AURORA, ONTARIO--(CCNMatthews - Jun 23, 2004) - Progress continues to be made on the Company`s DOS47 drug development program. A preliminary series of tumour screening studies have been completed using immune-deficient mice bearing various human cancer xenografts. These studies were conducted using different doses and treatment regimens to discern whether different tumour types would have different sensitivities to DOS47. Following intratumoural injections, tumour growth rate, cell culture studies and histological examination of biopsy samples were used to assess treatment effects. Among those human cancers studied, mice bearing human lung tumour xenografts appeared most sensitive showing notable growth retardation and tissue necrosis relative to controls. Sensitivity was also observed in mice bearing human ovarian cancer xenografts.

What is DOS47?

DOS47 is a new drug candidate under development at Helix that employs a novel approach to the destruction of cancer cells. This novel approach is based upon Helix`s theory that the urea cycle, a natural process in the body, can be modified to fight cancer.

Urea is a natural waste product found throughout the body. The key purpose of urea is to rid the body of ammonia, one of the primary metabolic waste products produced during the normal breakdown of proteins in cells. Urea is produced by combining ammonia and carbon dioxide in the liver through a metabolic process called the urea cycle.

DOS47 is based upon a naturally occurring enzyme called urease. It is thought to essentially reverse the urea cycle by breaking down urea into ammonia and carbon dioxide. The novel DOS47 treatment concept was conceived to reverse the urea cycle locally at the site of cancerous tumours in the body. By doing so, the local production of ammonia and carbon dioxide is believed to initiate a cascade reaction which suppresses the active metabolic machinery of tumour cells through a combination of effects. Among these theorized effects, the local production of ammonia at the site of cancerous tumours is thought to disrupt cancer cells via chemical action. As well, the local reaction is believed to raise the pH of the microenvironment surrounding the cancerous cells, effectively reversing the acidic extra-cellular conditions in which most cancer cells are believed to exist and multiply.

Next steps

Over the next 12 to 18 months, Helix intends to develop targeting agent conjugation methodologies and analytical capabilities, conduct acute toxicology studies, and perform a series of proof of concept studies in animals using specific DOS47 conjugates.

Central to Helix`s DOS47 program will be work to establish conjugation methodologies in order to combine DOS47 with biological targeting agents such as antibodies. The purpose of this developmental work is to enable DOS47 therapeutics to be designed to theoretically target different cancer types in the body, by using cancer type-specific targeting agents to anchor DOS47 to cancer cells for maximal tumouricidal effect. This would theoretically enable DOS47 to be used via systemic injection for metastasized cancers, as well as via direct intratumoural injection for solid tumours.

In order to facilitate this work, Helix has further upgraded its Edmonton, Alberta oncology laboratory facilities and hired additional scientific personnel.

NEUSTE INFORMATINEN ZU DOS47
Aurora, Ontario - Das Programm zur Entwicklung des Arzneimittels DOS47 macht weiterhin Fortschritte. Eine erste Reihe von Tumorvorsorgestudien mit iImmundefizienten Mäusen, denen verschiedene humane Tumor-Xenografts implantiert wurden, ist nunmehr abgeschlossen. Die Studien wurden unter Einsatz von unterschiedlichen Dosen and
Therapieregimen durchgeführt, um festzustellen, ob verschiedene Tumorarten unterschiedlich auf DOS47 reagieren.
Nach Verabreichung von intratumoralen Injektionen wurden die Wirksamkeit der Therapien anhand von Tumorwachstum,
Zellkulturstudien und histologischen Untersuchungen von Biopsiematerial beurteilt. Unter den im Rahmen der Studie
untersuchten menschlichen Krebsarten schienen Mäuse mit humanen Lungentumorxenografts am stärksten zu reagieren.
Sie wiesen gegenüber der Kontrollgruppe eine beachtliche Wachstumsverlangsamung und Gewebenekrose auf.
Außerdem schlug die Therapie auch bei Mäusen mit humanen Ovarialkarzinom-Xenografts an.

Was ist DOS47?
DOS47 ist ein neuer Medikamentenkandidat, der zur Zeit von Helix entwickelt wird und einen neuartigen Ansatz zur
Zerstörung von Krebszellen bildet. Dieser neuartige Ansatz beruht auf der Theorie von Helix, dass der Harnstoffzyklus,
ein natürlicher Ablauf im Körper, zur Bekämpfung von Krebs modifiziert werden kann.
Harnstoff ist als natürliches Abfallprodukt im ganzen Körper zu finden. Der Hauptzweck des Harnstoffzyklus’ besteht darin, den Körper von Ammoniak, welches Stoffwechselnebenprodukt insbesondere beim Abbau von Proteinen in den Zellen entstehen, zu entfrachten. Harnstoff wird im Rahmen eines Harnstoffzyklus genannten Stoffwechselprozess durch die Verbindung von Ammoniak und Kohlendioxid in der Leber erzeugt.
DOS47 basiert auf einem natürlich vorkommenden Enzym, das Urease heißt. Es wird angenommen, dass Urease den
Harnstoffzyklus im Prinzip umkehrt, indem es den Harnstoff in Ammoniak und Kohlendioxid aufspaltet. Das neuartige
DOS47 Therapiekonzept wurde entwickelt, um den Harnstoffzyklus lokal an der Stelle der Krebsgeschwulste im Körper umzukehren. Es wird angenommen, dass die lokale Produktion von Ammoniak und Kohlendioxid eine Kettenreaktion auslöst, die die aktiven Stoffwechselmechanismen von Krebszellen durch ein Zusammenspiel von Auswirkungen unterdrückt. Im Rahmen dieser theoretischen Reaktionen wird weiter vermutet, dass die lokale Produktion von Ammoniak am Ort der Krebsgeschwulste die Krebszellen mittels eines chemischen Vorgangs behindert. Außerdem glaubt man, dass durch die örtliche Reaktion der pH-Wert des die Krebszellen umgebenden Mikroumfeldes erhöht wird, wodurch die extra
zellularen säurereichen Verhältnisse, in denen die meisten Krebszellen existieren und sich vermehren, effektiv umgekehrt werden.

Nächste Schritte
Über die nächsten 12 bis 18 Monate beabsichtigt Helix, Methoden zur Konjugation von Targeting-Mitteln und analytische Ressourcen zu entwickeln, Studien zur akuten Toxikologie zu realisieren und eine Reihe von Konzeptnachweisstudien an Tieren unter Einsatz bestimmter DOS47-Konjugate durchzuführen.

Ein zentraler Punkt im DOS47-Programm von Helix wird darin bestehen, Konjugationsmethoden zu erarbeiten, um DOS47 mit einem biologischen Targeting-Mittel wie z.B. Antikörpern zu verbinden. Durch diese Entwicklungsarbeit soll es ermöglicht werden, DOS47-Therapeutika so zu gestalten, dass theoretisch unterschiedliche Krebsarten im Körper

einzusetzen, sowie auch direkt per intratumoraler Injektion bei soliden Tumoren. Um diese Arbeit zu ermöglichen, hat Helix seine Onkologielaboranlagen in Edmonton, Alberta, Kanada weiter ausgebaut und zusätzliche wissenschaftliche Mitarbeiter eingestellt.
