New York (aktiencheck.de AG) - David Moskowitz, Analyst von Caris & Company, bewertet die Aktie von Onyx Pharmaceuticals (ISIN US6833991093/ WKN 900778) nach wie vor mit "above average".

Das Unternehmen habe heute Morgen beim Treffen der "European Society for Medical Oncology" (ESMO) die vollständigen Ergebnisse aus der Studienphase IIb zu "Nexavar" veröffentlicht. Vor allem die Daten zur Effizienz seien positiv zu bewerten, allerdings sei bei den mit "Nexavar" behandelten Patienten ein höheres Auftreten von Nebenwirkungen (Hand-Fuß-Syndrom) zu beobachten gewesen. Bei Caris & Company sei man der Auffassung, dass die vorgelegten Daten ausreichen sollten, um eine dritte Studienphase in die Wege leiten zu können.


Da das Unternehmen in Bezug auf seine Medikamenten-Pipeline sehr aktiv sei, könne im weiteren Jahresverlauf mit einem kontinuierlichen Newsflow gerechnet werden. Neben zusätzlichen "Nexavar"-Studien für weitere Indikationen, hierunter fortgeschrittener Leberkrebs, seien auch noch vor Jahresende die Daten aus der ersten Studienphase zum Präparat "OMX-0803" zu erwarten. Nach Berechnung von Caris & Company werde Onyx Pharmaceuticals im gesamten Geschäftsjahr ein EPS von 1,15 USD erzielen. Für 2010 erwarte man einen EPS-Anstieg auf 1,19 USD. Das Kursziel der Aktie belasse man bei 37,00 USD.

Nexavar in Combination with Chemotherapy Demonstrates 74 Percent Improvement in Progression-Free Survival in Patients with Advanced Breast Cancer in Phase 2 Study
Berlin, Germany. — Sep. 23, 2009

Bayer HealthCare Pharmaceuticals and Onyx Pharmaceuticals, Inc. (Nasdaq: ONXX) today announced the full results from their first collaborative group-sponsored randomized, double-blind, placebo-controlled Phase 2 trial showing that Nexavar® (sorafenib) tablets in combination with the oral chemotherapeutic agent, capecitabine, significantly extended progression-free survival in patients with advanced breast cancer. The data were presented at the joint 15th European CanCer Organisation (ECCO) and 34th European Society for Medical Oncology (ESMO) Multidisciplinary Congress in Berlin.

Jose Baselga, M.D., chairman and professor of medicine at Vall d'Hebron Institute of Oncology in Barcelona, scientific chairman of SOLTI and the principal investigator of this study, reported that patients receiving Nexavar plus capecitabine had a 74 percent improvement in the time they lived without their disease progressing compared to those who received the chemotherapy alone. The difference in median progression-free survival of Nexavar plus capecitabine versus capecitabine plus placebo was statistically significant, 6.4 months vs. 4.1 months (HR=.576, p=0.0006).

“Onyx and Bayer have built a strong foundation with Nexavar in treating unresectable liver cancer and advanced kidney cancer- - both disease areas with a previously unmet treatment need,” said Todd Yancey, M.D., vice president of clinical development at Onyx. “These new results signify another step in understanding the potential role of Nexavar in breast cancer.”

The study evaluated Nexavar in combination with the oral chemotherapeutic, capecitabine, in patients with locally advanced or metastatic HER-2 negative breast cancer. Overall, treatment with Nexavar plus capecitabine was tolerable and resulted in no new side effects. Common grade 3 or 4 treatment-related adverse events included hand-foot skin reaction, diarrhea, dyspnea, neutropenia and mucositis.

Breast Cancer Trial Design
The randomized, double-blind, placebo-controlled Phase 2 study evaluated Nexavar in combination with the oral chemotherapeutic agent, capecitabine, in 229 patients. These patients had locally advanced or metastatic HER-2 negative breast cancer and had received no more than one prior chemotherapy in this setting. The primary endpoint of the study was progression-free survival. Secondary endpoints included overall survival, time to progression, and safety. Patients were randomized to receive 400 mg of oral Nexavar or matching placebo twice daily, in addition to 1000 mg/m2 of capecitabine twice daily for 14 days followed by a seven day rest from capecitabine.

“These data represent a potentially significant advance in the treatment of breast cancer, which is the second leading cause of cancer-related death in women,” said Dimitris Voliotis, vice president, Global Clinical Development Oncology, Bayer HealthCare. “In addition to the positive signal generated in this trial, Onyx and Bayer are committed to the development of Nexavar in breast cancer in a variety of settings through a robust clinical program.”

About the Nexavar Clinical Program in Breast Cancer
In a clinical development program known as Trials to Investigate the Effects of Sorafenib in Breast Cancer (TIES), Nexavar is being evaluated in a variety of treatment settings for patients with breast cancer in collaboration with investigators and cooperative groups. Among these clinical trials are three ongoing randomized Phase 2 studies, including a trial to evaluate Nexavar plus paclitaxel in the first-line setting, a trial to evaluate Nexavar plus gemcitabine or capecitabine in the second-line setting following progression on bevacizumab, and a trial to evaluate Nexavar plus docetaxel and/or letrozole in the first-line setting.

About Breast Cancer
Breast cancer was the most commonly diagnosed cancer among women worldwide in 2007-2008 (approximately 1.3 million cases), and the second leading cause of cancer-related death among women (approximately 465,000 deaths). It is the most commonly diagnosed cancer among women in the United States (1 in 4 cancer diagnoses is breast cancer). Nexavar's Differentiated Mechanism
Nexavar, an oral anti-cancer therapy, is currently approved in more than 80 countries for liver cancer and in more than 90 countries for the treatment of patients with advanced kidney cancer. Nexavar targets both the tumor cell and tumor vasculature. In preclinical studies, Nexavar has been shown to target members of two classes of kinases known to be involved in both cell proliferation (growth) and angiogenesis (blood supply) – two important processes that enable cancer growth. These kinases included Raf kinase, VEGFR-1, VEGFR-2, VEGFR-3, PDGFR B, KIT, FLT-3 and RET.

Nexavar is also being evaluated by the companies, international study groups, government agencies and individual investigators as a single agent or combination treatment in a wide range of cancers, including lung, ovarian and colorectal cancer and as an adjuvant therapy for liver and kidney cancer.

Important Safety Considerations For Patients Taking Nexavar
Based on the currently approved U.S. package insert for the treatment of patients with unresectable hepatocellular carcinoma and advanced kidney cancer, hypertension may occur early in the course of therapy and blood pressure should be monitored weekly during the first six weeks of therapy and treated as needed. In HCC patients, bleeding with a fatal outcome from any site was reported in 2.4% for Nexavar and 4% in placebo. The incidence of treatment-emergent cardiac ischemia/infarction was 2.7% for Nexavar vs. 1.3% for placebo. In RCC patients, incidence of bleeding regardless of causality was 15% for Nexavar vs. 8% for placebo and the incidence of treatment-emergent cardiac ischemia/infarction was 2.9% for Nexavar vs. 0.4% for placebo. Most common adverse events ?20% related to Nexavar for both HCC and RCC were fatigue, weight loss, rash/desquamation, hand-foot skin reaction, alopecia, diarrhea, nausea, and abdominal pain. Grade 3/4 adverse events in HCC and RCC patients, respectively, were 45% for Nexavar vs. 32% for placebo and 38% for Nexavar and 28% for placebo. Women of child-bearing potential should be advised to avoid becoming pregnant and advised against breast-feeding. In cases of any severe or persistent side effects, temporary treatment interruption, dose modification or permanent discontinuation should be considered.

ich bin vor kurzem bei 21,90$ eingestiegen,ich denke mal dass sie noch bis auf 21-21.50$ zurückkommen kann.
ich behalte sie im auge und werde gegebenenfalls noch mal nachkaufen.

Onyx Pharmaceuticals Announces Positive Top-Line Carfilzomib Data From Phase 2b Study
24 Percent Overall Response Rate and Duration of Response Exceeding Seven Months in Heavily Pretreated Advanced Multiple Myeloma Patients

EMERYVILLE, Calif., July 26 /PRNewswire-FirstCall/ -- Onyx Pharmaceuticals, Inc. (Nasdaq: ONXX - News) today announced positive top-line results from the Phase 2b 003-A1 study of single-agent carfilzomib, a selective, next generation proteasome inhibitor, in patients with relapsed and refractory multiple myeloma. In an independent review of the data, carfilzomib achieved an overall response rate (partial response or greater) of 24 percent and a median duration of response of 7.4 months in patients who entered the study after receiving a median of five prior lines of therapy (corresponding to a median of 13 anti-myeloma agents) and whose disease was refractory to their last therapeutic regimen. The clinical benefit rate (minimal response or greater) in the study population was 36 percent. Carfilzomib was well-tolerated and there were no new or unexpected toxicities observed. Full results of the trial will be presented at an upcoming scientific meeting. Based on these results, Onyx is continuing discussions with the U.S. Food and Drug Administration (FDA) regarding next steps in filing a new drug application (NDA) for carfilzomib, which the company expects to submit by year-end 2010 for potential accelerated approval in the U.S.

"Despite recent advances in treating multiple myeloma, all patients eventually relapse. The unmet medical need remains great, as the outlook for patients with relapsed and refractory disease is grim," said Michael G. Kauffman, M.D., Ph.D., Chief Medical Officer of Onyx Pharmaceuticals. Trial Design

The 003-A1 study was an open-label, single-arm Phase 2b trial. The trial evaluated 266 heavily-pretreated patients with relapsed and refractory multiple myeloma whose disease was refractory to their last treatment regimen and who had received at least two prior therapies, including bortezomib, either thalidomide or lenalidomide, an alkylating agent, glucocorticoids and an anthracycline. Refractory disease was defined as < 25% response or progression during therapy or within 60 days after completion of therapy.(ii) Patients enrolled in the 003-A1 trial had received a median of five prior therapeutic regimens, corresponding to a median of 13 anti-myeloma agents. Patients received carfilzomib at 20mg/m2 for the first cycle followed by 27mg/m2 thereafter for up to 12 cycles. Patients who completed the 12 cycles were eligible to enter an extension study. Responses and progression were determined according to the International Myeloma Working Group (IMWG) criteria. The trial was conducted in collaboration with the Multiple Myeloma Research Consortium (MMRC) and at additional sites in the U.S. and Canada.

"The patients in the 003-A1 trial represent an advanced population with significant unmet medical need who had received many lines of therapy and had limited options available to them outside of a clinical trial, strongly underscoring the need for new treatments. We are proud to have worked with the Onyx team that is developing carfilzomib and are encouraged by these results," said Kathy Giusti, Founder and CEO of the MMRC and a multiple myeloma patient. The MMRC initiated a relationship with the company (then Proteolix) in 2006, which included the participation of 11 MMRC Member Institutions in the 003-A1 trial, representing 36 percent of the total trial centers and 60 percent of enrolled patients.

Der Pharma- und Chemiekonzern Bayer erwartet in den kommenden Jahren durch den Verlust von Patentrechten keine großen Einbrüche im Arzneimittelgeschäft. "Wir haben keine wesentlichen Patentabläufe bis 2014. Das ist ein großer Unterschied im Vergleich zu anderen", sagte der Chef der Bayer-Pharmasparte, Andreas Fibig, am Dienstag im Gespräch mit Reuters. Dazu komme, dass die Pipeline an Wirkstoffen in der Entwicklung bei Bayer gut gefüllt sei. So habe Bayer etwa mit dem Wirkstoff VEGF Trap-Eye ein neues Medikament gegen Augenkrankheiten sowie einen Nachfolgewirkstoff zu dem lukrativen Krebsmittel Nexavar in der Hinterhand. Die Sparte HealthCare, zu der auch das Pharma-Geschäft gehört, will der Leverkusener Konzern auch durch den Kauf anderer Firmen ausbauen. "HealthCare ist bei der Bayer ein Wachstumsbereich. Wir würden durchaus auch in Zukäufe investieren", sagte der Manager am Rande der "Handelsblatt-Pharmatagung" in Frankfurt. Dazu gehört Fibig zufolge auch das Geschäft mit Tiermedizin-Präparaten. In dem Bereich ist Bayer früheren Informationen zufolge im Bieterprozess um Geschäfte der Pharmariesen Sanofi-Aventis und Merck & Co. Daneben will Bayer im Pharmageschäft seine Position in Indien weiter ausbauen. Eine Voraussetzung ist nach Angabe der Leverkusener aber, dass die intellektuellen Eigentumsrechte geschützt bleiben und geschützt werden. Auch Übernahmen sind für Bayer vorstellbar - "wenn es vernünftige Preis-Multiples gibt", betonte der Bayer-Manager.
