Mylotarg steht des Weiteren vor einer Rückkehr (Verbessertes Dosingsschedule, geringere Dosierung, in Kombination mit Chemo...)
Lässt hoffen, dass nicht CD33 als Target ein Problem darstellt, sondern die Tox der Payload in "normalen" Zellen...was für IMGN779 spräche, falls die Effektivität stimmt
Wie sich IMGN779 aber entwickeln wird, wird die Zeit zeigen...

Für IMGN ist das im Hinblick auf die unmittelbare Konkurrenzsituation zunächst positiv, die Börse hat es ja auch bereits mit einem kräftige Kurssprung honoriert. Die Frage ist nur, ob IMGN779 nicht auch noch in der Indikation AML scheitern wird ... bei SGN-CD33A hat es ja einige Jahre gedauert, bis sich die Probleme (Lebertoxizität, fatale Infekte) offenbart haben. Nicht zu vergessen auch, dass IMGN779 Konkurrenz von bispezifischen anti-CD33-Antikörpern und insbesondere der CAR-T-Technologie droht.

BOTHELL, Wash.--(BUSINESS WIRE)--Seattle Genetics, Inc. (Nasdaq: SGEN), a global biotechnology company, today announced that it is discontinuing the phase 3 CASCADE clinical trial of vadastuximab talirine (SGN-CD33A) in frontline older acute myeloid leukemia (AML) patients. Seattle Genetics took this action following consultation with the Independent Data Monitoring Committee (IDMC) and after reviewing unblinded data on June 16, 2017. The data indicated a higher rate of deaths, including fatal infections in the vadastuximab talirine-containing arm versus the control arm of the trial. Based on available data, the safety concerns in this trial do not appear related to hepatotoxicity. Seattle Genetics is suspending patient enrollment and treatment in all of its vadastuximab talirine clinical trials including the ongoing phase 1/2 clinical trial in frontline high risk myelodysplastic syndrome (MDS). Seattle Genetics will closely review the data and consult with the U.S. Food and Drug Administration (FDA) to determine future plans for the vadastuximab talirine development program.

AML is a devastating disease with a poor prognosis in most patients, and there is a great need for therapeutics against this disease. The phase 3 CASCADE clinical trial is a randomized, double-blind, placebo-controlled study evaluating vadastuximab talirine in combination with the hypomethylating agents (HMAs) azacitidine or decitabine compared to an HMA alone in older patients with newly diagnosed AML. Vadastuximab talirine is a novel investigational ADC targeted to CD33 utilizing Seattle Genetics’ proprietary ADC technology. CD33 is expressed on most AML and MDS blast cells.

IMGN779 zu sehen oder hören bekommen und ob das ausreicht Kursphantasien anzuheizen oder einen Selloff lostritt. Wie gesagt verstehe ich nicht wieso die Firma den Trigger für die KE noch nicht ausgeführt hat. Auf was warten die?

Probleme mit unerwünschten Nebenwirkungen sind bei SGN33A erst in späteren Studien im Zusammenhang mit allogenen Stammzelltransplantationen aufgetreten.
Was das Target CD33 betrifft, so wird IMGN779 auch der Konkurrenz von bispezifischen Antikörpern (z.B. von Amgen) sowie CAR-T-Präparaten ausgesetzt sein. Insoweit bin ich mit meinen Erwartungen an IMGN779 zum jetzigen Zeitpunkt noch zurückhaltend.

Hinweise darauf sollte man bei Immunogen inzwischen haben. In gut einer Woche sollten wir dann auch mehr wissen und erste Daten zu IMGN779 erfahren.
