap names) is the most direct and appropriate method, because in our experience all emerging biopharmaceutical companies are valued based on the assumption that they will join the ranks of profitable companies. We are using the current median P/E multiple of this group of profitable companies to minimize uncertainties around earnings visibility.

      Während die Zulassung von Folotyn (Generic name: Pralatrexate) zur Behandlung vpn peripherem T-Zell-Lymphom (PTCL) den Kurs - nebst zusätzlichem Potenzial nach oben - nach unten sehr gut absichert , ist die ganz grosse Phantasie von der Wirkung von Pralatrexate bei zusätzlichen Indikationen abhängig, und zwar beispielsweise bei nicht-kleinzelligem Lungenkarzinom. Sollte Pralatrexate bei dieser Indikation bessere Ergebnisse liefern als Tarceva, dann dürfte auch bei Roche ein verstärktes Interesse an Pralatrexate registriert werden, zumal Tarceva (generic name: Erlotinib) mittlerweile auch Blockbuster-Status erreicht hat.

      Zum Jahresende wurde das Depot modifiziert und zwar wurde die Hälfte des Bestandes von ALTH zum Kauf von 5000 Aktien des kanad. Biotech-Unternehmens Labopharm "geopfert". Diese Umschichtung ist nicht in einer potentiellen Schwäche von ALTH begründet, sondern vielmehr in der bevorstehenden Entscheidung der FDA über die Zulassung des Antidepressivums Trazodone. Der Wirkstoff ist bereits am Markt etabliert, lediglich die Formulierung stellt eine Weiterentwicklung dar, so dass dieser Serotonin-Wiederaufnahmehemmer nur einmal pro Tag appliziert werden muss.

      Obwohl die Kursentwicklung von Labopharm nach Zulassung von Trazodone fast ein wenig befürchtet wurde, ist diese dennoch enttäuschend. Da Trazodone bereits auch generisch in der ursprünglichen Formulierung angeboten wird, ist das Potenzial sicherlich überschaubar, weshalb die Position vollständig zum Verkauf angeboten wird.

      Allerdings konnten 2000 GenVec noch zu USD 1,82 eingesammelt werden - ursprünglich sollte auch der Verkaufserlös der Labopharm-Aktien für den Aufbau einer prominenten GenVec-Position verwendet werden. Zu GenVec später mehr.

      Allos Th. und Dendreon kommen nun wohl bald an einen entscheidenden Punkt wo sich die Richtung wo es hingehen wird zeigen sollte:confused:.
      Ich selbst habe immer noch Wilex im Depot wo eigentlich Mitte des Jahres auch wichtige Entscheidungen fallen werden. Was sagt Deine Spürnase zu diesem Wert und dessen Forschungsergebnissen?

      Ko0mmentar:
      Roche wird wohl demnächst für T-DM1 zur Drittlinienbehandlung bei metastatischem Brustkrebs einen Zulassungsantrag bei der FDA einreichen. T-DM1 (Trastuzumab-DM1-Konjugat) ist eine "intelligente" Weiterentwicklung des monoklonalen Roche-Antikörpers Trastuzumab (Herceptin), mit welchem die Basler derzeit einen Jahresumsatz von über 5 Milliarden CHF (2009: 5,266 Milliarden CHF) generieren. T-DM1 verbindet den Antikörper über eine Brücke (Linker) mit einem hochtoxischen Zytostatika (Maytansin). Der Vorteil des Antikörper-Arznei-Konjugates ist der kombinierte Wirkmechanismus sowohl des Antikörpers als auch des Zytostatikas. Der Antikörper bindet an die HER2-Rezeptoren der Brustkrebszellen und wird in die Krebszelle internalisiert. Das gebundene Toxin (DM1 oder Maytansin) wird erst in der Zelle freigesetzt und entfaltet dann seine hochgiftige, den Zelltod verursachende Wirkung. In einer Phase-2-Studie konnte eine viel versprechende Wirksamkeit bei einer sehr geringen Toxizität nachgewiesen werden, so dass Roche möglicherweise eine Zulassung 3rd line aufgrund dieser Daten bei der FDA beantragen wird. Mittelfristig könnte Roche mit T-DM1 die Anwendung von Trastuzumab vollständig ersetzen, und somit die Auswirkungen des auslaufenden Patentschutz von Trastuzumab (2014?) auf ein Minimum beschränken. Da Immunogen mit 6% an den Umsätzen beteiligt sein wird, ergeben sich alleine für diesen Wirkstoff glänzende Aussichten.

Immunogen wird allerdings erst bei Zulassung weitere Meilensteinzahlungen von Roche erhalten und danach vom Umsatz 6%. Das dürfte den Kurs jedoch zumindest in zweistellige Höhen beflügeln.

      The word “validation” is often used to accentuate the importance of events for small drug makers. In the case of ImmunoGen, which is approaching the filing and regulatory scrutiny of the most important candidate to emerge from its R&D labs to date, the phrase is no exaggeration.
      The drug in question is T-DM1, partnered with Roche, which is expected to file for approval from the FDA in third-line, metastatic breast cancer any time now. If the antibody-drug conjugate reaches the market great things are expected – the drug is being viewed as Roche’s Herceptin follow on. However Dan Junius, ImmunoGen’s chief executive, speaking EP Vantage at Asco earlier this month, says the company is focusing its attention on new pipeline candidates, which will no doubt be given a boost-by-association if all goes well for T-DM1.
      Another partnership?
      Mr Junius was at Asco to see the first presentation of phase I data on a new candidate emerging from ImmunoGen’s pipeline, IMGN388. Like T-DM1 the compound is an antibody-drug conjugate, this time consisting of the company’s cytotoxic agent DM4 attached to an antibody that targets integrin-expressing cells. Integrin is expressed on a wide variety of solid tumours and also on endothelial cells in the process of forming new blood vessels, this attracts the antibody and allows the cytotoxic agent to be delivered directly to tumours.
      “We are still in dose escalation, but the toxicology is looking good,” Mr Junius says. “It is early days, but we are already seeing stable disease.”
      The antibody ImmunoGen is using was licensed from Johnson & Johnson’s Centocor, which has the right to opt in on IMGN388 at an undisclosed early stage of development. The companies would then jointly develop the drug under a cost/profit share deal. With further data due on this compound later this year, hopes are growing that another ImmunoGen candidate is going to be snapped up by big pharma.
      Growing pipeline
      ImmunoGen’s pipeline, although early-stage, is certainly growing with a crop of new candidates moving into the clinic.
      “TDM-1 is not where we spend all our time, we are building our own proprietary pipeline. We have six candidates in the clinic with our technology, and might have 12 by the end of the year,” Mr Junius says, either in their own labs or the labs of partners.
      “We have really gained traction over the last year, and we are at the early stages of being able to demonstrate the full potential of our technology.”
      IMGN388 is not the only new candidate attracting attention. Also being watched closely are SAR3419, a phase I conjugate partnered with Sanofi-Aventis in phase I trials in non-Hodgkin’s lymphoma, and IMGN901 (huN901-DM1), in phase II studies in various solid tumours including small cell lung cancer.
      Fate intertwined
      Despite this full pipeline of follow-on candidates the fate of ImmunoGen is still closely linked to that of T-DM1; success will bring milestone payments and royalties. Happily, many are confident of success, including Roche, which has predicted sales of SFr2bn to SFr5bn ($1.8bn-$4.5bn), depending on the breadth of clinical activity it can demonstrate.
      Financial analysts have pencilled in sales of $330m by 2016, according to consensus data from EvaluatePharma. However, this figure masks some very different opinions suggesting if further data is positive and a launch happens next year as expected, albeit it in a niche third-line setting, consensus will rise.
      The conjugate joins the antibody Herceptin, widely used in Her2 positive breast cancer, with a proprietary chemotherapy agent DM1, developed by ImmunoGen. The company had to develop its own suite of highly toxic agents for these conjugates because currently available chemotherapies are not toxic enough at the low dosages used; just one of the barriers ImmunoGen had to overcome to get to this stage.

      Roche has said it plans to file for approval in a third line metastatic breast cancer setting this year, on the basis of phase II data. This is a patient population with no options left to try, hence the hope that this trial will be enough for approval (Roche picks up pace on ImmunoGen antibody drug conjugate, April 15, 2010).
      Broader utility
      Of course, T-DM1 will really only fulfil its blockbuster potential if evidence of broader utility is generated, and much data is due over the next couple of years. The most important on the horizon are phase II data due to be presented at the European Society for Medical Oncology meeting in October, the first look at a study pitting T-DM1 against Herceptin, in a first line setting.
      It will be considered a big win both clinically and commercially if T-DM1 shows equivalent efficacy, but without the toxicity or cost of the chemotherapy that must be given with Herceptin. Because T-DM1 is central to Roche’s strategy to extend its $5bn a year Herceptin franchise, a good showing would indicate the company might be able to price its follow-on candidate at a premium.
      Of course, phase III data is the ultimate proof, and one pivotal trial is already ongoing in a second line setting, pitting the drug against Xeloda and Tykerb in metastatic patients who have already failed on Herceptin. The study, called Emilia, is due to report in 2012. A second pivotal study in a first line setting is due to begin this year.
      True validation

      ImmunoGen currently boasts a market value of $555m, with a share price of $9.76, up 20% this year. For a company with a phase III product that is being touted as a follow-on blockbuster by the biggest oncology player in the world, a suite of earlier-stage candidates and not to mention full antibody generating capabilities, this valuation is arguably on the low side.
      However, ImmunoGen has been around for 29 years now, and it has taken a long time to get to this stage. A lot of potential is seen in antibody-drug conjugates, but the road to validation has indeed been long, is still being travelled and success is not in the bag just yet (Therapeutic focus – antibody drug conjugates set to improve on poor track record, May 29, 2009).
      Still, with the opportunities approaching and the $80m raised a few months ago, Mr Junius believes the potential for self sufficiency is on the horizon. That would be true validation indeed.

      Bei Immunogen wird demnächst eine positive Entscheidung über eine beschleunigte Zulassung von der FDA erwartet, so dass TDM1 zur Drittlinienbehandlung von Her+mBC möglicherweise noch vor Weihnachten 2010 zugelassen wird.
      Bei Dendreon läuft alles nach Plan, so dass bei den beiden Positionen derzeit kein Handlungsbedarf besteht.

      Levervmann, nachdem Genentech einen "Refuse to File Letter" von der FDA für T-DM1 erhalten hat, ist die Immunogen-Aktie um fast 50% abgestraft worden. Mit den grundsätzlichen Möglichkeiten von TDM1 hat dies jedoch nichts zu tun, da die FDA lediglich dem Umstand berücksichtigt hat, dass die Patientinnen in der vorgelegten Studie doch nicht vollst$ndig austherapiert waren und deswegen ein Zulassungsantrag mit Phase-II-Daten nicht zulässig ist. Auch wenn diese Begründung für mich logisch nicht plausibel ist - die HER2-positiven Patientinnen der vorgelegten Studie wurden lediglich nicht mit HER2-negativ indizierten Wirkstoffen behandelt -, hat dies zur Folge, dass die Drittlinienbehandlung dieses Jahr nicht zugelassen wird.
      Nichtsdestotrotz werden nächste Woche in Mailand beim Jahrestreffen der europ. Gesellschaft für klinische Onkologie (ESMO) weitaus wichtigere Daten veröffentlicht, und zwar die Ergebnisse einer Phase-2-Studie bei Erstlinienbehandlung von metastatischem Her2-positivem Brustkrebs:

      Nächste Woche werden in Mailand beim Jahrestreffen der europ. Gesellschaft für klinische Onkologie (ESMO) wichtiger Studienergebnisse veröffentlicht, und zwar die Ergebnisse einer Phase-2-Studie bei Erstlinienbehandlung von metastatischem Her2-positivem Brustkrebs mit T-DM1 im Vergleich zur Standardtherapie mit Herceptin und Docetaxel:

      Es ist auf alle Fälle zu erwarten, dass TDM1 bei den Nebenwirkungebn weitaus günstiger ist als die Standardtherapie mit dem Zytostatika Docetaxel. Sollte die Wirkung zumindest nicht schlechter sein als die Standardtherapie, dann dürfte hier ein neuer Stern aufgehen, welcher von allergrösster Bedeutung für den Fortbestand von Immunogen sein sollte.

      Bei Depotneuling Delcath hat ein potenzielles Übernahmegerücht von Bristol-Myers-Squibb den Kurs positiv beeinflusst. Obschon natürlich reichlich Übernahmephantasie vorhanden sein dürfte, ist dieses neuerliche Gerücht wohl eher mit dem bevorstehenden hexensabbat (Ablauf von Optionen) in Verbindung zu bringen.

      Da Delcath sich jedoch in der klinischen Onkologie mit einer "bahnbrechenden" Innovation zu etablieren scheint (dazu später mehr), ist für die Aktie reichlich Potenzial nach oben vorhanden.






      Conclusion

      There is actually no great treatment for any liver cancer. In our opinion DCTH's system will be the gold standard of care, and will open up numerous opportunities for other big pharmaceutical and biotech companies which could use this system to benefit and test other cancer drugs.

      The Market for USA only is estimated at roughly $750 million, and $5 Billion for off lable use. One other crucial point is that the drug delivery system is believed to be effective with most organs and cancers. The process also prevents chemotherapy drugs from getting into the patient's blood, thereby lessening damage and side effects. The potential market for Delcath's product is enormous. Management is highly motivated to succeed and seems to be on the right path.

      We believe the recent decline in the value of DCTH's shares is an overreaction, certainly due to the stock offering and the little delay to submit the rolling NDA. The fundamental outlook for Delcath company remains strong. They have excellent clinical data, significant market opportunity and a strong balance sheet with enough cash for at least the next 12 months. In addition, Delcath continues to pursue negotiations with potential partners for international marketing rights for the Delcath PHP system.

      At $7.25 and a market cap of only $273 million, for a company with such great potential, we think this is a great opportunity. We target the $25 for the run-up before PDUFA date which we estimate to be around June-July 2011.

      TDM1 ist ähnlich wirksam bzw. sogar noch ein wenig wirksamer (bei der Studiengrösse natürlich nicht statistisch signifikant) als die Standardbehandlung mit Docetaxel und Herceptin und dies bei weitaus geringeren Nebenwirkungen.
      Auch wenn Wall Street dieses Ergebnis nicht richtig würdigen wird, hier ist ein neuer Stern aufgegangen. Die klinische Onkologie darf sich freuen.
      Nun ist die IMGN-Aktie eindeutig als langfristiges Investment geeignet und werden deswegen gehalten!

      Allein die Tatsache, dass die 343. Wiedererkrankung nach meiner Sicht unerwartet lange dauert, lässt ein vielversprechendes Auswertungsergebnis vermuten. Hinsichtlich der gesamten Produktlinie halte ich Wilex für deutlich unterbewertet.

      In der Phase III-ARISER-Studie mit RENCAREX® erwartet WILEX die 343. Wiedererkrankung – den nächsten im Studienprotokoll
      definierten Meilenstein – voraussichtlich bis Jahresende. Danach werden alle vorhandenen Daten der 864 Patienten

      Zu Immunogen und ESMO:

      Die Ergebnisse der TDM1-ph2-Studie im Vergleich zur Standardtherapie (Docetaxel + Herceptin) --> TDM4450g sind hervorragend:

      Wirsamkeit:
      vorläufige Ergebnisse belegen eine "overall response rate" (ORR) von 47.8% bei TDM1 gegenüber 41.4% bei der Standardtherapie; die endgültige Studienanalyse einschl. Bestimmung des primären Endpunktes (progressionsfreies Überleben) wird auf Mitte 2011 erwartet

      Nebenwirkungen:
      TDM1 scheint ein wesentlich günstigeres Nebenwirkungsprofil zu haben als die Standardtherapie: z.B. nur 1.5% gegenüber 66.1% der Studienteilnehmerinnen verlieren ihre Haare, lediglich 7.5% gegenüber 57.4% sind von einer Neutropenie, also der Verminderung neutrophiler Granulozyten im Blut, betroffen.

      In der Summe aller schwerwiegenden Nebenwirkungen (Grad 3 und 4) sind unter TDM1 lediglich 37% gegenüber 75% der Studienteilnehmerinnen unter Standardtherapie betroffen.

Als wichtiger Bestandteil des NDA wurde eine mehrjährige Liefervereinbarung mit Synerx Pharma, LLC, und Bioniche Teoranta, einer Tochtergesellschaft von Mylan Inc., abgeschlossen, in Folge derer Delcath mit Melphalan-Hydrochlorid ( "Melphalan") beliefert wird.
      Kurse um USD15 sollten bis Ende Jahr möglich sein.

      Dies dürfte auch für T-DM1 eine zusätzliche Indikation möglich machen, obschon die Entwicklung bzw. die Klinik wohl noch einige Jahre in Anspruch nehmen wird.

      Curis könnte im Laufe des kommenden Jahres sehr erfolgreich sein, wenn der von Roche einlizenzierte "Hedgehog Pathway Inhibitor" GDC-0449 (Vismodegib) in einer Pivotalstudie zum "Basal Cell Carcinom" (BCC) erfolgreiche Daten liefert.       Pharmakokinetische Phase I-Studie zu dem Sonic Hedgehog-Antagonisten GDC-0449 bei pädiatrischen Patienten mit rezidivierendem oder refraktärem Medulloblastom: Eine Studie des Pediatric Brain Tumor Consortium (PBTC 25) (Abstract Nr. CRA9501) – Samstag, 5. Juni 2010, 16:45 – 17:00 Uhr CST, Raum S504.

      Diese Studien liefern Ergebnisse zu GDC-0449 (RG3616), einem oral verabreichten kleinmoleküligen Inhibitor des Hedgehog-Signalweges. Dieser wird bei bestimmten Krebsarten wie Kolorektalkarzinom, Medulloblastom (einer Art von Hirntumor) und Basalzellkarzinom (einer Art von Hautkrebs) in pathologischer Weise aktiviert. Man geht davon aus, dass GDC-0449 die überaktive Hedgehog-Signalgebung innerhalb der Krebszelle selbst (ausgelöst durch Mutationen im Signalweg), aber auch in den sie umgebenden Zellen (ausgelöst durch eine Hedgehog-Überexpression) hemmen kann.

      Genentech und Roche untersuchen derzeit in Zusammenarbeit mit dem Kollaborationsvertragspartner Curis, Inc. GDC-0449 in Phase II-Studien bei verschiedenen Tumorarten sowie in Kombination mit zugelassenen Behandlungen.



      Bei den ph2-Studien handelt es sich teilweise um Pivotal-Studien, so dass auf deren Grundlage ein Zulassungsantrag eingereicht werden kann, sofern diese Studien in ihren Endpunkten entsprechen.

      In February 2009, Genentech initiated this pivotal Phase II clinical trial of GDC-0449 as a single-agent therapy for patients with metastatic or locally advanced basal cell carcinoma (BCC). Genentech expects to evaluate GDC-0449 in approximately 100 patients with metastatic or locally advanced BCC in a global single-arm, two-cohort clinical trial. One cohort includes all patients with histologically-confirmed, RECIST measurable metastatic BCC. RECIST provides standard parameters to be used when documenting patient response for solid tumors. The second cohort includes histologically-confirmed, locally advanced BCC that is considered inoperable by the treating physician. All patients will receive a daily oral dose of GDC-0449. This Phase II pivotal study builds upon the Phase I safety and efficacy data demonstrated by GDC-0449, which showed clinical benefit in a substantial proportion of advanced BCC patients. There is currently no standard of care for patients with these types of BCC and Genentech has indicated that it designed this pivotal trial so that the data, if positive, may serve as the basis for NDA submission by Genentech. Roche has projected that data results will be available in the first half of 2011 and that, assuming favorable results, a potential regulatory submission for GDC-0449 in advanced BCC could also occur in 2011.

      In den letzten Jahreswochen werden vor allem bei Delcath kursbewegende Nachrichten erwartet und zwar die Einreichung des NDA bei der amerikanischen Arzneimittelbehörde FDA für Melphalan in Verbindung mit Delcath's Chemosaturation System zur Behandlung von Lebermetastasen.
      Die Immunogen-Aktie erhält Aufwind von Gerüchten, dass Roche angeblich an einer Übernahme interessiert sei. Da Novartis jedoch vor kurzem erst eine umfangreiche Kooperation mit Immunogen bekanntgegeben hat, ist eine Übernahme durch den Basler Konkurrenten trotz TDM1 zum jetzigen Zeitpunkt wohl mehr ein wünschenswertes denn realistisches Szenario - wobei jedoch ab und an auch Wünsche in Erfüllung gehen!

      Bei Curis sei noch einmal darauf hingewiesen, dass es sich um eine hoch spekulative Investition handelt, da der von Curis an Roche linzenzierte Hedgehog-Signalweg-Hemmer (Hedgehog pathway inhibitor) GDC-0449 (generic name: Vismodegib) bisher bei 2 Indikationen (Eierstock- und Darmkrebs) keine signifikanten Verbesserungen gezeigt hat. Da Roche jedoch die Entwicklung trotz dieser Negativergebnisse in weiteren Indikationen vorantreibt, besteht immer noch die Chance auf Zulassung vor allem beim "Basal Cell Carcinoma".

Im Unterschied zur FDA, welche Delcath Technologie wie ein Arzneimittel beurteilen wird, haben die europäischen Arzneimittelbehörden entschieden, Melphalan in Verbindung mit der Terchnologie als medizinisches System bzw. wie ein Apparat zu klassieren.

      Bei Delcath lässt der für 2010 angekündigte Zulassungsantrag bei der FDA für Melphalan/Chemosaturation auf sich warten, weshalb eine gewisse Nervosität am Markt zu beobachten ist:

      Je länger es mit dem Zulassungsantrag dauert, umso mehr dürfte der Druck auf nervöse Marktteilnehmer zunehmen und Kleinanleger mit StopLoss in Bedrängnis bringen. Es sei nur an das Dendreon-Feuerwerk erinnert, wonach durch eine StopLoss-Spirale der Kurs innerhalb 30 Minuten massiv nach unten gedrückt wurde. Dominante Marfktteilnehmer werden wohl immer bemüht sein, derartige Stress-Situatiuonen zu Ihren Gunsten auszunutzen.
      Deswegen ist in hochspekualtiven Märkten, vor allem wenn eine gewisse Marktenge gegeben ist - und bei den allermeisten KMUs der Biotechnologie ist dies der Fall - ist m.E. die "Absicherung" mit StopLoss eher kontraproduktiv.

However, primary or acquired resistance appears to present a major challenge to the success of inhibitors targeting the PI3K pathway due to the existence of redundant and compensatory pathways in cancer cells. Curis scientists believe that CUDC-907's synergistic inhibition of HDAC and PI3K may enhance anti-tumor activity and overcome these limitations through durable blockade of cancer networks as opposed to single target inhibition.

      About CUDC-907

      CUDC-907 is an orally bioavailable, multi-targeted small molecule that is designed to inhibit HDAC and PI3K, the combination of which Curis scientists believe have synergistic interaction against cancer cells. In vitro mechanism of action studies demonstrate that CUDC-907 is able to inhibit Class I PI3K and upregulate molecules involved in cancer cell death. CUDC-907 has also demonstrated the ability to suppress multiple nodes of other survival pathways as a result of the epigenetic modification resulting from the inhibition of its non-kinase HDAC target. Das ist zwar ein kleiner aber feiner Unterschied, d.h. beim Zulassungsantrag ist nachzubessern. Dies ist zawr äusserst unschön, ist aber immer wieder bei kleineren Unternehmen zu beobachten, die einen deratigen Zulassungsantrag zum ersten Mal einreichen - es sind "Tonnen" an Papier einzureichen!

      Ich bleibe vorsichtig optimistisch!
