Auch wenn Elan noch ein Weilchen bis zur Markteinfuehrung brauchen wird, publizieren tun sie in den angesehensten Journals - zuletzt im August in Nature Medicine.

One hallmark of Alzheimer disease is the accumulation of amyloid beta-peptide in the brain and its deposition as plaques. Mice transgenic for an amyloid beta precursor protein (APP) mini-gene driven by a platelet-derived (PD) growth factor promoter (PDAPP mice), which overexpress one of the disease-linked mutant forms of the human amyloid precursor protein, show many of the pathological features of Alzheimer disease, including extensive deposition of extracellular amyloid plaques, astrocytosis and neuritic dystrophy. Active immunization of PDAPP mice with human amyloid beta-peptide reduces plaque burden and its associated pathologies. Several hypotheses have been proposed regarding the mechanism of this response. Here we report that peripheral administration of antibodies against amyloid beta-peptide, was sufficient to reduce amyloid burden. Despite their relatively modest serum levels, the passively administered antibodies were able to enter the central nervous system, decorate plaques and induce clearance of preexisting amyloid. When examined in an ex vivo assay with sections of PDAPP or Alzheimer disease brain tissue, antibodies against amyloid beta-peptide triggered microglial cells to clear plaques through Fc receptor-mediated phagocytosis and subsequent peptide degradation. These results indicate that antibodies can cross the blood-brain barrier to act directly in the central nervous system and should be considered as a therapeutic approach for the treatment of Alzheimer disease and other neurological disorders.

Elan ist sicherlich eine spekulative Investition, mit aber durchaus positiven Aussichten. Wer das Risiko nicht scheut, koennte in ein paar Monaten zu den sehr gluecklichen Leuten gehoeren. Nicht auszudenken was mit dem Kurs passiert, wenn AN-1792 als Impfstoff im Menschen wirkt, wie es bereits fuer die PDAPP-Maus gezeigt wurde. Und bisher laufen die vorklinischen Tests sehr vielversprechend...
Wenn aus der der Alzheimer Story nichts wird, hat Elan aber auch andere Produkte in der Pipeline, die den Unternehmenswert ebenfalls steigern duerften. Ein Beispiel, das Brustkrebsmedikament MyocetT, eine galenische Verbesserung des zur Zeit schon verfuegbaren Doxorubicins. MycetT bringt den Wirkstoff Doxorubicin direkter zu den Krebszellen und verringert dadurch die Nebenwirkungen erheblich.

PS: Die Kurse von morgen sind die Gewinne von uebermorgen. Im Klartext: Wenn gezeigt wurde, dass AN-1792 beim Menschen wirkt, spielt es fuer den Aktienkurs keine Rolle ob das Medikament in 3 oder in 10 Jahren zugelassen wird!

bin neu hier und weiss nicht, ob dies jemand interessiert:

Ich habe gerade noch ein bischen über Elan rechiert.

Es gibt eine Zusammenarbeit zwischen Elan und dem Pharmagiganten American Home. Dies dürfte vor allem für die Markteinführung des Alzheimer-Impfstoffes wichtig sein. Eine relativ kleine Firma wie Elan hat für eine internationale Einführung keine entsprechende Marketforce (d.h. Pharmaberater, die ein neues Medikament in Krankenhäusern und bei Ärzten anpreisen).
Außerdem zeigt diese Kooperation m. E., dass große Firmen an den Ansatz, die Plaquebildung im Gehirn durch eine Impfung zu stoppen oder zu verhindern, glauben. Dies gilt auch nach dem Ausstieg von Eli Lilly aus der Kooperation mit Athena Neuroscience (dem eigentlichen Erfinder des Impfstoffes, der von Elan aufgekauft wurde).

--Partnership Joins Two Biotechnology Leaders to Address Important Medical Need--
Madison, NJ and Dublin, Ireland, April 5, 2000

A research alliance between United States pharmaceutical and biotechnology company American Home Products Corporation (NYSE: AHP) ("AHP") and the leading Irish pharmaceutical and biotechnology company, Elan Corporation, plc (NYSE: ELN) ("Elan"), was announced today to develop a vaccine that may be used for the treatment of mild to moderate Alzheimer`s disease and possibly to prevent the onset of the disease. This alliance in the fight against Alzheimer`s disease links two leaders with proven track records in biotechnology.
The framework of the alliance will enable AHP and Elan to collaborate in research, development and commercialization of an experimental therapy called AN-1792. Elan`s preclinical research has shown in transgenic mice that AN-1792 reduces and prevents the development of amyloid plaque, a substance that is believed to be associated with Alzheimer`s disease. Elan has commenced Phase I human safety studies in Alzheimer`s disease patients in the United States and United Kingdom.
"The pre-clinical data are very promising in light of the significant need for therapies that can alter the course of Alzheimer`s disease -- treatments that do not exist today," stated Donal J. Geaney, Chairman and Chief Executive Officer of Elan Corporation, plc. "We are excited about working closely with AHP, a leader in biotechnology and vaccine research. We hope to deliver a breakthrough for one of the most devastating diseases that is currently affecting the world`s aging population."
Commenting on the alliance, John R. This research alliance includes a five-year research program for the discovery and characterization of additional products to be developed under the collaboration. At the outset of the alliance, AHP will make an initial payment to Elan. Additional payments to Elan are outlined in the agreement for key development benchmarks and regulatory milestones. Costs and revenues in future years, from AN-1792 and other alliance products, will be shared equally by AHP and Elan.
