Wirk-Mechanissmus:
Phagen befallen die Einzeller (Bakterien) und nutzen deren Stoffwechsel um sich rasant zu vermehren

Zielgruppen Beispiele:
-in jedem Krankenhaus befinden sich Multi Resistente Staphylococcus -areus Keime (MRSA)
jedes Jahr sterben allein in D 23.000 Patienten an diesen Infektionen.
-Chronische Sinusitis,….
-Bakterien-Infektionen jeder Art
-neue Antibiotika wurden in letzter Zeit kaum noch entwickelt (wobei meinen Wissens Cempra, Tesaro und Karyopharm evtl. an Mitteln gegen Infektionen arbeiten, daß muß ich aber noch recherchieren)

in der Nahrungsmittelindustrie werden Phagen-Sprays zur Bekämpfung von Krankheitskeimen

Sicherheitsbedenken:
Viren sind ja allgemein als Krankmacher bekannt.
Phagen zählen zwar zu den Viren, haben aber nach heutigem Wissen, keinen Anteil an
Krankheiten.
Phagen sind überall, auf unserer Haut, im Darm,..
ein einziges Gramm Joghurt oder Rohmilchkäse soll hundert Millionen enthalten.
Phagen gelten als sehr sicher und darum wurde bereits 2013 ein Phagenspray von der FDA
für Lebensmittel zugelassen.
in den NL ist sogar die Konservierung von Bioprodukten erlaubt.

Business Update

AmpliPhi Biosciences Corp. ( APHB) is developing bacteriophage therapeutics for the treatment of bacterial infections. The company’s pipeline consists of treatments that specifically target infections caused by Staphylococcus aureus (AB-SA01), Pseudomonas aeruginosa (AB-PA01), and Clostridium difficile (AB-CD01). AmpliPhi has recently announced a number of updates for both AB-SA01 and AB-CD01, which are summarized below.

Bacteriophages

Bacteriophages are viruses that infect, replicate in, and kill bacterial cells while not harming eukaryotic cells. This exclusivity is vital to their use in combating infectious diseases, and their unique mechanism of action compared to traditional antibiotics means they can be used in place of or in conjunction with those therapies.

While bacteriophages have been known for over 100 years, up until now their use as anti-infectious agents has been restricted to limited geographic regions (mainly Eastern Europe and Russia). This is due to the rise of antibiotics during the middle part of the 20 th century that proved to be cheap, plentiful, and effective therapeutics. However, the rise of antibiotic resistant organisms has resulted in a clear need for alternative treatment options, specifically for ones that no longer respond to once effective antibiotic therapies.

Bacteriophage therapies potentially hold a number of advantages over antibiotics, including:

- Multiple modes of application
- Less severe side effects
- Localized and highly specific effect
- Very low doses
- Can treat antibiotic-resistant strains
- Unique mechanism of action makes resistance less likely

AB-SA01 Update

AB-SA01 is being developed for the treatment of Staphylococcus aureus infections, including methicillin-resistant S. aureus (MRSA) infections. It contains three phage constituents that target greater than 96.7% of S. aureus isolates from a global diversity panel.

AB-SA01 in Chronic Rhinosinusitis.

On November 19, 2015, AmpliPhi announced a clinical trial research agreement has been entered into with the University of Adelaide for the purpose of conducting a Phase 1 clinical trial of AB-SA01. The study is titled “A Phase 1 Investigator Initiated Study to Evaluate the Safety, Tolerability and Preliminary Effectiveness of AB-SA01 in Patients with Chronic Rhinosinusitis Associated with Staphylococcus aureus infection”.

The trial is being conducted at the Queen Elizabeth Hospital, Adelaide, Australia and AmpliPhi is supplying the bacteriophage drug product for the clinical study. In addition, AmpliPhi is providing scientific input, protocol recommendation, and assisting the study’s principal investigator with data management support. The trial is expected to enroll nine patients who suffer from chronic rhinosinusitis (CRS) and are also positive for S. aureus by nasal swab. Since this is a Phase 1 study, the primary endpoint is safety, however the company will also be collecting anecdotal evidence about how bacteriophage treatment affects the patients symptoms.

As part of the standard of care these patients flush their sinuses twice a day with a sterile saline solution. The trial involves the addition of different concentrations of bacteriophage to the saline solution that the patients use to flush out their sinuses. Cohort 1 was dosed with 108 phage/mL in sterile saline twice daily for one week. Cohort 2 was dosed with 108 phage/mL in sterile saline twice daily for two weeks. Cohort 3 is currently being dosed with 109 phage/mL in sterile saline twice daily for two weeks.

On September 20, 2016, the company provided an update on the Phase 1 trial. Thus far, seven patients have completed dosing and the final two patients should be enrolled into the trial very soon. The preliminary safety results from the first seven patients show that AB-SA01 was well tolerated as there was no change in body temperature following treatment and no difference in blood panels before or after treatment. In addition, no drug-related serious adverse events were reported.

Importantly, the company announced that eradication (=AUSROTTUNG) of S. aureus infection was seen in two out of three patients in Cohort 2. None of the patients in Cohort 1 experienced eradication of S. aureus. Patients in both Cohorts 1 and 2 reported improvements in symptoms on days 0, 7, and 14 by Visual Analog Scale (VAS) and Sino-Nasal Outcome Test (SNOT-22) score.

These preliminary results are very encouraging, and with eradication seen with low-dose phage treatment for 14 days, it stands to reason that very similar results should be seen in Cohort 3 as they are receiving high-dose phage for 14 days. We anticipated bacteriophage treatment being safe and well-tolerated, as bacteriophages are ubiquitous in the environment and people are constantly exposed to them, but it is encouraging to see administration of bacteriophage does not result in any unanticipated safety signals. We anticipate full results from this study before the end of 2016.

AB-SA01 for Wound Care

In addition to treating CRS, AmpliPhi is also developing AB-SA01 in collaboration with the U.S. Army as a topical treatment for wound infections. On September 13, 2016, AmpliPhi announced topline results from a Phase 1 clinical trial designed to test the safety and tolerability of AB-SA01 administered topically to the skin. A total of 12 healthy adult volunteers were dosed in two cohorts of six participants each, with each volunteer receiving either low-dose (1x108 PFU/mL) or high-dose (1x109 PFU/mL) of AB-SA01 on the forearm under an occlusive bandage. Placebo was administered to the opposite forearm, thus allowing each participant to serve as his or her own control. AB-SA01 and placebo were administered daily for three consecutive days. Results showed that AB-SA01 was well-tolerated and that no drug-related serious adverse events were reported. This study was required before a trial involving wounds infected with S. aureus could get underway, and we are glad to see that it was completed successfully with no unanticipated safety signals arising.

AB-CD01 Update

AB-CD01 is under development for the treatment of Clostridium difficile infections. C. difficile is a gram-positive, spore-forming bacterium that is best known for causing antibiotic-associated diarrhea. According to the U.S. CDC, It is estimated to cause upwards of 300,000 infections and 30,000 deaths every year in the U.S.

There are only three antibiotics that are utilized for the treatment of C. difficile infections, thus there is a need for more treatment options as relapse rates are very high. Approximately 20% of patients will have a relapse infection, and once a patient has had one recurrence the chance of having another recurrence rises to 65%.

On September 12, 2016, AmpliPhi announced the publication of data from a preclinical study in both in vitro and in vivo models of C. difficile infection (Nale et al., 2016). For the in vitro studies, a 4-phage cocktail was added to pre-established biofilms of C. difficile and live cells were quantified following treatment. The following chart shows an approximately 0.5-1.0 log reduction in vegetative cells for 1-4 day old biofilms, however there did not appear to be an effect on spore formation.

In summary, bacteriophages have several characteristics that make them potentially attractive therapeutic agents. They are (i) highly specific and very effective in lysing targeted pathogenic bacteria, (ii) safe, as underscored by their extensive clinical use in Eastern Europe and the former Soviet Union and the commercial sale of phages in the 1940s in the United States, and (iii) rapidly modifiable to combat the emergence of newly arising bacterial threats. In addition, a large number of publications, some of which are reviewed in this minireview, suggest that phages may be effective therapeutic agents in selected clinical settings. Granted, many of these studies do not meet the current rigorous standards for clinical trials and there still remain many important questions that must be addressed before lytic phages can be widely endorsed for therapeutic use. However, we think that there is a sufficient body of data—and a desperate enough need to find alternative treatment modalities against rapidly emerging, antibiotic-resistant bacteria—to warrant further studies in the field of phage therapy.

They were able to show the M13 phage dissolved amyloid-beta plaques when the phage was delivered through the rodents’ nasal passages. The phage even worked against the amyloids in prion diseases (a class that includes Creutzfeldt-Jakob disease). Im Osten hat das ganze angefangen, als es damals noch keine Antibiotika gab.
Erst als das Penicillin 1928/29 entdeckt und mit Ausbruch d. WW2 in Massen prodziert wurde,
ist das ganze im Sande verlaufen.

Wer weiß, in welchem Umfang es jetzt wieder ausgegraben wird, nachdem immer
mehr Superbugs resistent werden/sind.

Wo ist das Geld aus der letzten Kapitalerhöhung (Ende 2016) geblieben?
Wenn jetzt 7M Aktien 1,50 USD verkauft werden, entspricht das einem Anteil von 7/8,65 des Unternehmens. Dieser Teil wird also zu 10,5MUSD verkauft.

Das gesamte Unternehmen wird daher mit 10,5*8,65/7 MUSD=12,975 MUSD bewertet.

Abzüglich der neuen Barmittel bewertet das Management sein Unternehmen aktuell mit etwa 2,5 MUSD, und das trotz aller Vorleistungen und dem Mittelzufluss in 2016. Eine Zumutung für (alt-)Aktionäre.

Zudem soll die neue KE bis zum 10.5. abgeschlossen sein. was soll die Hast?

1) Die der (kleinen) Altaktionäre. Ich habe kein Angebot der Firma erhalten, neue Aktien durch die KE zu erwerben. Das ist schon ziemlich frech. Vor der KE war eine Institution mit 20% dabei, ich hab noch keine Neuigkeiten gefunden, wie hoch die jetzt investiert sind.

Ausserdem wurde ich als Aktionär auch nicht befrgat, ob ich meiner Enteignung zu den Konditionen dieser KE zustimme. Dass das in den USA geht, hätte ich NIE für möglich gehalten (In DE habe ich so einen Scheiss auch schon erlebt). Die Ampliphi-Chefs haben Ihre (kleinen) Eigentümer um ihr eingesetztes Kapital betrogen, weil sie es an die neuen Eigentümer (oder an grosse alte) verschenkt haben.

2) Die der neuen Aktionäre. Wer die Aktien jetzt kauft, kriegt die gesamte Entwicklungsarbeit der Firma und den Einsatz der Altaktionäre quasi geschenkt! Beim aktuellen Aktienkurs gibt es wahrscheinlich sogar den baren Dollar für 80Cent. Und die, die kürzlich mit knapp 10 Mille eingestiegen sind, gehen ziemlich sicher davon aus, dass aus dem Einsatz mehr wird.

Fazit: Interessantes Business, absolut opportunistisches Management, das auf seine Aktionäre scheisst.

Good morning, good afternoon everyone.

As Paul mentioned, our strategy for the next 6 to 12 months is to provide all therapeutic candidates AB-SA01that targets Staphylococcus aureus and AB-PA01 that targets Pseudomonas aeruginosa to patients under the expanded access regulatory guidelines, also known as compassionate use that I established by the FDA in the U.S. and by the Therapeutic Goods Administration in Australia.

As we had previously reported, AB-SA01 demonstrated activity against 97% of multi-drug resistant clinical isolate globally over a multiyear period and AB-PA01 against 70% to 80% of such multi-drug resistance Pseudomonas clinical isolates.

Drug resistance Staph and Pseudomonas bacteria are both on the World Health Organizations priority pathogens list published earlier this year. We plan to provide AB-PA01 and AB-SA01 that upgrade used at our GMP manufacturing facility to patients who suffer from serious or life-threatening multi-drug resistant infections such as endocarditis, bacteremia, prosthetic joint infections, cystic fibrosis, complicated urinary tract infections and others.

Our objective is to treat at least 10 patients with AB-SA01 or AB-PA01 by the end of 2017 and additional patients in early 2018. Helping such seriously ill patients who don't respond to conventional antibiotics will also enable us to demonstrate real-world clinical evidence without therapeutic candidates.

Based on the clinical and microbiological data that we're currently collecting, we plan in the first half of 2018 to select lead indications for further clinical development and in consultation with the FDA and other regulatory agencies define the potential path to regulatory approval.

As you may recall in a meeting with AmpliPhi earlier this year, the FDA expressed a commitment to working with us to design a regulatory path for phage therapies. Positive data over the next 6 to 12 months may enable us to initiate Phase 2 studies of AB-SA01 and/or AB-PA01 which may potentially be registrational studies as early as the second half of 2018.

Implementation of this strategy is proceeding as planned. We recently supplied AB-PA01 to a major U.S. teaching hospital for patient suffering from life-threatening multi-drug resistant Pseudomonas lung infection.

Under an emergency IND along with the FDA, the patient received multiple doses of AB-PA01 administered intravenously and by an inhaler. The treatment was well-tolerated. We're pleased with the results so far and the analysis of the data is ongoing and we anticipate that the academic and clinical team involved in this patient's treatment will submit the detailed results for presentation at the future medical conference.

We achieved several important milestones for AmpliPhi in this case. It was the first-in-human administration of our AB-PA01 therapeutic candidate. It was also the first intravenous and inhaled administration of AB-PA01.

As a final topic, we're pleased to share with you that our manufacturing facility successfully completed the periodic regulatory GMP inspection and our GMP certification was renewed. AmpliPhi has the world's only GMP certified facility that is dedicated to manufacturing bacteriophage therapeutic candidates for human use and this certification enables us to continue supplying drug product to support our strategy.

Now I'd like to turn the call over to Steve Martin for review of our financial results. Steve?

I'd like to share some highlights from our financial results provided in our quarterly report filed with the SEC today and as announced in our press release. In May 2017, we completed an underwritten public offering and we received net proceeds of approximately $9 million after underwriting commissions in all related expenses.

Cash and cash equivalents were $9 million as of June 30, 2017 compared to $5.7 million as of December 31, 2016. With this financing and based on our updated operating plans focused on single patient expanded access treatments, we believe our existing cash resources will be sufficient to fund our operations until mid 2018. The company currently has 8.7 million common shares outstanding.

Bis jetzt glänzten Sie mit Dillution und Resplits!
Der nächste Resplit könnte schon in den Startlöchern stehen.

Andererseits waren alle letzten News zur Technologie an Sich durchwegs positiv.
Der Kurs sollte meiner Meinung nach nicht soooo tief stehen, aber die Investoren fürchten, wie bei
vielen anderen Bio Smallcaps auch, eben einen ständig Verwässerung,… …bis hier was rüberkommt
der Einstiegs-Zeitpunkt ist entscheidend sonst braucht man zu viel Geduld und die hat nicht jeder.

Ich kann mir vorstellen, aktuell den Boden zu sehen. Zum AbwärtsGap wären
dann schnell 0,52USD möglich.

Was AMPH für die Shareholder draus macht, ist eine andere Geschichte.

Hab erst heute wieder einen Bericht gelesen, dass durch die Behandlung
mit einem Phagencocktail die bösen Bakterien ihre Resistenzen gegenüber
den vorher wirkungslosen Antibiotika wieder abbauen können.
Vielleicht weil sie durch den Angriff der Phagen sich mit dem potenziellen mächtigeren

Wie im Artikel beschrieben: der öffentliche Leidensdruck ist noch nicht
groß genug und es springt zu wenig für die Pharma Firmen raus.
Darum zu wenig Phagen Forschung und zu viel Antibiotika im Einsatz

Und hoffentlich wird das nicht zu teuer. Und hoffentlich ist das Management der ehemals private Held C3J besser drauf. Und hoffentlich fängt das Geschäft mal an zu laufen ...

Das sehe ich auch so..
Mgmt. hat Verbesserungspotential.

Die letzten Jahre wurden Kleinaktionäre gemolken, ist aber meist so bei solchen MicroCaps
im Entwicklungsstadium.

Das öffentliche Interesse muss noch viel mehr werden, das ergibt sich aber die nächsten Jahre von allein, wenn mehr und mehr Superbugs auftauchen.
Dann wird der Druck auf die Pharma Companies steigen um in Phagen mehr zu investieren.
Bis Dato ist mit Antibiotika einfach mehr verdient.
Phagen Firmen sind eher Exoten unter seines Gleichen.

Die Studien sind nach wie vor vielversprechend. (84% Treatment Succses)
Mit der eigenen ausgebauten Phagen Prod. und den neuen Möglichkeiten mit C3J (Synt. Phagen)
Scheinen die Weichen für die Zukunft richtig gestellt und es kann hier evtl. was Größeres heran wachsen.

Ich warte einen ggfs. noch anstehenden R/S Split ab, dann steige ich wieder ein.
Vielleicht bringt ja dieses Jahr die Wende und ab 2020 geht's up.
