      In an April 29 research report, analyst Jotin Marango reported that ROTH Capital Partners raised its price target on ArQule Inc. (ARQL:NASDAQ) to $10 per share in anticipation of a clinical update on its ARQ531 dose escalation study in chronic lymphocytic leukemia (CLL) patients, to be given at the European Hematology Association (EHA) Congress this June.

      "This spring, we are increasingly bullish on ArQule," and "remain high conviction buyers of ARQL shares," Marango wrote. The stock is currently trading at around $6.10 per share.

      The study results ArQule will present in six weeks will likely be from the sizeable group of CLL patients treated at higher doses of ARQ531, a BTK inhibitor, Marango noted. This would include cohort 6, whose dose was increased to 65 milligrams (65 mg) from 45 mg, and the expanded cohort 7, which received 65 mg. (Data from cohort 8, which is receiving 75 mg, likely will not be ready in time for the EHA meeting.)

      Marango highlighted that a positive update could elicit a favorable market reaction, particularly if patients experienced tumor shrinkage. The new data could move ArQule closer to determining the recommended dose and positioning of a Phase 2 study. Also, a positive report could "rekindle mergers and acquisitions speculation around ARQ531," he noted.

      Were the latter to happen, Marango purported, the interest would likely come from a handful of larger companies: AbbVie Inc. (ABBV:NYSE), Celgene Corp. (CELG:NASDAQ), Bayer AG (BAYRY:OTCMKTS; BAYN:XETRA), Daiichi Sankyo Co. (4568:TYO; DSKYF:OTCPK) and Jazz Pharmaceuticals Plc (JAZZ:NASDAQ). "Overall, we note that there certainly appear to be more potential apt buyers than there are reversible BTK inhibitors in development," he added.

      ROTH has a Buy rating on ArQule.

      Roth spricht eher gegen ein Investment in ARQL, Unseriöse Bude, gucke bei Cytori - seit Jahren helfen die mit, das die Firma weiterlebt.

      ARQL macht einen guten Job, die Investoren auf die Daten der EHA vorzubereiten. Erwartungsmanagement könnte man das nennen.
      Aus der Präsentation heute Nacht:
      - EHA Daten werden vor allem aus Kohorte 7 bestehen (mit 1-2 Scans) und Daten von 8 der 10 Teilnehmer beinhalten.
      -Davon 6 CLL Patienten mit C481 Mutation, 1 DLBCL und 1 Richter Transformation.
      -Von Kohorte 8 wird es noch keine Daten zur Effektivität geben.

      Die wichtigste Frage wird vor allem sein, wie hoch die Ansprechrate sein wird. 1 PR ist ja schon bekannt. Bei 1 weiteren PR wäre man schon bei 30% und bei 2 weiteren bei 50% Ansprechrate. (s. Analystenkommentare)
      Auch der 1 Patient mit Richter Transformation dürfte viel Beachtung bekommen. Bin wirklich sehr gespannt!

Ich würde es aber nach wie vor begrüßen, wenn man das Potenzial von 531 für andere Indikationen erstmal testet. So wie man es ja auch plant.

      Das perfekte Beispiel einer gelungenen KE. Macht Sinn und konnte zu einem sehr guten Kurs durchgeführt werden. Im Rahmen der KE wurden noch ein kleiner Ausblick auf die 8. Kohorte gegeben.


      Also auch eine PR in DLBCL. Die Dosis scheint auch gefunden zu sein. Sehr schön! Hier verkaufe ich momentan gar nichts!

      Interessant dürfte sein, wie es bei den hochdosierten Patienten (gelb markiert) weitergeht. Ich könnte mir durchaus vorstellen, dass man da schon vor ASH Neues präsentieren wird. Zumal man ja die Dosis gefunden zu haben scheint, mit der man jetzt weitergehen will. Man will ja schon dieses Jahr mit der expansionstudy beginnen.
      Die PR wurde auch bei einem 2. Patienten mit einem 2. Scan bestätigt (und hat sich sogar verbessert). Jetzt also 2 confirmed PRs (Patienten 27 und 30).

      KE war 4fach überzeichnet. Cash soll jetzt bis 2022 reichen.

      Am Mittwoch kommen Zahlen von ARQL. Zumindest denkbar ist ein Update zur 8. Kohorte bzw. 531. Oder zumindest nähere Aussagen dazu, wann mit einem Update zu rechnen ist. ASH fände ich unnötig spät, zumal man ja dieses Jahr noch mit der expansionstudy starten will.

      Bitter! ARQL heute Tagesverlierer mit -13%! Grund ist die Ankündigung von Lilly, dass man Ende des Jahres P1 Ergebnisse des BTK (LOXO305) veröffentlichen wird. Das ist der Kandidat aus der LOXO Übernahme. Ich denke es ist gar nicht so sehr die Ankündigung an sich (das ist ja keine Überraschung), sondern das Timing dies im Zuge phantastischer Daten des RET Kandidaten bekannt zu geben. Das macht erstmal Angst! Aber man muss sicher auch sehen, dass der Kandidat nicht aus der ARRY Kollaboration stammt sondern von Redx entwickelt und verkauft wurde. Man wird sehen...

      Ich bin schon vor einigen Wochen bei 7,2 Euro ausgestiegen. Habe einiges an Gewinn liegen lassen, aber ich war auch viel zu hoch in ARQL investiert. Bin bei 3,5 Euro eingestiegen und was man hat, das hat man. Bin nach wie vor von Arqule und deren Pipeline überzeugt. Bin zurzeit in CLVS investiert, vielleicht magst du Dir die Aktie mal anschauen. Ist nichts für schwache Nerven, aber das ist bei Biotechwerten ja immer der Fall.
      Ich denke ARQL könnte das Gap bei $6 noch schließen. Sollte das der Fall sein, werde ich wohl wieder einsteigen.

      ich bin für ARQL weiter sehr bullish. Die größte Gefahr sind sicherlich bessere Ergebnisse eines Konkurrenzprodukts. Den Kurseinbruch der letzten Tage halte ich aber für übertrieben. Bin gespannt auf die Ergebnisse auf der ASH. Zudem hat ARQL ja noch weitere Eisen im Feuer.
      Zu CLVS: Da geht es m.E. nur noch um Pleite oder Übernahme. Alleine können die langfristig nicht überleben. Die Konkurrenz ist einfach zu groß. (AZN und Merck sowie GSK) Der Kurs ist aber sicherlich sehr niedrig dafür, dass sie ein zugelassenes Medikament haben.

      Gerüchte hinsichtlich einer Übernahme von CLVS gibt es immer wieder. Aktuell ist Marktkapitalisierung von CLVS niedrig, vielleicht wird ein Großer Player demnächst zugreifen.
      Eine kurzfristige Pleite halte ich für ausgeschlossen. Nach der jüngsten Finanzierungsmaßnahme schätze ich die liquiden Mittel auf 550M. Aufgrund der aggressiven Entwicklungs- und Vermarktungsstrategie ist der cash burn sehr hoch. Dieser wird sich in den nächsten Jahren auf ca. 150M p.a. reduzieren. Die Firma ist zumindest für die nächsten 2 Jahre finanziert. Der Kursverfall war brutal, von $80 auf unter $5. Ein Grund für den Absturz war die Zulassung eines Wettbewerbsproduktes. Der Wettbewerb ist groß, allerdings ist das Marktpotential für PARP-Inhibitors riesig, so dass jeder ein großes Stück vom Kuchen bekommen kann. CLVS hat schon ein zugeladenes Produkt und wir mit diesem 2019 ca. 140M erzielen. Bin knapp über $5 eingestiegen und werde den Wert zeitnah verfolgen.

      Bei ARQL bereitet man die Investoren wieder sehr gut auf die anstehenden Daten - diesmal auf der ASH vor. Aus der letzten Konferenz:

      -Es werden auf der ASH die finalen Daten der P1 präsentiert.
      -Schwerpunkt ist die Dauerhaftigkeit des Ansprechens. Alle Patienten haben mindestens 2 oder sogar 3 Scans gehabt (erster Scan nach 2, dann nach 5 und 9 Monaten) Von daher wird es spannend, ob Patienten mit PR diese auch halten konnten bzw. ob sich über die Zeit sogar noch einige Verbesserungen im Vergleich zur EHA zeigen lassen. Hier nochmal die Daten zur EHA:



      Die P2 Expansion Cohort Studie läuft bereits. Neu ist hier die Kohorte mit Patienten, die nicht länger auf Ibrutinib ansprechen aber nicht die C481S Mutation haben. Das war vorher nicht geplant -macht aber Sinn.

      ArQule, Inc. (NASDAQ:ARQL) is a clinical stage pharmaceutical company pursuing multiple programs in biomarker-defined oncology and rare disease indications.

      ARQL stock saw some action in the last session, trading over 3 times of average volume at a 6% increase from the previous close. We will see why the stock is attracting market attention and if it is justified.



      Catalyst

      The immediate catalyst that triggered the market enthusiasm is the October 2, 2019, announcement of initiation of dosing in the phase 1/2 MOSAIC trial for ARQ 092 (miransertib) in overgrowth diseases. Topline data from this trial is expected in 1H-2021.

      However, the lead candidate presently at advanced stage is derazantinib (ARQ 087). Topline data from the ongoing phase 2 "Pivotal Study of Derazantinib in Patients With Inoperable or Advanced Intrahepatic Cholangiocarcinoma (iCCA) and FGFR2 Gene Fusions or FGFR2 Gene Mutations or Amplifications" is expected in 1H-2020.

      Pipeline

      Other product candidates and indications in the company's clinical pipeline include:

      ARQ 531 - a reversible dual inhibitor of both wild type and C481S-mutant Bruton's tyrosine kinase (BTK) that is in Phase 1 clinical development for B-cell malignancies refractory to other therapeutic options.

      ​Miransertib (ARQ 092) - a selective inhibitor of protein kinase B (AKT), a serine/​threonine kinase. A registrational clinical trial of miransertib for the treatment of Proteus syndrome (PS) and PIK3CA-Related Overgrowth Syndromes (PROS) initiated in October 2019. Miransertib is also in phase 1b clinical development in oncology in combination with anastrozole hormonal therapy.

      ​ARQ 751 - a selective inhibitor of AKT that is in phase 1 clinical development for solid tumors harboring AKT, phosphoinositide 3-kinase (PI3K) or phosphatase and tensin homolog (PTEN) mutations or that are PTEN null.

      About Derazantinib (ARQ 087)

      Derazantinib is an oral drug that inhibits FGFR and especially FGFR2. FGFR is a family of kinase, fibroblast growth factor receptors, which are involved in key cellular behaviors like proliferation, cell differentiation, cell migration, cell survival and angiogenesis. A number of cancers owe their origin to FGFR dysregulation; these include iCCA, cholangiocarcinoma, bladder, endometrial, breast, gastric, lung and ovarian.

      About iCCA or intrahepatic cholangiocarcinomas, this is what the company says:

      "iCCA is a rare type of bile duct cancer that originates from the intrahepatic biliary ductal system and forms an intrahepatic mass. The disease is often diagnosed late because it presents as asymptomatic. There are currently no approved therapies for iCCA."
      Derazantinib has been granted orphan drug designation for iCCA by both the FDA and EMA. The drug has shown a positive response rate in biomarker-defined iCCA population with FGFR2 fusion.

      Trials

      ArQule conducted a phase 1a and phase 2 clinical trial of derazantinib for the treatment of patients with iCCA. A 21% response rate was observed with 6 partial responses out of 29 evaluable patients. An 83% disease control rate and median time was observed on treatment of over 26 weeks. The drug also demonstrated a manageable side effect profile.

      A registrational, single-arm, biomarker- and response rate-driven trial of approximately 100 second-line iCCA patients with FGFR2 fusions was initiated in 4Q-2017, in the United States and European Union. It was later transferred to Basilea after the granting of exclusive license to them in April 2018. In January 2019, Basilea announced results of an interim analysis conducted after 42 patients had been enrolled, with a subset of 29 evaluable patients who had at least one post-baseline imaging assessment. The objective response rate (ORR) was 21% in the 29 evaluable patients. The disease control rate was 83%. The safety data obtained from all 42 patients was also consistent with the previous clinical trial results.

      Execution

      ArQule entered into an agreement with Sinovant in February 2018, granting them an exclusive license to develop and commercialize derazantinib in Greater China. ArQule received an upfront payment of $3 million and a development milestone of $2.5 million in 1Q-2019. Sinovant will be responsible for all costs and expenses of development, manufacture and commercialization of derazantinib in Greater China. The company is also eligible for up to an additional $82 million in regulatory and sales milestones. ArQule is entitled to receive double-digit royalties in the low teens from Sinovant on net sales of derazantinib post-commercialization in Greater China.

      ArQule also entered into an agreement with Basilea in April 2018, granting them an exclusive license to develop and commercialize derazantinib in the United States, European Union, Japan, and the rest of the world, excluding Greater China. An upfront payment of $10 million was paid to ArQule, with eligibility for up to $326 million in regulatory and commercial milestones. Upon commercialization of derazantinib, ArQule will also receive staggered royalties on future net sales, ranging from high-single digits to the mid-teens, and mid-single digits to low-double digits, on direct, and indirect sales, respectively. All costs and expenses of development, manufacture, and commercialization in its territory will be the responsibility of Basilea. Under certain circumstances, ArQule may promote derazantinib in the United States directly.

      ARQ 751: two issued patents in the United States covering the composition of matter or analogs and pharmaceutical composition or analogs, expiring in June 2032. Granted patents in the European Union, Japan and other foreign jurisdictions will expire in June 2032.

      One issued patent in the U.S. covering methods of using ARQ 092 or ARQ 751 will expire in September 2035. A pending U.S. application covering pharmaceutical combinations comprising ARQ 092 or ARQ 751, if issued, will expire in November 2039.

      Market and competition

      The global market of cholangiocarcinoma (CCA) was estimated to be USD 385.07 million in 2018.

      Several companies are working in the FGFR inhibition area according to the company 10-K - these include AstraZeneca (NYSE:AZN), Bayer (OTCPK:BAYZF), BioClin Therapeutics, Debiopharm Group, Boehringer Ingelheim International GmbH, Eisai Co., Ltd. (OTCPK:ESALF), Five Prime Therapeutics (NASDAQ:FPRX), Incyte (NASDAQ:INCY), Johnson & Johnson (NYSE:JNJ), Novartis (NYSE:NVS), Pfizer (NYSE:PFE), Principia Biopharma (NASDAQ:PRNB), Servier and Taiho Oncology.       Risks

      The company has outlicensed its lead drug, and while the iCCA market does not have currently approved competition, this seems to be a pretty small market. That problem should be mitigated by extending the molecule's FGFR inhibition MOA to other indications.

      Opinion

      On the one hand, the fact that ARQL has outlicensed its lead drug to other companies in all relevant global territories means it will only earn royalties and milestone payments. On the other hand, the company will not have to spend anything on the molecule's further development. This sort of R&D-only approach works well if the company has a strong R&D pipeline; this, it has because of the news from ​miransertib, which will probably be license-ready in the next year or so. Given this sort of approach, and the company's current price and strong cash balance, a small position may be warranted.

      ARQL CC zum Q3

      Hauptsächlich ging es um 531. Einerseits zum Update auf der ASH aber auch um die Kommunikation mit der FDA (End of P1 Meeting).
      ASH Abstrakt ist Mitte Juli eingereicht worden (einige Wochen mehr an Daten seit EHA) und wird am kommenden Donnerstag (6.11.) veröffentlicht.
      Zur ASH kommen dann die aktuellen Daten, die sowas wie die finalen Daten sind. (Anfang Dezember)
      Expansion Kohorte P2 ist eröffnet. Interessant auch, dass es einen Arm gibt für Patienten, die Ibrutinib intolerant sind also auch aufgrund der Nebenwirkungen die Behandlung mit Ibrutinib abbrechen. Das wäre nochmal ein sehr großer Markt. Das habe ich vorher anders/falsch geschrieben.
      Man hat mit präklinischen Studien begonnen, um zügig auch Kombinationstherapien im Rahmen der P2 expansion Studie testen zu können.
      Grundsätzlich auch ein guter Ausblick auf ASH, dass es bis jetzt keine ganz schweren Nebenwirkungen gegeben zu haben scheint (keine Dose limiting toxicity bzw. gab es da ja einen Fall)
      Interessant auch die These, dass der Ibrutinib Einsatz dann noch stark ansteigen dürfte, wenn das Patent abläuft. Das würd den Markt für 531 nochmal stark erhöhen. Momentan macht Ibrutinib weltweit bereits 10 Milliarden Umsatz im Jahr!

      Es ist noch nicht genau klar, wie ARQL über den Verlauf der Gespräche mit der FDA berichtet. Insbesondere ist die Frage zu klären, ob die P2 als zulassungsrelevant für C481S CLL und eventuell auch Richters Syndrom gelte kann bzw. wie genau die Studien dann vom Umfang, Endpunkt etc. aussehen müssen. Auch wie die FDA zum Arm Ibrutinib intolerant steht soll Thema sein. Es klingt so, als wäre ein Update zur ASH möglich oder zum CC Q4.

      Miransertib P3 Studie läuft. Bzw. laufen: Protheos und PROS.

      751 finale P1 Daten kommen noch.

      Das nennt man glaube ich „sell the News“. Ich konnte in der heutigen Pressemitteilung nichts negatives ausmachen. Langfristig hat sich an den guten Aussichten nichts geändert.

      Das "Problem" sind die Daten von Lilly/LOXO. Die sehen auf den ersten Blick auch ganz gut aus. Problematisch finde ich, dass Lilly die Daten sehr geschönt präsentiert.
      The first eight patients were evaluable for initial response and 7 tumor responses (87.5%) were observed (by disease-defined criteria): 5/5 CLL patients (1 PR and 4 PR-L

      87,5% tumor response ist aber was anderes als 87% Ansprechrate.
      Auch gibt man nicht PR sondern auch PR-L an. Zudem ist das eine andere Population. ARQL hat vor allem CLL mit C481S Mutation. Man kann die Daten also gar nicht miteinander vergleichen. Aber die Daten von Lilly sehen auf jeden Fall auch nicht schlecht aus. (für einen ersten Blick)
      Man wird jetzt die Daten zur ASH abwarten müssen...

      Hallo Kmasta,

      So, mein Sonnenstrand hat mich nach Hause entlassen.

      Ich habe gestern Abend Deine ArQule - Grafik gelesen, auch den Anstieg mitbekommen.

      Im Urlaub wollte ich nicht handeln, weil auch noch den Trend nach unten besteht, seit dem Juli – mit Anstieg im Anfang Oktober.

      Dieser wurde jetzt durch den Fall von ca. 9,0 auf ca. 7,0 US $ unterbrochen.

      Die beginnende Aufholung könnte sich fortsetzen.
      Ich werde versuchen einen Einstiegspunkt zu finden.

      Um es kurz zu machen: Merck kauft ARQL für 20$

      Da kann man sich die heutigen Daten auf jeden Fall ganz entspannt anschauen!
      Langfristig wäre m.E. mehr drin gewesen, aber in Anbetracht der Konkurrenz durch Lilly evtl. auch ein sinnvoller Zug.

Leider ist aus Aktionärssicht meiner Meinung nach der Preis nicht so wirklich erfreulich. ARQL besteht ja nicht nur aus ARQ-531 und Übernahmen sollten aktuell eher noch ein Premium enthalten. Wieviel Wert wird ARQ-531 damit zugemessen?

      Ich hätte mittelfristig (1-3 Jahre) hier mehr als 10 Mrd USD erwartet, wenn es weiter gut mit ARQ-531 läuft, also mehr als das vierfache dessen, was MRK heute zahlt. ARQ-531 sieht eher nach einem Tagrisso aus, wobei das Potential für einen solchen BTK-Hemmer eher höher liegt. ARQ-531 kann gut ein besseres Imbruvica sein... Abbvie hat am Ende für 50% Imbruvica 21 Mrd USD hingelegt.

      Naja, es besteht allerdings auch noch das Risiko von gravierenden Nebenwirkungen, dann wäre das heute ein guter Deal.

      Man hätte sicher erst die ASH-Daten abwarten können. Dann wäre der Kurs auch so in Richtung 20$ gelaufen. Andererseits ist der Deal sehr gut für die Patienten. Merck kann den Wirkstoff jetzt viel schneller und breiter entwickeln. Ich will mich da insgesamt auch gar nicht groß beschweren! 🙂
      Ich habe erstmal vor, einen Großteil der Aktien zu halten, weil ich eine andere Offerte nicht ausschließen würde. Eigentlich ist ja Merck auch eher eine Überraschung und JnJ bzw. ABBV hätten viel besser gepasst...?
