US FDA advisers turn down ViroPharma common cold drug
(UPDATE: Adds background on Picovir)

GAITHERSBURG, Md., March 19 (Reuters) - A U.S. advisory panel on Tuesday rejected ViroPharma Inc.`s (NasdaqNM:VPHM - news) common cold treatment Picovir, the first drug to attack the cause of the illness and not just the symptoms.

The panel voted unanimously to urge the Food and Drug Administration (FDA) to decline to approve Picovir. The agency usually follows its panels` advice.

Aventis SA (NYSE:AVE - news) is co-developing Picovir, known generically as pleconaril.

The drug strikes viruses that cause more than half of all colds, and ViroPharma`s studies showed it cut the illness short by about one day.

Patients need to start taking Picovir within 24 hours after symptoms start. Treatment lasts five days, and the drug must be taken with food three times a day. Approval by the FDA would make Picovir available by prescription only.

Picovir works by sliding into a pocket on the surface of picornaviruses. By occupying the pocket, Picovir prevents the virus from copying itself and infecting healthy cells.

GAITHERSBURG, Md., March 19 (Reuters) - A drug that treats the common cold by attacking its cause, rather than just symptoms, suffered a setback Tuesday as U.S. advisers unanimously said regulators should not approve the medicine.

Picovir, a pill made by ViroPharma Inc (NasdaqNM:VPHM - news) and co-developed with Aventis SA (NYSE:AVE - news), is the first medicine to strike viruses responsible for more than half of the 1 billion colds that afflict Americans each year. Over-the-counter remedies only mask symptoms such as coughing and a runny nose.

A U.S. Food and Drug Administration (FDA) advisory committee urged the agency to reject ViroPharma`s application to sell the drug in the United States. The FDA usually follows its panels` advice.

Panel members said Picovir produced a modest benefit in some patients. In clinical trials, patients` colds cleared up about a day faster on average than with a placebo.

Members said they were not convinced, however, that the drug was safe enough to treat an illness that is not life threatening and goes away on its own.

``I have not seen adequate (data) from my point of view to conclude this drug is safe as we anticipate it will be used,`` said Dr. Brian Wong, a panel member.

FDA staff raised various safety concerns, including the possibility that Picovir may reduce effectiveness of oral contraceptives. Two women treated with Picovir for six weeks in a study testing whether the drug may prevent colds became pregnant while using birth control pills.

Panel members also worried that the drug may stimulate development of resistant cold germs.

Patients need to start taking Picovir within 24 hours after symptoms start. Treatment lasts five days, and the drug must be taken with food three times a day. If approved, Picovir would be available by prescription only.

Picovir targets picornaviruses, the viruses that cause more than half of all colds, and up to 80 percent in the peak spring and fall seasons.

Picovir works by sliding into a pocket on the surface of picornaviruses. By occupying the pocket, Picovir prevents the virus from copying itself and infecting healthy cells.

``The development of the first antiviral drug to treat the predominant cause of the common cold is a landmark development in the history of anti-infectives,`` Dr. Ellen Cooper, a ViroPharma vice president, told the FDA panel.

ViroPharma is based in Exton, Pennsylvania.

ViroPharma stock plummets after Fulcrum says `sell`
NEW YORK, March 14 (Reuters) - Shares of ViroPharma Inc. (NasdaqNM:VPHM - news) tumbled nearly 24 percent to their lowest level in a year on Thursday after an analyst said he expects U.S. regulators to reject the biotechnology company`s drug for the common cold.

Next week a U.S. Food and Drug Administration advisory committee will make a recommendation on whether the FDA should approve ViroPharma`s Picovir drug, which is also designed to treat respiratory infections. The committee`s recommendations are usually, but not always, followed by the agency.

Darren Mac, an analyst at Fulcrum Global Partners said in a report that late-stage trials of Picovir showed levels of efficacy that were ``highly variable,`` and that the FDA could ask for additional information for how the drug works in patients such as children and the elderly that are at higher risk for viral respiratory infections.

``We believe that the upcoming Antiviral Drugs Advisory Committee meeting for Picovir will result in a recommendation against approval to the Food and Drug Administration,`` Mac said. ``Our expectations are also supported by what we view as a very weak co-promotion partnership and recent selling and a hedging arrangement by a director.``

Exton, Pennsylvania-based ViroPharma`s shares fell $4.12, or 23.9 percent to $13.32 in afternoon trading on Nasdaq. Mac said in a report he believes the shares could fall as low as $7.50 a share if the drug is rejected on March 19.

Mac said he expects the FDA to scrutinize the drug`s safety particularly carefully since the common cold is not usually a life-threatening disease. He said that in trials 3.3 percent of patients dropped out because of side effects, compared to 2 percent in the control group.

Viropharma officials were not immediately available for comment.

ViroPharma last September formed a co-promotion partnership with Aventis SA . But Mac said investors should not consider this validation for Picovir since $20 million of the $25 million Aventis has paid to ViroPharma is repayable over the next two years if certain milestones are not met.

Mac also said recent stock sales and a forward contract by a director of the company also raised concern.

Das Medikament ist für die Behandlung von Erkältungen gedacht. Dieser Schritt ist für das Unternehmen schwerwiegend, da das Mittel ein Hauptproduktkandidat für Viropharma ist. Derzeit ist die Aktie noch vom Handel ausgesetzt (BörseGo berichtete).

NEW YORK (Reuters) - Shares of ViroPharma Inc. (NasdaqNM:VPHM - news) tumbled nearly 59 percent on Wednesday, a day after a U.S. regulatory panel failed to approve its experimental drug for the common cold.

Shares of the Exton, Pennsylvania-based biotechnology company fell $7.91 to $5.50 on Nasdaq, making it the percentage loss leader on the tech-heavy composite Wednesday. The shares, halted for trading on Tuesday on the news, also hit a 52-week low, with the prior low at $12.32.

ViroPharma said after the market closed Tuesday that an advisory committee of the U.S. Food and Drug Administration rejected the drug, Picovir, saying it was not convinced it was safe enough to be widely prescribed.

The panel`s recommendation, which is usually followed by the FDA, is a severe blow to ViroPharma, which has no other drugs in late-stage development.

Company officials said they will meet with the FDA to discuss what it needs to do to persuade regulators to clear the drug. ViroPharma said studies underway may be able to answer questions raised by the FDA.

``This drug is not dead,`` said Mark McKinlay, a ViroPharma spokesman said after the panel reached its decision.

The panel questioned whether cold germs may mutate, making the drug ineffective. It was also concerned that Picovir could reduce the effectiveness of birth control pills.

Picovir was to have been marketed in the United States and Canada by Aventis SA , the European drug company. Aventis paid $25 million for the marketing rights, but ViroPharma may have to refund $20 million of that amount if it fails to meet certain milestones.

Na ja, wundert mich nicht, daß sie Picovir wieder abgelehnt haben.
Habe nie verstanden, daß viele auch nach den immer wiederkehrenden schlechten Meldungen an VPHM festgehalten haben, teilweise sogar wieder eingestiegen sind.
Kann mich da an einige heiße und teilweise ein bißchen unfreundliche Diskussionen erinnern, in diesem Board; dies ging so weit, daß ich frustriert aus der Diskussion ausgestiegen bin.
Ich halte gar nichts von Scarface, aber bei VPHM hat er einfach recht.
Do not buy; das ist meine Empfehlung!
Diese Aktie ist zur Zeit was für 100%tige Zocker, keine Aktie für "vernünftige" (nennen wir sie mal etwas langfristiger anlegende) Anleger.
Wenn ihr einen gefallenen Engel haben wollt: Kauft euch MAXM, bei denen bin ich überzeugt, daß sie wieder auf die Beine kommen werden. Dies wird aber JAHRE dauern!!!
Für Picovir sehe ich einfach schwarz.

Die können von der FDA immer noch die Zulassung erhalten, die arbeiten gerade sehr an den Beanstandungen. Die FDA nimmt das Beratergremium normalerweise als Empfehlung, aber normalerweise ist nicht immer und wenn Dinge nachgereicht und besprochen werden, kann es auch mal anders kommen s. United Therap.

Natürlich werden sie wieder steigen; fragt sich bloß von wo, wie lange und bis wohin.
Ob ihr die jetzigen Kurse alsbald wieder sehen werdet ist fraglich aber nicht unmöglich.
Ihr seid Optimisten, daß ist schön, an der Börse aber nicht immer angebracht.
Sicher besteht die Möglichkeit, daß die FDA sich anders entscheidet als das Beratergremium.
Kommt ja mal vor, alle Jubeljahre einmal.
Hätten bei den letzten Versuchen von Picovir auch geschehen können. Hat aber nicht.

@tester: Vergiss es! Ohne jede Führung und bei Sicherheitsbedenken wird die FDA sich niemals sich für eine Zulassung entscheiden. Die FDA vermindert augenblicklich radikal die Typ I Fehler auf Kosten der Typ II Fehler, was bei einer Behörde auch nicht weiter verwundern muss.

@TheRed: Picovir ist vom Gremium als effektiv deklariert worden. Der Ablehnungsgrund sind ungenügende Sicherheitsdaten. Und Picovir wird weiter bei Säuglingen getestet werden.

Das war doch auch schon beim letzten mal der Ablehnungsgrund, wenn ich mich richtig erinnere.

Ich habe das Gefühl, daß die FDA (bzw. das Gutachtergremium) VPHM gefressen haben, wie schon häufiger wohl vorgekommen (frag jetzt nicht nach einem Beispiel, ich wußte mal ein paar, hab sie aber wieder vergessen).
Die werden auch wohl weiterhin große Schwierigkeiten haben.
Na ja, immerhin haben sie noch mehr als ausreichend Kapital!

Das war deren erstes NDA!
Ich glaube das Verhältnis der FDA zu Gliatech ist schlechter, aber das Thema dürfte gegessen sein. Die eigentlichen FDA-Gutachter waren auffallend neutral. Unglaublich, wie unterschiedlich die ihre Daten präsentieren. Einmal als neutraler Anwalt, mal als Teufels Advokat.

Einige Hauptkritikpunkte des Expertengremiums:

a)Seltene zwischenmenstruale Blutungen und Interferenz mit Kontrazeptiva

b)Imbalanz in der Studienteilnehmer zugunsten der Frauen (2/3 zu 1/3). Das wiegt IMHO eigentlich relativ schwer, da Frauen scheinbar besser auf Picovir ansprachen. Es wurde überraschenderweise vom Gremium kaum diskutiert. Aber da Picovir ganz offensichtlich auf den Östrogenhaushalt einwirkt, könnte ein krankheitsreduzierender Effekt eben durch diese Nebenwirkung erzeugt worden sein. Es wird sogar die Wirkung von Östrogenen auf Alzheimer untersucht, warum sollten die nicht auch Einfluss auf das Krankheitsbild von Erkältungen haben?

c) Sorge vor Kreuzresistenzen mit anderen antiviralen Wirkstoffen

Ansonsten war der grobe Tenor: Angst vor bewusster und unbewusster Fehlbenutzung und keine Nebenwirkungen bei solchen leichten Krankheiten.
Bei konsequenter Fortsetzung dieser Denkweise hätte die FDA dann aber auch nie Viagra und diverse andere Blockbuster zulassen dürfen.

Picovir dürfte IMHO für die Anwendung VRI tot sein. Bei drei Gutachtern (u.a. Dr. Brass) hatte man den Eindruck, die hätten Picovir auch nicht zugelassen wenn die Erkältung sofort verschwunden wäre und die "schwerste" Nebenwirkung temporäre Übelkeit gewesen wäre.

Bin an VPHM aber nicht mehr interessiert, deshalb verfolge ich die Entwicklung nicht mehr regelmäßig.
Übrigens, wo hast du die Daten (Ablehnungsgründe) her, die Quelle klingt ganz interessant.
