Summary
Proteostasis Therapeutics completes enrollment quicker than anticipated, now expects data from three 28-day phase 2 studies using doublet and triplet therapy for CF patients by Q4 2019.

Company tumbled on phase 1 CF data for its doublet and triplet studies. However, changes to dosing/clinical trial design/longer treatment duration should achieve a better outcome in lung function improvement.

The triplet therapy of PTI-801/PTI-808/PTI-428 is promising because, at day 14, there was no plateau in lung function improvement observed, which means efficacy could improve with more treatment days.

Proteostasis has a major competitor to overcome, Vertex Pharmaceuticals, which has been a powerhouse in the CF space and had recently received FDA approval for its triplet therapy Trikafta..

Es sieht so aus, dass Vertex nun Konkurrenz im Bereich der Behandlung der Stoffwechselstörung Mukoviszidose, auch zystische Fibrose (CF) genannt, bekommt .

Vertex hat bei seinen Patienten mit seiner "Triple Behandlung "nach 24 Wochen eine Verbesserung der Lungenfunktion von 10% verzeichnet, PTI konnte nach nur 14 Tagen bei Verabreichung einer niedrigerer Dosierung bereits eine 8%ige Verbesserung der Lungenfunktion bei " krankeren" Patienten feststellen.

Dann warten wir mal auf den Dezember - mit hoffentlichen guten results.

Hab jetzt gestern erstmal meine Teile in USA versilbert , Bio Aktien sind halt immer ein heißes Eisen, und stehen und fallen mit den Daten der klinischen Versuchen. Mir persönlich kommt es im Moment
so vor als ob die power etwas verloren geht. Handelt mit Bedacht und nicht zu gierig werden.
Ich werde sie auf dem Schirm behalten.

Hier dann noch enige Zahlenspielereien,von User torture, die ich mal unkommentiert stehen lassen möchte.

Doublet generates about $1 bil in revenues and PTI should be the dominant player there. with 9x revenue (conservative) .
PTI will be $9 bil market cap company once doublet is on the market = $150-$180/share.
At this point im just safely gonna assume that triplet is a total #$%$up and no CF patients wants it.

PTI will still be $150/share.

Volume today after the webcast confirms my thesis because tutes were all over it , making it hard for shorters to cover

So wie es nach den gestrigen Aussagen von CEO Meenu Chhabra nun vermuten lässt, scheinen die Doublet-Daten ( PTI 801 /PTI 808 ) phänomenale Ergebnisse aufzuweisen. Das wäre schon einmal die sehr gute "Oberhammer-Nachricht" wenn diese bei der Veröffentlichung der Daten in den nächsten Tagen bestätigt werden würde.

Was bei den Triplet- Daten ( PTI 428 / PTI 801 / PTI808 ) herauskommt, wird in den USA Foren ( Yahoo , stocktwits ) jetzt mehr als "Bonus" angesehen - die aktuelle Ausgangslage wird grundsätzlich jetzt schon als Win-Win-Situation eingeschätzt.

Wenn dem so ist, sollte wir als PTI-Investoren , dann doch mehr als zufrieden sein.

VRTX hat bereits eine Zulassung für Trikafta , einer Drei-Medikamenten-Kombination für CF-Patienten, bekommen. ( Prognosen: 4 Billionen Umsatz ) PTI könnte quasi ein Direkt-Konkurrent von VRTX bzgl. der Behandlung von CF Patienten werden.

Verkaufspreise und Anforderungskriterien sind so hoch, so dass viele Länder in Europa keinen Zugang zu dem Medikament Trikafta haben werden - deshalb suchen viele europäische Länder verzweifelt nach Lösungen für CF Patienten.
Und jetzt kommt der Aspekt von den Zwei-Medikamenten-Kombi-Behandlungen ins Spiel – diese sind leichter zu genehmigen, weil u.a. weniger Medikamentenkombinationen geprüft werden müssen.

In diesem Zusammenhang könnten die Worte von CEO Meenu Chhabra von Bedeutung sein :
„ should move forward with doublet commercialization as quickly as possible because it
is the best in class doublet so far"....."in Europe our doublet can do at least".

BOSTON, Dec. 11, 2019 /PRNewswire/ -- Proteostasis Therapeutics, Inc. (NASDAQ: PTI), a clinical stage biopharmaceutical company dedicated to the discovery and development of groundbreaking therapies to treat cystic fibrosis (CF), today announced positive, initial ex-vivo results of PTI's proprietary cystic fibrosis transmembrane conductance regulator (CFTR) modulators, PTI-801, PTI-808, and PTI-428, in individuals with CF who are ineligible for the current standard of care CFTR modulator therapies due to their genotype. The data are part of a pan-European strategic initiative, known as HIT-CF (Human Individualized Therapy of CF), which seeks to accelerate the development of, and access to, personalized therapies for CF patients, beginning with those for whom no currently approved CFTR modulator therapy is indicated.   

HIT-CF is sponsored by the European Commission Horizon 2020 program, in which CF-Europe, a patient organization representing more than 48,000 individuals with CF, collaborates and with the European Cystic Fibrosis Society Clinical Trial Network (ECFS-CTN), which is recruiting adult CF patients into the ex- vivo ­study through its 43 clinical trial centers.  HIT-CF collects tissue samples from CF patients and develops organoids, or miniaturized organs, that are genetically identical to the patient donor, and share the same micro-anatomy as the organ from which they were derived.  

As of today, rectal organoids from over 300 subjects have been collected for functional profiling and of those, 65 have been tested for response to PTI's investigational drugs. Early results support the initiation of enrollment of responding subjects into HIT-CF's clinical trial known as "CHOICES" (Crossover trial based on Human Organoid Individual response in CF - Efficacy Study), which is designed to evaluate the translation of organoid ex-vivo response to potential clinical benefit, such as changes in FEV1 and sweat chloride. CHOICES, which is expected to initiate in mid-2020, will be the first ever personalized medicine-based study in CF, with initial data expected by the end of 2020.  Fully funded by the HIT-CF, this trial is a placebo controlled, double blind, crossover study with an 8-week treatment period and 6 months of uninterrupted dosing.  The results may serve as the basis for a potential Marketing Authorization Application with the European Medicines Agency (EMA) in 2021 through a novel regulatory pathway which is being pursued jointly by Proteostasis and HIT-CF.  The CHOICES clinical study is part of PTI's broader clinical development strategy for its CFTR modulator candidates that is already separately funded for the common genotypes.

Results from the HIT-CF project to date will be presented at the Keystone Symposia on Tissue Organoids titled "Tissue Organoids as Models of Host Physiology and Pathophysiology of Disease (J1)" taking place on January 19-23, 2020 in Vancouver, BC, Canada.

Nein, das waren noch nicht die News zu den PH II Daten.

So wie ich als Biotech-Laie die Nachricht vom 11.12..19 verstehe, scheint PTI jetzt einen sehr großen Fuß in den Europäischen CF Markt gesetzt zu haben. Die CF Patienten , die für die derzeitigen CFTR - Modulator-Therapien aufgrund ihres Genotyp nicht geeignet sind, werden genauer fokussiert.

Erste positive Ergebnisse einer ex-vivo Studie - es wurden rektale Organoide von über 300 Probanden für die funktionelle Profilerstellung gesammelt und davon 65 auf ihre Wirkung auf PTIs Prüfpräparate ( 801-808-428 ) getestet - unterstützt die Einleitung der Einschreibung der antwortenden Probanden in die klinische Studie von HIT-CF . Diese zielt nun darauf ab, die Ergebnisse der ex-vivo-Reaktion der Organoide auf potenzielle klinische Vorteile, wie Veränderungen in FEV1 und Schweißchlorid, zu bewerten.

Die daraus geplante Crossover- Studie ( CHOICES ) , die voraussichtlich Mitte 2020 beginnen soll , wird dann die erste personalisierte, medizinisch basierte Studie mit CF sein. Das heißt, dass mit Entwicklung und Bereitstellung neuartiger CFTR-Modulatoren viele Patienten mit seltenen Mutationen zukünftig zu behandeln wären.

Erste Daten zum HIT-CF-Projekt werden bis Ende 2020 erwartet.

Erste Zwischenergebnisse des HIT-CF-Projekts werden auf den Keystone Symposien am 19. bis 23. Januar 2020 in Vancouver, BC, Kanada, vorgestellt.

Proteostasis stock tumbles on cystic fibrosis trial data

Shares of Proteostasis Therapeutics Inc. PTI, -46.44% fell 35% in premarket trading on Tuesday after the biopharmaceutical company shared mixed findings from a mid-stage study for its experimental triple combination cystic fibrosis therapy.

Proteostasis still plans to launch a Phase 3 trial next year. It is also conducting a personalized medicine study for patients with cystic fibrosis. The company held a call with investors on Tuesday morning to discuss the findings.

Die News von heute rechtfertigt m.M.n. nicht diesen extremen Rückgang. ( aktuell Minus 56 % !! ) Ich denke Hedgefonds beherrschen aktuell die Aktie und manipulieren den Aktienhandel bis zum "geht nicht mehr".

Vielleicht waren die Erwartungen bzgl. der Phase II Daten auch zu groß – ( dass die PTI Daten besser sind als VRTX Daten ) Auch wenn dies nicht der Fall ist, so sind die Daten aber zumindest so gut ( positive topline results) , dass PTI in die Phase III gehen wird.

Data was not as good as I was hoping but this is a panic sell. Clearly they need more trials. Data is confusing but shows the the drugs work and are safe and have potential in future trial - now Back to sleep, i will discuss more later

Der Bereich 0,75 - 0,85 € sieht für mich interessant aus.

Der Vergleich der PTI - Phase-2-TripleRx-Daten zu den Daten von VRTX hat diese Sell-off Panikreaktion ausgelöst und wurde/wird zugleich von den Hedgefonds ausgenutzt. Durch die positiven Aussagen auf der Konferenz wurden zu hohe Erwartungen auf die Ergebnisse der Phase II geweckt, die leider nicht erfüllt wurden.

Ich habe mir im Laufe des Tages die Trading-Spielchen der BIGs genaustens angeschaut und zugleich die Diskussionen in den USA Foren verfolgt. Es besteht aktuell sehr viel Unsicherheit bei den Biotechexperten bzgl. der Interpretation der Triple - Daten.

Da ich denke, dass PTI weiterhin zum Spielball der Hedgies wird und der Aktienkurs so manipuliert wird wie es den BIGs gefällt, habe ich mich heute dazu entschlossen, meinen gesamten Aktienbestand mit knapp 35 % Gewinn , zu verkaufen.

Ich bleibe jetzt zumindest erst mal an der Seitenlinie und verfolge die Newslage.

Ich wünsche allen Noch-Investierten auf jeden Fall „ Good Luck“

What happened
Shares of Proteostasis Therapeutics (NASDAQ:PTI) were down 56% at 12:07 p.m. EST on Tuesday after the company released what it called "positive" data from a phase 2 clinical trial testing its triple drug combination in patients with cystic fibrosis.

Investors rightfully didn't see the results as positive enough.

So what
Proteostasis' headline is technically correct: Patients responded to the triple combination of dirocaftor, posenacaftor, and nesolicaftor. After 28 days, cystic fibrosis patients with two copies of the F508del mutation treated with the triple combination improved their percent predicted forced expiratory volume in one second ppFEV1- a measure of lung function -- by 8 percentage points compared with placebo.

Unfortunately, the improvement isn't as good as the 13 to 14 percentage point improvement that Vertex Pharmaceuticals (NASDAQ:VRTX) saw with its recently approved triple combination called Trikafta.

The data for patients with one copy of the F508del mutation was murkier. Proteostasis noted that the triple combination improved sweat chloride -- a measurement of the protein that's mutated in cystic fibrosis patients -- but didn't improve ppFEV1 by a statistically significant margin versus placebo.

Now what
As biotech companies are apt to do in the face of lackluster results, Proteostasis is pressing on to a phase 3 program with two clinical trials expected to start next year . The More trial will test patients with two F508del mutations, while the Choices trial will test patients with rare mutations.

There could be an opportunity for Proteostasis to pick up Vertex's scraps if it can find a niche of patients who don't respond to Trikafta. Considering that Trikafta is expected to be a multibillion-dollar drug, the leftover patients could still be a substantial opportunity.

The biggest issue investors should be watching at this point is Proteostasis' cash, which stood at $77.8 million at the end of the third quarter. At this low market cap, raising additional funds would dilute current shareholders substantially.

Habe bisher nichts verkauft, habe es auch nicht vor.
Wegen den Feiertagen wird sich hier vermutlich nicht mehr viel tun. Anfang 2020 sollen neue Infos kommen, dann gibts neuen Schwung...

Nachdem sich die Kursmanipulationsphase doch relativ schnell beruhigt hat, habe ich seit Ende Dezember peu à peu wieder nachgelegt und bin jetzt wieder "voll" dabei .

Dann warten wir mal auf die ersten Zwischenergebnisse des HIT-CF-Projekts , die auf den Keystone Symposien am 19. bis 23. Januar 2020 in Vancouver, BC, Kanada, vorgestellt werden.

Ja bin ich....
PTI ist momentan der Einzige, der dem Monopolist Vertex gefährlich werden könnte.
Und genau darauf setze ich.
Cash reicht bis Ende 2021
Corona verzögert lediglich Studien, stoppen sie aber nicht.
Und der Markt an CV Patienten ist riesig.
Ich kaufe Monat für Monat und lass es liegen....
In einem Jahr sehen wir weiter....
Ich persönlich hoffe, dass der Kurs sogar noch etwas runter geht... so gibts mehr Aktien für mich...

Proteostasis Therapeutics (PTI) and Yumanity Therapeutics have completed their reverse merger.


The combined company will operate under the name Yumanity Therapeutics and its shares will commence trading on the Nasdaq Capital Market on December 23, 2020, under the ticker symbol "YMTX."


Following the completion of the merger, all of Yumanity's outstanding equity was converted into Proteostasis common stock and securities convertible into or exercisable for Proteostasis common stock. Further, the former PTI and Yumanity shareholders will own ~29.7%% and ~70.3% of the combined company, respectively.


The combined company will have nine directors including two Proteostasis directors, David Arkowitz and Kim Drapkin, and the existing Yumanity Directors; Dr. Tony Coles will serve as executive chairperson. Yumanity plans to explore strategic alternatives for the Proteostasis assets.

All unsere Hoffnungen von vor einem Jahr wurden erst einmal zerstört.
- kein Konkurrent zu Vertex
- kein Verkauf von Pti 428 ( Deal könnte zwar noch kommen, ich glaube aber nicht dran)
- ein Management, das keinen Wert auf Aktionäre legt

(.... Ein ehemaliger Inhaber eines 800 Mann Unternehmens sagte mal, nachdem man mit der Ausgabe von Aktien 70 Mio eingesammelt hat: Aktionäre sind das letzte, sie wollen ein Essen bei der Versammlung und dann noch Dividenden) ich persönlich sage da nur: Thema verfehlt, ähnlich wie Meenu...

Ich persönlich kann nur hoffen, dass Ymtx es deutlich besser macht.
Sollte der Kurs mal wieder bei 60-80 stehen (3-4 Pti) bin ich raus.
