The trial (REO 014) is a Phase II, open-label, single agent study whose primary objective is to measure tumour responses and duration of response, and to describe any evidence of antitumour activity of intravenous, multiple dose REOLYSIN® in patients with bone and soft tissue sarcomas metastatic to the lung. REOLYSIN® will be given intravenously to patients at a dose of 3x1010 TCID50 for five consecutive days. Patients may receive additional five-day cycles of therapy every four weeks for a maximum of eight cycles. Up to 52 patients will be enrolled in the study.
Eligible patients must have a bone or soft tissue sarcoma metastatic to the lung deemed by their physician to be unresponsive to, or untreatable by standard therapies. These include patients with osteosarcoma, Ewing sarcoma family tumours, malignant fibrous histiocytoma, synovial sarcoma, fibrosarcoma and leiomyosarcoma.

ANALYSIS OF NEWS-ORPHAN DRUG STATUS FOR REOLYSIN (Not rated) 11-Apr-07 03:55 am The following is my analysis of the significance of the recent news.Orphan Drug Status is possible for Reolysin used for metastisized to the lung sarcoma since the condition incidence is low and since the current treatment relies on surgery,alone which is not very effective.Orphan Drug Status would allow approval of Reolysin if some Phase II patients had complete tumor remission along with evidence of tumor supression or Reolysin bioactivity against the tumor(new FDA rules).In this case, a very early Reolysin approval as soon as 6 months after completion of Phase II could be expected under FDA Orphan Drug Status rules.Early approval of Reolysin for this condition would provide funding for further trials using Reolysin in combination with radiation and other chemo agents.Reolysin would also be considered first line treatment for this condition since Chemotherapy does not work after surgery.Excerpted from the enclosed link:"Soft tissue sarcoma is a rare neoplasm: there are approximately 6600 cases annually in the United States. 1 Sarcoma may arise virtually anywhere, but the extremity is the most common primary site.
