ist ADX71149 im Gegensatz zu bisherigen antipsychotischen
Mitteln in der Lage, die Symptome der Schizophrenie
(übernormale Wahrnehmungen, Halluzinationen, emotionale
Defizite, kognitive Defizite) zu dämpfen, und zwar ohne Compliance-
limitierende Nebenwirkungen, wie Sedation, extrapyramidale
Symptome, Hormonstörungen und Gewichtszunahme
Diese Produkteigenschaft soll auch bei Angstzuständen, die im
Zusammenhang mit Depressionen stehen, helfen. Die Analysten
von Wedbush (10.09.12) gehen auf Grund der neuartigen
Funktionsweise des Medikaments von Produktumsätzen von
über 2 Mrd. USD für das Produkt aus.

Dabei habe das Medikament bei niktotinabhängigen Mäusen die somatischen Erscheinungen des Entzugs gemildert und teilweise die in diesem Zusammenhang gesteigerte Schmerzempfindlichkeit herabgesetzt. Addex fühlt sich durch die Ergebnisse in dem Ansatz bestätigt, die Aktivierung des GABA-B-Rezeptors als Strategie zur Raucherentwöhnung zu verfolgen, teilte das Unternehmen am Donnerstag mit.

Addex Therapeutics (SIX: ADXN), a leading company pioneering allosteric modulation-based drug discovery and development announced today entering a collaboration with the National Institute on Alcohol Abuse and Alcoholism (NIAAA), a component of the National Institutes of Health (NIH), to evaluate the pharmacology of ADX71441, a GABAB receptor positive allosteric modulator (PAM), in preclinical models of alcohol use disorder. The collaboration will evaluate Addex drug candidate, ADX71441, in a battery of preclinical models to study its potential as a treatment for alcohol use disorder.

Both clinical and pre-clinical data suggest that activation of GABAB receptors offers a unique therapeutic opportunity to address the needs of patients with alcohol use disorder by suppressing multiple alcohol-related behaviors, including intake, reward and motivation, and seeking behavior. GABAB receptor PAMs have a similar pharmacological profile to the orthosteric agonist, baclofen, while showing a more favorable tolerability profile.

Addex has published positive data with ADX71441 in two models of alcohol abuse in mice, the ethanol binge-like drinking, drinking-in-the-dark (DID) and a model of long-term, excessive drinking, intermittent access to alcohol (IAA) (Hwa et al. Psychopharmacology 2014;231(2):333-343).

"The evaluation of ADX71441 in additional preclinical models established in the laboratory of clinical and translational studies at NIAAA will provide Addex with invaluable information to guide the clinical development of ADX71441 in alcohol use disorders," said Sonia Poli, CSO at Addex. About Alcohol Use Disorder
Problem drinking that becomes severe is given the medical diagnosis of "alcohol use disorder" or AUD. AUD is a broad term for problems with alcohol, and is generally indicative of compulsive and uncontrolled consumption of alcoholic beverages. It is medically considered a disease, specifically an addictive illness. The World Health Organization estimates that about 140 million people throughout the world suffer from alcohol dependence. Patients with alcohol use disorder suffer major changes to the brain structure and chemistry. Excessive alcohol consumption damages almost every organ in the body and the cumulative toxic effects can cause both medical (cirrhosis of the liver, pancreatitis, heart disease, peptic ulcers, sexual dysfunction) and psychiatric (epilepsy, dementia, psychosis, anxiety & depression) problems. Treatment of alcoholism is complex with a current standard of care typically being prescribed to patients with heavy drinking but largely being unable to prevent them from relapsing. Approximately 7.2 percent or 17 million adults in the United States age 18 and older had an AUD in 2012. This includes 11.2 million men and 5.7 million women. In addition, an estimated 855,000 adolescents age 12-17, in the United States, had an AUD in 2012.

About GABAB Activation and ADX71441
Activation of gamma-aminobutyric acid subtype B (GABAB) receptor, a Family C class of GPCR, is clinically & commercially validated. Generic GABAB receptor agonist, baclofen, is marketed for spasticity and some spinal cord injuries, used off label for overactive bladder (OAB) and alcohol use disorder, but use is limited due to variety of side effects of the drug and rapid clearance. ADX71441 is a potent selective positive allosteric modulator (PAM) which potentiates GABA responses at the GABAB receptor. ADX71441 is a novel, first-in-class, oral, small molecule that has demonstrated excellent preclinical efficacy and tolerability in several rodent models of pain, anxiety, addiction and OAB and has also proven efficacy in a genetic model of Charcot-Marie-Tooth Type 1A disease (CMT1A). ADX71441 differs from the generic drug baclofen in that it is a positive allosteric modulator rather than an orthosteric agonist at the GABAB receptor. ADX71441 only acts when the natural ligand (GABA) activates the receptor, and therefore respecting the physiological cycle of activation. It has been proposed that PAMs produce less adverse effects and lead to less tolerance than direct agonists (May and Christopoulos 2003; Langmead and Christopoulos 2006; Perdona et al. 2011; Urwyler 2011; Gjoni et al., 2008).

Addex and Dystonia Medical Research Foundation Announce Partnership to Explore the Therapeutic use of Dipraglurant in the Treatment of Dystonia


Geneva, Switzerland /Chicago, USA, 19 January 2015 – Addex Therapeutics (SIX: ADXN), a leading company pioneering allosteric modulation-based drug discovery and development and the Dystonia Medical Research Foundation (DMRF) announced today entering a collaboration to explore the use of dipraglurant to treat dystonia, the third most common movement disorder following essential tremor and Parkinson’s disease. Dipraglurant, a novel small molecule inhibitor of the metabotropic glutamate receptor 5, has shown promise in the treatment of levodopa-induced dyskinesia and dystonia in Parkinson’s disease. Dipraglurant has also been shown to normalize the effects of the TOR1A/DYT1 dystonia mutation in the brains of mice. The objective of the collaboration is to design a detailed development plan and regulatory path as well as identifying key option leaders and patients for a Phase 2 clinical trial. In addition, Addex recently reported on 9 January, plans to start clinical testing of the therapeutic effect of dipraglurant in patients with cervical dystonia in collaboration with Professor Dirk Dressler of The Hannover Medical School.

Dystonia is a neurological disorder characterized by persistent or intermittent muscle contractions causing abnormal, often repetitive, movements, postures, or both. The movements are usually patterned and twisting, and may resemble a tremor. Symptoms originate from an imbalance of neurotransmitters in the brain. There are multiple forms of dystonia, and up to 100 diseases and conditions include dystonia as a prominent symptom. Dystonia may affect a single body area or be generalized throughout multiple muscle groups. Dystonia affects men, women, and children of all ages and backgrounds. In a double-blind, placebo-controlled, US and European Phase 2 study in PD-LID, data showed that dipraglurant met the primary objective of the study by exhibiting a good safety and tolerability profile. Dipraglurant also demonstrated a statistically significant reduction in LID severity with both 50 and 100 mg doses. Dipraglurant reduced dystonia severity in addition to chorea, the two major LID components. Efficacy was measured using the modified Abnormal Involuntary Movement Scale (mAIMS), patient diaries documenting "off-time" (impaired voluntary movement), "on-time" (with or without dyskinesia) and sleep. Additional endpoints include the Unified Parkinson's Disease Rating Scale (UPDRS), the Clinician & Patient Global Impression of Change (CGIC & PGIC), and an evaluation of the patients' mood using the Hospital Anxiety & Depression Score. The trial was supported by a grant from The Michael J. Fox Foundation for Parkinson's Research.

Das Westschweizer Pharmaunternehmen Addex Therapeutics hat im Rahmen einer Privatplatzierung Kapital im Umfang von 2,8 Mio CHF aufgenommen und sich damit die Finanzierung bis ins Jahr 2017 hinein gesichert.

Die Ausgabe neuer Aktien sei von der Investmentgesellschaft Herculis Partners durchgeführt worden, wobei sich Herculis-Kunden und das Addex-Management an der Transaktion beteiligt hätten, teilt Addex am Montag mit. Nach Abschluss der Platzierung halten die von Herculis betreuten Fonds 4,1% an Addex.
Von den neuen Aktien wurden 921'667 zu einem Preis von 3 CHF je Titel bei Investoren und weitere 604'369 zu 1 CHF bei der Tochter Addex Pharma SA platziert. Somit seien ab dem (heutigen) Montag insgesamt mehr als 1,5 Mio, die einen Anteil von 13% am gesamten Kapital repräsentieren, neue Addex-Aktien kotiert. Die Aktien wurden aus dem genehmigten Kapital geschaffen.
"Mit den zusätzlichen Mitteln können wir unsere Finanzierung bis ins Jahr 2017 ausweiten", wird Addex-CEO Tim Dyer in der Mitteilung zitiert. Damit sei man in der Lage, das Produktportfolio in Zusammenarbeit mit den Partnern weiterzuentwickeln. 10.03.2015 - Der Westschweizer Wirkstoffentwickler Addex Therapeutics AG hat im Rahmen einer Privatplazierung rund 2,8 Mio. CHF aufgenommen. Damit sei die Finanzierung bis in das Jahr 2017 hinein gesichert, heißt es.

Die neuen Aktien wurden von Kunden der Investmentgesellschaft Herculis Partners sowie dem Addex-Managment gezeichnet. Nach Abschluss der Plazierung halten die von Herculis betreuten Fonds 4,1% an dem Unternehmen in Plan-les-Ouates. Von den neuen Aktien wurden rund 920.000 zu einem Stückpreis von 3 CHF bei Investoren und weitere 600.000 zu 1 CHF beim Tochterunternehmen Addex Pharma SA plaziert.

„Mit den zusätzlichen Mitteln können wir unsere Finanzierung bis ins Jahr 2017 ausweiten“, sagte Addex-CEO Tim Dyer. Damit sei man in der Lage, das Produktportfolio in Zusammenarbeit mit den Partnern weiterzuentwickeln. Hauptentwicklungskandidat von Addex ist der niedermolekulare Wirkstoff Dipraglurant zur Behandlung von Levodopa-induzierten Dyskinesien bei Parkinson (PD-LID). Der negative allosterische Modulator des metabotropen Glutamatrezeptors 5 (mGluR5 NAM) wurde bereits in der Phase II getestet und wird in Zusammenarbeit mit der Michael J. Fox Foundation entwickelt.

In Zusammenarbeit mit der Universität Cork in Irland habe man an Laborratten die Wirksamkeit von ADX71743 bei stressinduzierten Eingeweideschmerzen aufzeigen können, teilte das Genfer Unternehmen am Mittwoch mit.
Der Versuch zeige, dass "negative allosterische Modulatoren (NAM) des metabotropen Glutamatrezeptors 7 (mGluR7)" eine neuartige und wirksame Ansatz in der pharmakologischen Behandlung von stressinduzierten Eingeweideschmerzen sein könnten, heisst es weiter. Das sei ein weiterer Schritt zur Validierung solcher Wirkstoffe in einem Gebiet, wo es ein Bedürfnis nach medizinischer Behandlung gebe.

Dipraglurant is an oral, small molecule allosteric modulator that inhibits selectively the metabotropic glutamate receptor 5 (mGluR5), a Class C G-Protein Coupled Receptor (GPCR), with potential to be used in combination with levodopa or dopamine agonists or as a standalone treatment for Parkinson's disease levodopa-induced dyskinesia (PD-LID), motor and non-motor symptoms of Parkinson's disease and other movement disorders. Der Verlust kam durch die Baby-Franzosen herein heute, weil die Franzosen offensichtlich keine Bilanzabzüge für Forschungskosten erlauben. Man habe erfolgreich den ersten Teil eines Projektes abgeschlossen, das von der Schweizer Kommission für Technologie und Innovation (CTI) finanziert wurde, heisst es in einer Mitteilung vom Montag. Dabei wurden die beiden allosterischen Modulatoren mGluR4 und mGluR7 in Modellen von neurodegenerativen und psychiatrischen Erkrankungen getestet.


So wurde der Wirkstoffkandidat ADX88178, ein selektives mGluR4 PAM, in einem vorklinischen Modell bei Parkinson getestet. Ausserdem wurde die elektrophysiologische Wirkung von ADX71743, einem negativ-allosterischen Modulator, erforscht.


Mit dem erfolgreichen Abschluss der beiden Studien, habe das Team einen zweiten Zuschuss des CTI über 660'240 CHF erhalten, um die Programme mit den beiden Wirkstoffen weiter voranzutreiben, heisst es weiter.

Addex erinnert mich einwenig an Santhera wie in 2014 nur leider ist Addex noch ca 4-5 Jahre entfernt vom Zulassungsantrag aber trotzdem ist die Aktie wegen der sehr niedrigen bewertung interessant . Auch interessant ist das der CMO von Acadia Pharma (ACAD) zu Addex gewechselt ist was hier zu bemängeln ist , ist der niedrige cashbestand .

Für ein Investment sind sie für mich persönlich noch nicht weit genug trotzdem ist die Aktie attraktiv mit einer Marktkap von unter 30M CHF . Vielleicht sollte Santhera hier zuschlagen und Addex übernehmen würde die Pipeline ganz schön aufhübschen .

Das Geld hat keinen Wert mehr und nachdem die Zentralbanken beschlossen haben, dass sie ihre Programme langsam zurückfahren werden, geht es auch bei guten Nachrichten einfach nicht mehr höher. Die Finanzkrise ist einfach noch nicht ausgestanden.

Wer den Long-Chart vom Dow kennt der wird feststellen, dass ein solcher Zusammenbruch gut und gerne 30 Jahre Zeit braucht.

nach dieser meldung von heute habe ich mir etwas zeit genommen und addex etwas genauer angeschaut.

die pipeline ist noch in einem frühen stadium, es hat aber einige wirklich interessante und inovative wirkstoffe dabei. mit einem marktwert von ca. 30 mio. ist addex in meinen augen sicherlich nicht zu hoch bewertet. das interesse scheint im moment eher klein zu sein und somit scheinen momentan auch keine pusher dabei zu sein, was ich sehr begrüsse. so nach dem sprichwort: "totgeglaubte leben länger"

wer etwas risikokapital und einige jahre zeit hat scheint mir ein investment durchaus interessant zu sein. ich habe mir heute morgen jedenfalls eine kleine position um 2.20 gekauft.

einfach dass es gesagt ist: ich will auf keinen fall den kurs pushen und das risiko von einem totalverlust oder verwässerung durch neue kapitalerhöhungen ist natürlich nicht zu unterschätzen. wollte euch hier einfach mal meine überlegungen mitteilen.

Habe mir gestern ein paar Addex zugelegt im vorfeld des Nasdaq listings in diesem Quartal das bestimmt ordentlich Schwung reinbringen wird .Firma ist günstig bewertet und sehr gut finanziert deshalb denke ich stehen die Chancen sehr gut für steigende Kurse .

Schon jetzt hat Addex bereits starke Investoren wenn noch Nasdaq dazu kommt werden mit Sicherheit weitere Institutionelle einsteigen wollen und das wird kaum gehen ohne den Kurs zu pushen deshalb eben mein vorzeitiger Einstieg . Die Aktie wird wahrscheinlich irgendwo zwischen $6-10 (ADS) an die Nasdaq gebracht.

Die Ausgliederung ist ein genialer finanzieller Schachzug und ermöglicht bei guten Zahlen einem Interessenten die Möglichkeit Addex völlig problemlos zu übernehmen und zu integrieren. Keine Kosten, keine Mitarbeiter etc. die übernommen / abgefunden werden müssten. Ich könnte mir vorstellen das dieser Deal der erste Schritt dahin ist.
