      Additional Data Document Increases in HIV Drug Resistance; New ViroLogic Viral Fitness Assay Has Potential Role in Guiding Future HIV Therapy

      CHICAGO--(BW HealthWire)--Feb. 8, 2001-- Fourteen new research presentations utilizing ViroLogic Inc.`s (NASDAQ:VLGC) PhenoSense(TM) HIV technology were presented here this week at the 8th Conference on Retroviruses and Opportunistic Infections.

      The studies identified ways in which patients, physicians, and payors can benefit from personalized drug therapy guided by phenotypic technology. Additional ViroLogic studies presented here also revealed that the challenge of managing and overcoming HIV drug resistance is becoming more common in physicians` offices, as transmission of drug resistant virus continues to increase.

      Among the presentations here that utilized ViroLogic`s PhenoSense HIV technology were the following: Clinical Cut-Offs "Determination of a Clinically Relevant Phenotypic Resistance `Cutoff` for Abacavir Using the PhenoSense Assay," E. R. Lanier, Abstract 254

      Researchers from GlaxoWellcome and ViroLogic identified clinically relevant cutoff values for abacavir, a commonly prescribed AIDS drug. Clinical cutoffs define the level of drug resistance beyond which patients are less likely to respond to treatment, and allow more accurate interpretation of phenotypic assays. According to the researchers, abacavir`s clinical cutoff is 4.5-fold. That is, when a patient`s virus becomes more than 4.5 times more resistant to abacavir than normal, that patient is less likely to respond to treatment with the drug.

      ViroLogic began incorporating clinical cutoffs for abacavir and other antiretroviral drugs in December. PhenoSense HIV is the only drug resistance test to employ clinically relevant drug cutoffs; genotypic resistance tests are unable to provide quantitative clinical cutoffs.

      Drug resistance persists despite structured treatment interruptions (STIs), according to data presented by Dr. Steven Deeks of the University of California, San Francisco. Dr. Deeks led a team of scientists in tracking 19 patients who discontinued all antiretroviral drug therapy for a limited period of time. Using the PhenoSense HIV drug resistance assay, they found that although resistant virus became undetectable during the treatment interruptions, it often re-emerged after treatment was restarted.

      "These findings highlight the need for accurate, quantitative drug resistance testing when considering any break in therapy," said Dr. Hellmann, a co-author of the study. "The persistence of resistance despite treatment interruptions indicates that drug resistance testing should be used by all patients considering STIs prior to stopping therapy and prior to restarting therapy, in order to identify potential obstacles to successful treatment." Transmission of Drug-Resistant Virus "Antiretroviral Drug Susceptibility and Response to Initial Therapy Among Recently HIV-Infected Subjects in North America," S.J. Little, Abstract 756

      Rates of transmission of drug resistant HIV are rapidly increasing, according to a study presented by Dr. Susan J. Little of the University of California, San Diego. Using the PhenoSense HIV assay, a research team led by Dr. Little examined a cohort of 408 newly infected patients in 8 cities across the United States and Canada in 1999 and 2000, and found that high-level resistance to non-nucleoside reverse transcriptase inhibitors and protease inhibitors increased dramatically over earlier surveys. Overall, 10% of subjects harbored virus highly resistant to at least one AIDS drug and 4% harbored multi-drug resistant virus.

      The researchers concluded that the study results strongly support the use of resistance testing in the initial selection and management of therapy in all newly infected patients, since there is no way to identify which patients might be infected with resistant virus without testing.       In a related study, Dr. Viviana Simon of the Aaron Diamond AIDS Research Center led a team of scientists in New York and Montreal that examined 61 newly infected patients. Using PhenoSense HIV and a genotypic assay, the researchers recorded an increasing prevalence of drug resistant HIV in these patients. Of note, two-thirds of individuals in the study with multi-drug resistance were infected in the later half of the study, suggesting that multi-drug virus transmission is increasing. Integrating Individualized Patient Data "A Novel Approach to Integrate Pharmacologic and Virologic Characteristics: An In Vivo Potency (IVP) Index for Antiretroviral Agents," C. V. Fletcher, Abstract 732

      Researchers have created a new, comprehensive and accurate predictor of treatment success by integrating individualized patient data from a variety of laboratory tests, including PhenoSense HIV. The combined measurement, dubbed "in vivo potency," or IVP, combines pharmacokinetic data (which measures the amount of drug actually present in a patient`s body) with results from the PhenoSense HIV drug resistance assay. The IVP may be used guide physicians in designing individualized dosing regimens for patients in the future. Measuring Viral Fitness "Protease Inhibitor Resistant HIV-1 Has Marked Decreased Fitness in Vivo," R. M. Grant, Abstract LB4

      For the first time, researchers have demonstrated the ability of a new clinical assay developed by ViroLogic to accurately measure viral fitness, which denotes the ability of HIV to replicate effectively within the body. This discovery could have important clinical implications for physicians, who may use viral fitness testing to personalize HIV therapy in the future.

      The new study appeared in a latebreaker abstract presented by Dr. Robert Grant of the Gladstone Institute at the University of California, San Francisco. Dr. Grant`s research team measured viral fitness in a cohort of 12 patients using a rapid assay based on ViroLogic`s PhenoSense technology, and compared the results to the actual replicative performance of each patient`s virus in vivo. They found that the results of the ViroLogic assay correlated very well with in vivo measurements.

      "The strong correlation demonstrated in this study points toward a possible future clinical role for this fitness assay," said Dr. Grant. "The data to date in this area suggests that measuring viral fitness may provide clinically relevant new information on a patient`s prognosis."

      exel ist durchaus kaufenswert. die firma ist enorm vielversprechend. obwohl sie im bereich der grünen biotechnologie arbeitet, besteht hier jedoch sehr viel potential, aus pflanzlichen produkten effiziente wirkstoffe herzustellen. ich beobachte die aktie schon länger, ein kursziel liegt ca. bei 30€. ausserdem wurde exel ende 2000 von börseonline als eine firma beschrieben, die sich langfristig oben festsetzen kann. (neben riesigen firmen wie millennium und protein design labs!!)

      oooops, falsch gepostet. nun zu vlgc:
      ebenso kaufenswert. allerdings sehr riskant, da sie sich sehr stark auf ihre photo-sens therapie bei hiv konzentrieren. allerdings befindet sich die aktie im bbbiotech-portfolio, mit verhältnismassig hoher beteiligung! investment wird sich lohnen.

      ViroLogic ist ein Biotech-Unternehmen, das Produkte für die verbesserte Behandlung
      von Viruserkrankungen entwickelt. Mit einer Technologie, die unter dem Namen
      PhenoSense firmiert, möchte ViroLogic eine wichtige Rolle bei der Behandlung von
      AIDS und Hepatitis B und C spielen.

      PhenoSense-HIV – schnelles und präzises Testverfahren zu reduzierten
      Kosten

      PhenoSense-HIV ist ein so genannter Resistenztest, der die Widerstandsfähigkeit
      und Verträglichkeit der HIV-Infektion des Patienten gegen Antivirus-Medikamente
      misst. Mithilfe dieses Tests sollen die behandelnden Ärzte mit Informationen
      versorgt werden, um bessere therapeutische Entscheidungen treffen und
      Therapieversagen auf Grund von Resistenzen reduzieren zu können.

      Der Grundgedanke besteht darin, dass jeder Patient individuell die Fähigkeit hat,
      bestimmte Arten von HIV zu bekämpfen. Um nun eine effektive Behandlung
      entwickeln zu können, müssen die Ärzte wissen, an welcher HIV-Art der Patient
      erkrankt ist und wie diese Art auf die eingesetzten verschiedenen
      Medikamentenkombinationen reagiert.

      Früher – vor PhenoSense-HIV – war die Festlegung einer wirksamen, individuellen
      Behandlung wegen der sich verändernden Natur dieser Krankheit schwierig. Doch
      der neue Resistenztest hat gegenüber herkömmlichen Methoden viele Vorteile, die
      sich verändernde Widerstandsfähigkeit bei den HIV-Patienten zu testen. So
      benötigt PhenoSense-HIV nur eine Bearbeitungszeit von 14 Tagen, während die
      Standardtests 3 bis 4 Wochen brauchen. Außerdem liefert PhenoSense sehr präzise
      (95%), reproduzierbare und individualisierte Ergebnisse in Bezug auf die
      Medikamentenverträglichkeit.

      Keine langen und teuren FDA-Zulassungsverfahren

      Auf detailliertere Erläuterungen möchte ich verzichten. Aber Sie sollten wissen, dass
      diese Art labortechnischer Analyse keine Zustimmung durch die FDA benötigt,
      sondern durch die Health Care Finance Administration (HCFA) kontrolliert wird. Damit
      bleiben ViroLogic lange und oft ungewisse FDA-Zulassungsverfahren erspart.

      Der so genannte PhenoSense-HIV Select Panel kostet 775 US$, der etwas
      umfassendere PhenoSense-HIV Comprehensive Panel 895 US$. Die Kosten sind
      höher als für die herkömmlichen Tests, aber PhenoSense-HIV bietet präzisere und
      schnellere Ergebnisse und reduziert somit die Kosten beträchtlich.

      Nutzen Sie den günstigen Einstiegspunkt als Reaktion auf den Ablauf der Haltefrist
      bei Altaktionären. ViroLogic geht davon aus, die gesamten Verluste von 10,4 US$
      pro Aktie über den Zeitraum der letzten 4 Quartale im Jahr 2001 auf 0,63 US$ pro
      Aktie begrenzen zu können. Für das
      3. Quartal werden Verluste von 0,31 US$ pro Aktie erwartet.

      Seit Mitte September befindet sich die Aktie auf einer Kurstalfahrt von über 22 US$
      auf unter 10 US$. Hintergrund ist die am 29. Oktober 2000 erloschene Haltefrist, bis
      zu der es Altaktionären und bestimmten Investorengruppen nicht erlaubt war,
      Aktien des Unternehmens zu verkaufen. Somit stehen 10 Millionen neuer Aktien für
      den Verkauf zur Verfügung. Schon im Vorfeld des Endes der Lock-Up Frist hatten
      sich freie Aktionäre von ViroLogic Aktien getrennt.

      Ich empfehle Ihnen, auf jeden Fall unterhalb von 7 US$ zu kaufen. Die Bereitschaft
      der Altaktionäre (Firmenangehörige und Risikokapitalgeber, die vor dem Börsengang
      investiert waren) ist groß, einige oder alle ihrer Aktien zu verkaufen. Dies sollte den
      Kurs noch einmal drücken.
