guilford ist wirklich interessant. Der Börsenbrief Global
Biotech Investing gibt für diesen Wert ein Kursziel von 150 € an, und
das bei einem aktuellen Kursniveau von ca. 36 €!!! Die Krebsforschung
ist wohl eine der wichtigsten Biotech-Segmente, und Guilford ist hier
sehr gut positioniert. Ich kann mir gut vorstellen dass dieses Kurs-
niveau schon deutlich früher erreicht wird. Biotech boomt und Guilford
ist, obwohl schon auf der Kaufliste vieler amerikanischer Invest-
menthäuser, immer noch ein Geheimtip! Wer jetzt nicht kauft ist selber
Schuld!

Guilford Acquires Exclusive Worldwide Rights to Novel Pro-Drug of Propofol, One of the World`s Best Selling Anesthetic Agents
BALTIMORE, March 2 /PRNewswire/ -- Guilford Pharmaceuticals Inc. (Nasdaq: GLFD - news) today announced that it has acquired exclusive worldwide development and commercialization rights to a novel anesthetic compound developed by ProQuest Pharmaceuticals Inc., a privately held pharmaceutical company based in Lawrence, Kansas. The anesthetic agent, PQ-1002 (GPI-15715) is in late-stage preclinical development and is a pro-drug of propofol -- currently the world`s best selling anesthetic agent. Preclinical studies suggest that GPI-15715 could have significant advantages over existing propofol formulations.

Propofol is an intravenous drug that is widely used in a hospital or out-patient setting to induce and maintain anesthesia during surgery. It is also frequently used to sedate patients in intensive care settings or for patients undergoing certain diagnostic or medical procedures. Worldwide sales of propofol in 1999 were estimated to exceed $750 million.

Despite its commercial success, propofol has several reported side effects. These include cardiovascular side effects (such as reduced heart rate and decreased blood pressure), depressed respiration, elevated blood lipid levels (hypertriglyceridemia), pain at injection site, and the potential for bacterial contamination, which could lead to infection. Many of these reported side effects may result from the product`s formulation, which is composed of a non-soluble lipid-based emulsion.

In contrast, GPI-15715 is a water-soluble pro-drug of propofol that is rapidly converted in the body into propofol after intravenous injection. It is formulated in a clear aqueous solution that does not contain a lipid emulsion.

Preclinical studies suggest that GPI-15715 may lack many of the side effects associated with the original propofol formulation. Specifically, in preclinical studies, GPI-15715 did not significantly affect cardiovascular function at doses that produced rapid and effective anesthesia and sedation. In addition, because a lipid emulsion is not required for administration, GPI-15715 may also avoid some of the limitations of the original propofol formulation, such as hypertriglyceridemia, and may be less likely to cause bacterial contamination. Moreover, the pro-drug may have significantly improved stability.

``We view GPI-15715 as an important addition to Guilford`s neurological product pipeline and feel that the acquisition of this technology represents an important opportunity to achieve balanced risk diversification within our Company,`` remarked Craig R. Smith, President and Chief Executive Officer of Guilford. ``Since GPI-15715 is a pro-drug of a successful commercial product, we believe the development risks associated with this product are less than for our novel neurological product candidates that target breakthrough discoveries in neuroscience. Another significant advantage of the product is the opportunity for relatively rapid clinical development. In exchange, ProQuest will receive $1 million in cash and Guilford will receive an equity position in the company. In addition, ProQuest will be eligible to receive milestone payments if the product progresses through clinical development and royalties on any product sales.

Bin ähnlicher Meinung. Hatte bisher fast außschließlich Hightech-Firmen in meinem Depot und habe seit Mitte Dezember etwas umdisponiert. Habe jetzt folgende Biotech-Aktien in meinem Depot:

Andrex (ADRX) -> Generika-Hersteller (Kauf bei 40 $)
Enzon (ENZN) -> Kauf bei 59,30 $
Guilford (GLFD) -> Kauf bei 33 $
Maxim Pharmac. (MMP) -> Kauf bei 58,80 $

Habe aber auch sehr lukrative OTC BB Aktien mit FECC und BLLS, die beide innerhalb von nur einigen Wochen schon über 100 % gemacht haben.

Ich halte die Nasdaq-Werte für viel substandsvoller als die des Neuen Marktes, die sehr oft hohe Kursausschläge durch einzelne Analysten haben. Zudem ist deren Markt oftmals auf Deutschland begrenzt und damit nicht so zukunftsträchtig wie viele amerikanischen Firmen, die einen ganz anderen Markt vor der Tür haben.

bin seit anfang-mitte januar drin. einstiegskurs 22 euro. habe damals
nur eine kleine position gekauft. der tip kam von einem geschäftsfreud
aus new york. werde ihn bei gelegenheit mal zum essen einladen dafür,
wenn ich mal wieder drüben bin. ich habe vor bei gelegenheit noch ein
paar stücke von guilford nachzukaufen, bin wirklich überzeugt von der
story. bevor ich 150 euro für die aktie bekomme gebe ich keine mehr
her! bleibt guilford treu, es wird sich lohnen!

sind die umsätze zu niedrig. wenn du trotzdem unbedingt da kaufen
willst, unbedingt ein limit setzen.
wichtig ist dass du überhaupt kaufst, denn ansonsten würdest du dich
über die verpasste chance sehr ärgern..

Auf jeden Fall werde ich die Aktie unter 100 € nicht abgeben, aber ich bin überzeugt,
dass ich darauf nicht lange warten muss! Fundamentale Daten habe ich
mir bei finance.yahoo.com herausgesucht, kurz und knapp und man kann sich
seinen eigenen Reim drauf machen! Dort findet Ihr auch das aktuelle
Rating der Analysten und das sieht durchaus postitiv aus: 1,19!

Guilford Licenses Novel Discovery From Johns Hopkins University

BALTIMORE, Feb. 16 /PRNewswire/ -- Guilford Pharmaceuticals Inc. (Nasdaq: GLFD) today
announced that it has licensed a discovery from the Johns Hopkins University relating to a unique
biological target, serine racemase, that could be used to develop treatments for neurodegenerative
diseases.
"This is an exciting discovery because serine racemase represents a novel way to potentially block
the harmful effects of glutamate over-stimulation," remarked Dr. Solomon H. Snyder, Director of
Neuroscience at the Johns Hopkins University School of Medicine. Glutamate`s Role in
Degenerative Disorders

The overproduction of glutamate has been implicated in a large number of acute and chronic
degenerative conditions including stroke, epilepsy, peripheral neuropathies, chronic pain and
Parkinson`s, Alzheimer`s and Huntington`s disease.

Glutamate is one of the most abundant amino acids in the body. It is found both in the peripheral
and central nervous system and controls many important functions. During stroke and other
degeneration conditions, however, excessive glutamate is released, which is highly toxic to neurons.
Scientists believe that the harmful effects of excessive glutamate occur principally through activation
of a subtype of the glutamate receptor called the NMDA receptor. The Role and Activity of Serine
Racemase & D-Serine

NMDA receptors have not one, but two sites that must be activated in order for neurotransmission
to take place. Scientists formerly thought that the amino acid, glycine, working in tandem with
glutamate, was the sole activator of this site. Recently, however, scientists at Johns Hopkins, led by
Dr. Snyder, discovered that the D-isomer of the amino acid, serine (D-serine), was the principal
activator of this site. Dr. Snyder`s lab was responsible for isolating and cloning the enzyme, serine
racemase, which converts the L-isomer to the D-isomer.

"Since glutamate is so abundant in our bodies, nature developed a failsafe mechanism to prevent
over-excitation of NMDA receptors by glutamate. It`s very much like a lock that requires two
separate keys. We discovered that D-serine, acting in tandem with glutamate, is required for
NMDA activation," explained Dr. Snyder.

This finding suggested that D-serine plays an important regulatory role in the neurotoxic cascade
which contributes to cell injury and cell death, and that it accomplishes this by acting as a check on
glutamate.

"This discovery provides us with an interesting new biological target for drug development, since
small molecule inhibitors of serine racemase could be anticipated to prevent glutamate neurotoxicity
in conditions like stroke and other neurodegenerative conditions," continued Dr. Snyder.

These findings were recently published in the November 9, 1999 issue of the Proceedings of the
National Academy of Sciences. Building a Neuroprotectant Franchise

Previous therapeutic approaches have focused on blocking glutamate`s ability to bind at the NMDA
receptor site, however, these approaches have been associated with deleterious side effects in
clinical studies. In contrast, Guilford`s NAALADase inhibitor program focuses on blocking
excessive glutamate pre-synaptically. Similarly, the identification and cloning of serine racemase
expands and complements Guilford`s existing neuroprotectant franchise by presenting an additional
opportunity to explore novel ways of blocking glutamate over-stimulation.

As part of the company`s agreement with Johns Hopkins, Guilford has obtained an exclusive license
covering the rights to this discovery and its corresponding intellectual property. In addition, Guilford
acquired a proprietary screening system to identify novel inhibitors of serine racemase.

"The over-stimulation of NMDA receptors by glutamate is believed to be an important initial
pathologic event in stroke and several neurodegenerative diseases. Finding an alternative method of
blocking glutamate activation of the NMDA receptor -- by selectively blocking the synthesis of
D-serine -- could potentially offer new treatment alternatives for conditions like stroke and chronic
degenerative disorders like Parkinson`s and Alzheimer`s disease," remarked Dr. Peter Suzdak,
Senior Vice President of Research and Development at Guilford. "We are excited about pursuing
this novel approach as part of our neuroprotectant research and development program."

TECH INVESTOR Guilford Joins Md. Biotech Highfliers

This may be the Internet Millennium, but it`s biotechnology month in the stock market, with three Maryland biotech stocks hitting 52-week highs yesterday.

MedImmune Inc. of Gaithersburg was up $16.31 1/4, to $178, EntreMed Inc. of Rockville gained $5.50, to $46.87 1/2, and Baltimore`s Guilford Pharmaceuticals Inc. closed up $2.75 at $27.87 1/2.

"There`s general enthusiasm for the biotech sector," said Scott Koonce of Koonce Securities in Rockville.

Institutional investors and fund managers are starting the year by adding new stocks to their biotech portfolios, branching out from established names such as MedImmune and EntreMed.

Guilford has joined the group not only because the biotech rally seems to be broadening, but also because the firm has announced three important developments--involving three different products, said Guilford communications director Stacey Jurchison.

Yesterday Guilford executives told their story to another group of potential investors at a conference in New York sponsored by First Union Securities.

While many biotech firms are pursuing a single avenue of research, Guilford is going down a pair of paths, focusing on drug-delivery systems for fighting cancer as well as a cluster of neurological drugs.

On the drug-delivery side, Guilford recently began a new round of tests involving 240 patients for its treatment for brain tumors. To create a time-release tumor treatment, Guilford packs chemotherapy drugs into a little biodegradable wafer that`s implanted in the cavity left when a tumor is removed surgically. Already used for recurrent tumors, the wafer is now being tried as a first-time treatment.

In December, Guilford announced another drug-delivery research initiative by filing a request to test a new ovarian cancer treatment that packs the drug Taxol into tiny "microspheres" that can be injected into the body.

Guilford was one of 11 firms whose Taxol research was detailed in a year-end report by Sturza`s Institutional Research, a New York firm that specializes in medical stocks. Taxol is now the largest-selling anti-cancer drug, generating sales of about $1.5 billion last year, the study noted.

And in partnership with the big biotech firm Amgen Inc., Guilford is getting ready for the second phase of trials of a drug that treats Parkinson`s disease by stimulating the regrowth of neuron cells deep in the brain.

"What I like about them is that they are not just a one-product deal. They`ve got a research pipeline of four or five different products they`re working on," Koonce said. "If any one of them hits, the stock could take off."

Koonce said Guilford`s shares didn`t catch the biotech wave when it began to rise last year because investors were focused mostly in genomics and antibody research. "Everything else kind of languished."

Guilford still reports regular losses but its finances have matured beyond the cash-burning stage that`s common in biotech.

The company`s picked up a $5 million payment from Amgen in December and its brain tumor wafers are generating about $20 million in sales via a pair of big drug company partners. Jurchison said about 35 percent of that flows directly to Guilford, which manufactures the product and collects royalties on its research.

Guilford`s balance sheet also is unusual by biotech standards--showing $145 million in cash. Roughly one-third of that came in last September when the firm sold $45 million worth of stock in a private placement managed by Prudential Vector Healthcare Group.

The institutions that bought those 3.4 million shares must be among the investors most satisfied with the recent run-up in Guilford stock. They paid just $13.50 a share for their stock and have doubled their money in less than four months.

jahres in der biotechszene. guilford ist ja noch beinahe unentdeckt
und hat eine blendende zukunft vor sich. gerade der artikel den du
gepostet hast ist vielvesprechend, die zukunft von guilford hängt eben
nicht nur von einem einzigen produkt ab wie bei anderen biotechs. ich
denke ich werde in dieser woche nochmal ein paar stück nachlegen.

Ich bin ganz ehrlich-ich bin zwar absolut ueberzeugt,dass guilford eine sehr gute aktie ist,aber ich wuerde dir es auch nicht uebel nehmen,wenn du dir denkst ,der wastl ,der empfiehlt mir nur die aktie um selbst davon zu profitieren!

Also ich sag nur - tue das was du fuer richtig hälst-verlass dich auf keinen anderen!!!!
Sonst wirst du schamlos ausgenutzt!!
Besorg dir einfach ein paar artikel aus den usa-eventuell noch ein paar analystenstimmen und entscheide dann ,was du machst!!!

As the wafer erodes in the brain, it releases the cancer chemotherapeutic drug, carmustine (BCNU), directly to the tumor site in high concentrations over an extended period of time.

Gliadel was first approved in the United States by the FDA in the fall of 1996 for use as an adjunct to surgery to prolong survival in patients with recurrent GBM for whom surgical resection is indicated. In 1996, Guilford Pharmaceuticals and Rhône-Poulenc Rorer (RPR)—now Aventis Pharma—entered into a licensing agreement granting RPR worldwide marketing rights (currently excluding Scandinavia and Japan), for Gliadel.

Gliadel offers an important treatment option for neurosurgeons and complements other standard therapies for brain cancer. In a randomized clinical trial, Gliadel was shown to prolong survival in patients with recurrent GBM for whom surgical resection is indicated. In this multicenter study involving 222 patients with recurrent malignant glioma, Gliadel increased six months survival from 36% with placebo to 56% among the 145 patients with recurrent GBM.

It is currently being studied in a Phase III trial in conjunction with surgery and radiation in patients with newly diagnosed malignant glioma. This randomized, double-blind, placebo-controlled study is being conducted at 42 sites in 12 countries, and the primary endpoint is overall survival 12 months after patient enrollment ends.

The spectrum of adverse events observed in patients with recurrent malignant glioma who received Gliadel or placebo was consistent with that encountered in patients undergoing craniotomy for malignant glioma. Clinically relevant side effects that were more common with Gliadel versus placebo included healing abnormalities, brain edema, and intracranial infections. Seizures that occurred in patients receiving Gliadel had a much earlier onset, although the incidence of seizures was the same in the placebo and Gliadel groups.

hallo rama,
scheint keine schlechten nachrichten von guilford zu geben, der markt
zieht sie halt mit runter. ich würde sagen das ist eine gute gelegenheit
zum nachkaufen. es mag sein dass die biotechbranche schon gut gelaufen
ist, ich denke aber auch dass firmen wie guilford, die sich nicht nur
auf ein produkt spezialisieren, die besten chancen haben werden in
zukunft. und ein kurs von 100 € ist das mindeste was ich mir von guilford
erwarte. ich werde erst verkaufen wenn sie bei 150 sind, und ich bin mir
sicher dass das noch dieses jahr sein wird!
