Corr developed this trial in collaboration with Clovis Oncology. CU Cancer Center will be the primary site, Additionally, this trial will be nationally enrolling patients at two other institutions; The University of Pennsylvania and Fox Chase Cancer Center.
"The Phase II clinical trial aims to determine the effectiveness of the anti-cancer drug Rucaparib as maintenance therapy for patients with metastatic or recurrent endometrial cancer that have been treated initially with chemotherapy," explains Corr. "Very basically, Rucaparib, a PARP inhibitor, works by stopping cancer cells from repairing their DNA appropriately. Cancer cells can hijack the body's system to repair their own DNA, which allows them to multiply. Without the ability to repair damaged DNA, the cancer cells eventually die. Rucaparib is an FDA- approved oral drug used in other cancer types and has been well studied."
Currently PARP inhibitors are utilized mostly in BRCA ovarian cancer patients, with outstanding sucess. However, preclinical data has demonstrated that patients with a PTEN deficiency may respond to PARP inhibitors as if they had a BRCA mutation. The PTEN mutation is found in over 80% of endometrial cancer. This trial will be one of the first to use this drug class in endometrial cancer.
"I believe that this clinical trial could be the foundation step to practice-changing therapy," says Corr. "There are currently no approved maintenance therapies in endometrial cancer, and if this trial demonstrates similar success rate to what we have seen in ovarian cancer, I believe this drug class could become a part of standard care for some patients."

Clovis up 3% premarket on Rubraca data in pancreatic cancer

Clovis Oncology (NASDAQ:CLVS) is up 3% premarket on light volume in reaction to interim results from an investigator-initiated Phase 2 study evaluating Rubraca (rucaparib) as first-line maintenance therapy in pancreatic cancer patients following induction with platinum-based chemo. The data were presented at AACR in Atlanta.

Preliminary results suggest that treatment with the PARP inhibitor provides a disease control benefit in patients with BRCA1, BRCA2 or PALB2 mutations, a population representing 5 - 8% of pancreatic cancer cases.

The primary endpoint of the 42-subject study is progression-free survival (PFS). In 19 evaluable patients, median PFS was 9.1 months from the start of Rubraca treatment. Median overall survival has not been reached. The response rate was 36.8% (n=7/19), including one complete responder. The disease control rate (responders + stable cancer) was 89.5% (n=17/19) at week 8.

Pancreatic cancer is one of the most treatment-resistant cancers with a five-year survival rate of only 8.5%.

GlaxoSmithKline (NYSE:GSK), recent acquirer of PARP inhibitor developer TESARO, is up 1% premarket.

Okay, die haben in 2018 ca. 95 Mio $ Umsatz mit ihrem Hauptprodukt Rubraca gemacht.
Rubraca ist jetzt auch für die EU und zugelassen und in Deutschland bereits erhältlich. Der Wirkstoff in Rubraca, Rucaparib, hemmt die Aktivität einer als Poly(ADP-Ribose)-Polymerase (PARP) bezeichneten Proteinfamilie, die an der Reparatur beschädigter DNA in den Zellen (sowohl normaler als auch Krebszellen) beteiligt ist. In normalen Zellen besteht ein alternativer Mechanismus für die DNA-Reparatur, dieser alternative Mechanismus funktioniert jedoch nicht ordnungsgemäß in Krebszellen mit Mutationen in den BRCA-Genen. Wenn also die PARP-Enzyme blockiert werden, kann die beschädigte DNA in den Krebszellen nicht repariert werden. Dies hat zur Folge, dass die Krebszellen absterben.
Derzeit wird Rubraca nur für Patientinnen verwendet, die bereits mit mindestens zwei platinbasierten Arzneimitteln behandelt wurden, jedoch nicht länger mit diesen Arzneimitteln behandelt werden können, daher der derzeit noch verhaltene Umsatz. Clovis will seinen Marktanteil durch eine Zunehmende Anwendung in der Weiterbehandlung (second-line maintenance treatment) für diese Krebsart steigern. Dazu hat die FDA Rubraca als „Breakthroug Therapy“ für BRCA mutiertem metastasiertem kastrationsresistenten Prostatakrebs anerkannt. Hinzu kommen zahlreiche klinische Studien für die Anwendung bei einer Vielzahl an soliden Tumoren einschließlich Prostatakrebs und Blasenkrebs. Hier gibt es auch bereits eine breite Kooperation mit Bristol-Meyers Squibb für die Kombination von Rubraca mit deren Produkten.
Es gibt also reichlich Phatasie und Wachstumspotential. Der Laden hat noch satte 520 Mio $ cash und das bei einer Marktkapitalisierung von 1,4 Mrd $...also ich sehe hier ein mehr als ordentliches Chance/Risiko Verhältnis und erwarte einen positiven News Flow…bei den Daten dürfte auch das ein oder andere Übernahmegerücht aufkommen…ich riskiere mal eine kleine Position…

Hi, ich bin wieder bei Clovis eingestiegen.
Ausschlaggebend war der kurze Bericht in der letzten Börse Online, in dem
spekuliert wird, dass Clovis demnächst übernommen werden könnte.
Ich kann jedoch nicht den wahren Wert des Medikament einschätzen.
Hat hier jemand eine fundiertere Einschätzung?

die Phantasie bei Clovis liegt doch hauptsächlich in der potenziellen Übernahme, oder?
Bei Tesaro hat das ja super funktioniert, wenn auch der Kurs vorher komplett abgestürzt ist.
