zunägst einmal möchte ich sagen, daß dieser thread weder zur verbreitung von pusher noch von basher geöffnet worden ist. hier sollen ausschließlich fakten oder ganz klar fundierte meinungen, theorien oder chartanalysen veröffentlicht werden.
wer meint er oder sie könnte hier (wie schon so oft woanders)seiner miesen laune durch absichtlicher provokation luft machen, ist hier nicht gern gesehen !!!
aber soviel nur dazu, ich denke wir sind ja alle erwachsen...
in der neuen ausgabe von "der aktionär" (ja, ich lese ihn immer noch...), nr.10/2002 27.02., geht es im titelthema wieder mal um "biotechnologie: comeback der giganten", wobei VRTX u.a. (wieder mal) empfohlen wird:
kurs: 25,90 €
ziel: 50 €
stopp: 17 €
im prinzip schreibt schweinebacke förtsch nix neues, nach wie vor soll sich das investment langfristig lohnen...
meiner meinung nach ist dies aus den schon oft erwähnten gründen (volle pipeline...usw.) richtig, ich versuche aber durch gelegentliches verkaufen nach extremen anstiegen, wie etwa vor ca. 2 wochen (22% in drei tagen) meine stückzahl zu vermehren...volatil genug ist der verlauf der aktie... wie auch bei vielen bios !!!

6,1 % im plus, nachbörslich leicht um 0,4 % wieder runter...
auffällig im guten sinne ist es, daß die umsätze an tagen wie gestern, wenn es nach oben geht, deutlich höher sind, als an schlechten...ich rede dabei von der nasdaq, denn dort wird der kurs gemacht !!!
der NBI bei 764,9 pkt (plus ca. 2%)...ich halte zwar nicht viel von charts, die 800 pkt. kommen aber langsam näher...

Habe Vrtx damals mit der ersten Musterdepotaufnahme im Aktionär gekauft. Nach ersten guten Gewinnen habe ich sie später frustriert und mit Verlust verkauft.
Meiner Meinung nach ist die Aktie irgendwie tot. Ok., gestern gings mal wieder rauf, aber irgendwie scheint mir Vrtx nie so richtig zu explodieren. Bei einigen großen Biotechfonds ist Vrtx zwar noch in den Topholdings, aber dieser Flop mit dem einen Produkt hat wohl einen gewissen Vertrauensverlust hinterlassen. Von der Produktpipeline siehts zwar sehr gut aus, aber irgendwie habe ich nicht das Gefühl das Vrtx in den nächsten Monaten den Biotechbereich outperformt.
Ich habe mich inzwischen auf Medx, Ctic und Pdli konzentriert.

Für den Optionsschein der Citibank (WKN: 582437)sollte daher ein Limit von 0,40 Euro platziert werden. Die nächsten Widerstandslinie - und damit potenzielle Kursziele - sind der Bereich um 35 und 46 US-Dollar. Für den Warrant würde dies zu einem Kursanstieg von bis zu 350 Prozent führen. Aus fundamentaler Sicht dürfte die Aktie ebenfalls vor einer Höherbewertung stehen: Vertex verfügt über eine ganze Reihe vielversprechender Produktkandidaten. Beispielsweise hat man bereits Ende 2000 für das HIV-Medikament Agenerase, das in Zusammenarbeit mit GlaxoSmithKline entwickelt worden sei, die Zulassung bekommen. Von diesem Präparat erwartet Vertex ein Marktpotenzial von bis zu 500 Millionen US-Dollar. Elf weitere Wirkstoffe gegen Brustkrebs oder Hepatitis C befindet sich derzeit in unterschiedlichen Phasen der präklinischen und klinischen Entwicklung. Für ein Präparat zur Bekämpfung des HI-Virus, das sich in der Phase III der klinischen Studien befinde, erwarte man die Zulassung für das Jahr 2003. Zur Verlustbegrenzung sollte für die Gesamtposition ein Stop bei 20,65 US-Dollar berücksichtigt werden.

guten morgen
eine gute woche liegt hinter VRTX, der abwärtstrend ist deutlich gebrochen, jetzt gilt es die 200 tage linie zu überwinden, wobei das alles etwas schnell passiert...
die umsätze sind auch schön hoch, jedoch denke ich, daß eine kleine korrektur fällig wird, noch einmal 10 - 15 % runter, dann kann es losgehen...

eine gute woche liegt hinter VRTX, der abwärtstrend ist deutlich gebrochen, jetzt gilt es die 200 tage linie zu überwinden, wobei das alles etwas schnell passiert...
die umsätze sind auch schön hoch, jedoch denke ich, daß eine kleine korrektur fällig wird, noch einmal 10 - 15 % runter, dann kann es losgehen...

Noch im Januar 2001 habe die Biotech-Branche eine Menge neuer Produktgenehmigungen für das Jahr erwartet, wodurch sich sowohl Umsatzwachstum als auch eine positive Verfassung der Branche ergeben sollten. Es seien 25 Produktgenehmigungen erwartet worden, von denen die Aufsichtsbehörde FDA in den Vereinigten Staaten von Amerika dann tatsächlich 18 erteilt habe. Dies berichten die Anlageexperten der DWS in einer aktuellen Brancheneinschätzung.
Obwohl dies für die Branche wie eine durchaus vernünftige Erfolgsquote aussehen würde (72 Prozent Genehmigung gegenüber einem historischen Durchschnittswert von 80 Prozent für P3-Produkte), sei die Marktverfassung dadurch belastet worden, dass zahlreiche herausragende Produkte zurückgestellt worden seien.
Obwohl die Biotech-Branche unter diesen Rückschlägen gelitten habe, hätte es trotzdem einige positive Einsichten zu verzeichnen gegeben. Zunächst bestehe nach Ansicht der DWS-Experten ein sehr großer Unterschied zwischen der Ablehnung eines Produkts und einer Zurückstellung. Falle ein Produkt bei den klinischen Tests durch, dann gehe der Wert dieses Produkts auf Null. Würde aber ein Produkt zurückgestellt, würde dies bedeuten, dass durch einige Nachbesserungen das Produkt immer noch eine vernünftige Chance besitze, irgendwann in der Zukunft auf den Markt zu kommen.
Die Mehrzahl der negativen Nachrichten im Jahr 2001 hätten sich nach Angaben der Biotech-Experten der DWS auf Zurückstellungen und nicht auf Ablehnungen eines Produkts bezogen. Alles in allem verfüge die Biotech-Branche nach Einschätzung der DWS-Analysten über ein solides Finanzpolster. Die Produkt-Pipeline der Branche sei voll und reife heran. Vor allem erzeugten neue Produkte neue Umsätze und neue Gewinne. Dieser Punkt bleibe ihres Erachtens das Hauptkriterium einer Investition in Biotech-Aktien.

gestern wurde erst mal kasse gemacht, jedoch haben sie nicht auf tagestief geschlossen, das lag nochmal fast 1 $ unter dem schlußkurs
die umsätze waren allerdings mit 1,6 mio mehr als doppelt so hoch wie am vortag, an dem noch ein leichter gewinn zu verbuchen war....
...ich denke, da ist noch ein bischen platz nach unten, bis ca. auf das alte zwischen-hoch bei etwa 24 $, dann heißt es meiner meinung nach aber "kräftig nachkaufen" und auf zu neuen höhen !!!!

das find ich hochinteressant, langsam scheint VRTX wieder in den blickpunkt vieler zu gelangen, die beiträge in den verschiedenen threads nehmen wieder zu, jetzt bin ich nur gespannt, ob der wert endlich mal ausbricht, oder ob er wieder zurück kommt...
ich habe vor einigen tagen erst mal kasse gemacht, zwar nicht auf dem höchstpunkt, aber das klappt eh nie !!!
ich fahre nun eine noch viel riskantere schiene, bin verstärkt in STEM rein, ich hoffe dort mal schnell ein paar prozente zu machen, um dann in VRTX wieder mit höherer stückzahl einzusteigen...?!?!

als ich angefangen habe, mich mit aktien zu beschäftigen, bin ich über einen bekannten an den "aktionär" geraten, das war die zeit, als jeder mit jeder empfehlung ins schwarze getroffen hätte...
zu dieser zeit fand ich die förtsch-gang sehr gut, mit der zeit stellte ich dann fest, die labern genau so viel müll wie jeder andere, der empfehlungen gibt, auch !!!
was er jedoch geschaft hat, ist mein interesse für biotechs zu endecken...
aus dieser zeit habe ich noch ein abo, das ich dummerweise versäumt habe nach dem ersten jahr zu kündigen...die kündigung ist jetzt aber raus...die hefte, die noch bekomme, lese ich natürlich auch, zumindest das, was mich interessiert....
soviel dazu....
...ich denke vor ostern passiert nicht mehr viel, wenn nicht gerede wahnsinns news kommen, ziehe ich mich bis dienstag zurück, kleiner entspannungsurlaub mit reichlich bier...allen ein paar schöne tage und dicke eier.....

Getting Serious about Stopping STDs

In the public mind, AIDS looms as by far the most frightening sexually transmitted disease [STD]. But the biotech industry is also scrambling to find drugs and vaccines to fight a host of less-lethal but more prevalent STDs -- from the closely related B and C strains of the hepatitis virus and the two forms of herpes to the most common of all, human papilloma virus, or HPV.

The human toll taken by these diseases is staggering. According to the U.S. Centers for Disease Control & Prevention [CDC] in Atlanta, 1 in 20 people will be exposed to hepatitis B at some point in their lives -- even though a vaccine is available today that can protect against it. An estimated 200,000 people in the U.S. are infected with the disease each year, and as many as 25% of them are eventually diagnosed with liver disease or liver failure.
Hepatitis C is even more alarming because there`s no vaccine to prevent this newer strain. And it`s a chronic infection that remains with its victims throughout their lives, destroying their liver and in many cases necessitating a transplant. About 40,000 new cases are reported in the U.S. annually -- roughly equal to the number of new HIV infections each year.
URGENT NEED. Given the scope of the threat, several companies -- including such powerhouses as Merck (NYSE:MRK-news) and GlaxoSmithKline (NYSE:GSK-news), as well as small biotech players such as Gilead Sciences (NasdaqNM:GILD-news) and Vertex Pharmaceuticals (NasdaqNM:VRTX-news) -- are now locked in a race to be the first to market a vaccine.
The potential payoff is huge. Analysts estimate that a vaccine for a common STD could rake in more than $1 billion a year in sales in the U.S. alone. For example, if an effective vaccine against genital herpes could be found and gain approval from the federal Food & Drug Administration, it could well end up being recommended by doctors as a preventative measure for all teenagers in the U.S.
The need is urgent: The CDC estimates that 1 in 5 adolescents and adults nationwide are infected with genital herpes. While the disease may cause no symptoms in many cases, it can be an embarrassing and extremely painful lifelong affliction. The CDC figures that exposure to HPV is even higher, with the majority of sexually active people exposed to one or more of the dozens of strains of HPV at some point in their lives.
WOMAN-KILLER. While an HPV infection almost never poses a danger to men, it`s the precursor to nearly all cases of cervical cancer in the U.S., which is often fatal and is diagnosed in more than 15,000 American women annually.
Merck is usually thought of as the leader in treating sexual diseases because of its highly effective AIDS cocktail Crixivan. It now has worldwide clinical trials on an experimental AIDS vaccine under way. But Merck also may be sitting on a powerful vaccine for HPV. The drug was developed for the prevention of genital warts [a common symptom of the virus] and cervical cancer, and is now in late-stage testing. Comprising virus-like particles derived from yeast containing no genetic material, the vaccine is a coat of viral proteins meant to foster an immune response.
Merck won`t know for sure how well the vaccine works for three or four years, but the early signs are promising, says Dr. Kathrin Janssen, director of Merck`s HPV vaccine program. She believes that the vaccine, if effective, would be an ideal preventative measure for every young woman. ``We feel that it`s a very important health concern and that you have to start early in educating adolescents about the risks -- before they cut loose, so to speak.``
MODESTLY EFFECTIVE. Glaxo could turn out to be Merck`s main rival. It has an HPV vaccine that`s roughly a year behind Merck`s and that takes a different scientific approach. Glaxo`s vaccine uses virus-like particles derived from an insect and an additional catalyst to help the immune system recognize the virus and attack it, says Dr. Moncef Slaoui, a senior vice-president for new products at Glaxo. At this point, it`s too early to tell which of the two vaccines might prove most effective.
Glaxo, however, is also researching a broad range of treatments against STDs. The British drugmaker already is selling Valtrex, a modestly effective treatment for hepatitis B. More important, Glaxo is testing a vaccine for genital herpes. In trials already completed, it helped protect women against infection but was ineffective in men. As a result, researchers have altered the dosing and trial design in an effort to see if it can protect men, too.
``We feel that this could be a very important,`` Slaoui says. ``From a strict medical standpoint, there are other [more deadly] STDs for which vaccine development is more critical. But psychologically, genital herpes can have a profound impact on people who develop its symptoms.``
MILDER SIDE EFFECTS. Medically speaking, the most pressing need is to stop the spread of hepatitis. With few treatment options available, some patients have no choice but to wait for a liver transplant. Within months, however, the virus can begin to infect the transplanted liver, putting patients in danger of liver failure again. Adds Shlomo Dagan, chief scientific officer for XTL Biopharmaceuticals, an Israeli company that`s developing new antibody therapies for hepatitis B and C: ``Many people are not aware that they carry the disease. And in some carriers the symptoms may not appear for 10 to 15 years.``
Vertex Pharmaceuticals may be the furthest along in hepatitis C research. Doctors now rely on powerful immunotherapy such as alpha interferon for treating the disease, but that produces almost unbearable side effects, leaving many patients too weak to go to work. Getting Serious about Stopping STDs

In the public mind, AIDS looms as by far the most frightening sexually transmitted disease [STD]. But the biotech industry is also scrambling to find drugs and vaccines to fight a host of less-lethal but more prevalent STDs -- from the closely related B and C strains of the hepatitis virus and the two forms of herpes to the most common of all, human papilloma virus, or HPV.

The human toll taken by these diseases is staggering. According to the U.S. Centers for Disease Control & Prevention [CDC] in Atlanta, 1 in 20 people will be exposed to hepatitis B at some point in their lives -- even though a vaccine is available today that can protect against it. An estimated 200,000 people in the U.S. are infected with the disease each year, and as many as 25% of them are eventually diagnosed with liver disease or liver failure.
Hepatitis C is even more alarming because there`s no vaccine to prevent this newer strain. And it`s a chronic infection that remains with its victims throughout their lives, destroying their liver and in many cases necessitating a transplant. About 40,000 new cases are reported in the U.S. annually -- roughly equal to the number of new HIV infections each year.
URGENT NEED. Given the scope of the threat, several companies -- including such powerhouses as Merck (NYSE:MRK-news) and GlaxoSmithKline (NYSE:GSK-news), as well as small biotech players such as Gilead Sciences (NasdaqNM:GILD-news) and Vertex Pharmaceuticals (NasdaqNM:VRTX-news) -- are now locked in a race to be the first to market a vaccine.
The potential payoff is huge. Analysts estimate that a vaccine for a common STD could rake in more than $1 billion a year in sales in the U.S. alone. For example, if an effective vaccine against genital herpes could be found and gain approval from the federal Food & Drug Administration, it could well end up being recommended by doctors as a preventative measure for all teenagers in the U.S.
The need is urgent: The CDC estimates that 1 in 5 adolescents and adults nationwide are infected with genital herpes. While the disease may cause no symptoms in many cases, it can be an embarrassing and extremely painful lifelong affliction. The CDC figures that exposure to HPV is even higher, with the majority of sexually active people exposed to one or more of the dozens of strains of HPV at some point in their lives.
WOMAN-KILLER. While an HPV infection almost never poses a danger to men, it`s the precursor to nearly all cases of cervical cancer in the U.S., which is often fatal and is diagnosed in more than 15,000 American women annually.
Merck is usually thought of as the leader in treating sexual diseases because of its highly effective AIDS cocktail Crixivan. It now has worldwide clinical trials on an experimental AIDS vaccine under way. But Merck also may be sitting on a powerful vaccine for HPV. The drug was developed for the prevention of genital warts [a common symptom of the virus] and cervical cancer, and is now in late-stage testing. Comprising virus-like particles derived from yeast containing no genetic material, the vaccine is a coat of viral proteins meant to foster an immune response.
Merck won`t know for sure how well the vaccine works for three or four years, but the early signs are promising, says Dr. Kathrin Janssen, director of Merck`s HPV vaccine program. She believes that the vaccine, if effective, would be an ideal preventative measure for every young woman. ``We feel that it`s a very important health concern and that you have to start early in educating adolescents about the risks -- before they cut loose, so to speak.``
MODESTLY EFFECTIVE. Glaxo could turn out to be Merck`s main rival. It has an HPV vaccine that`s roughly a year behind Merck`s and that takes a different scientific approach. Glaxo`s vaccine uses virus-like particles derived from an insect and an additional catalyst to help the immune system recognize the virus and attack it, says Dr. Moncef Slaoui, a senior vice-president for new products at Glaxo. At this point, it`s too early to tell which of the two vaccines might prove most effective.
Glaxo, however, is also researching a broad range of treatments against STDs. The British drugmaker already is selling Valtrex, a modestly effective treatment for hepatitis B. More important, Glaxo is testing a vaccine for genital herpes. In trials already completed, it helped protect women against infection but was ineffective in men. As a result, researchers have altered the dosing and trial design in an effort to see if it can protect men, too.
``We feel that this could be a very important,`` Slaoui says. ``From a strict medical standpoint, there are other [more deadly] STDs for which vaccine development is more critical. But psychologically, genital herpes can have a profound impact on people who develop its symptoms.``
MILDER SIDE EFFECTS. Medically speaking, the most pressing need is to stop the spread of hepatitis. With few treatment options available, some patients have no choice but to wait for a liver transplant. Within months, however, the virus can begin to infect the transplanted liver, putting patients in danger of liver failure again. Adds Shlomo Dagan, chief scientific officer for XTL Biopharmaceuticals, an Israeli company that`s developing new antibody therapies for hepatitis B and C: ``Many people are not aware that they carry the disease. And in some carriers the symptoms may not appear for 10 to 15 years.``
Vertex Pharmaceuticals may be the furthest along in hepatitis C research. Doctors now rely on powerful immunotherapy such as alpha interferon for treating the disease, but that produces almost unbearable side effects, leaving many patients too weak to go to work. Aventis Pharma and Vertex Pharmaceuticals to Expand Development of Pralnacasan, Oral Cytokine Inhibitor for Inflammatory Diseases Clinical Proof-of-Mechanism and Good Tolerability for 24 Weeks Indicated in Phase II Rheumatoid Arthritis Clinical Trial
BRIDGEWATER, N.J., and CAMBRIDGE, Mass., Apr 24, 2002 /PRNewswire-FirstCall via
COMTEX/ -- Aventis Pharma (NYSE: AVE) and Vertex Pharmaceuticals Incorporated
(Nasdaq: VRTX) today announced that they are expanding clinical development of
the interleukin-1 beta converting enzyme inhibitor pralnacasan, based on
clinical proof-of-mechanism data obtained in a recently-completed Phase II
clinical trial in rheumatoid arthritis (RA). Pralnacasan is an oral, small
molecule drug candidate licensed for development by Aventis from Vertex. In this
Phase II clinical trial, proof-of-mechanism for pralnacasan as a novel
anti-inflammatory drug candidate was obtained based on an assessment of the
compound`s clinical activity and tolerability. With this result, pralnacasan has
met Aventis` criteria for expanded clinical development.
The data from the Phase II clinical trial in RA supports further clinical
development of pralnacasan in RA, as well as initial Phase II clinical trials in
osteoarthritis and other potential indications. Aventis will evaluate the timing
of initiating such trials.
"These results demonstrate that pralnacasan, an oral cytokine inhibitor, has
anti-inflammatory effects in rheumatoid arthritis," said Frank Douglas,
executive vice president and head of Drug Innovation & Approval at Aventis. "We
and Vertex look forward to completing our analysis of this trial and designing
additional rheumatoid arthritis studies. At the same time, we will consider
initiating clinical studies in osteoarthritis and other indications, if
appropriate. This will allow us to define fully the clinical applications and
market opportunity for pralnacasan."
Pralnacasan is an orally administered inhibitor of interleukin-1 beta converting
enzyme (ICE), an important enzyme regulating inflammatory processes. ICE
regulates the production of interleukin-1 beta (IL-1 beta) and IL-18, key
proinflammatory cytokines that initiate and sustain the progression of
inflammation. Inhibiting ICE may be a useful strategy for curtailing damaging
inflammatory processes common to a number of acute and chronic conditions.
Study Design
The Phase II clinical trial was designed to evaluate the safety, efficacy and
pharmacokinetics of pralnacasan alone and in combination with other treatments
in patients with active RA. The clinical trial was conducted in Europe and
enrolled adult patients diagnosed with mild to moderate RA at least 6 months
prior to the start of the study. Patients in the study were able to continue
their treatment with anti-inflammatory therapies. Upon study entry, patients
were required to have received either: 1) a fixed dosage regimen of RA therapies
that included one or more disease-modifying anti-rheumatic drugs (DMARDs) and/or
corticosteroids for at least three months, or 2) no additional RA drug therapy.
The clinical trial was a 12-week, double blind, randomized, placebo-controlled
study. After the initial 12 weeks of the trial, patients had the option of
extending treatment for an additional 12 weeks. In the study, patients were
randomized to receive one of two doses of pralnacasan or placebo, in addition to
any other RA therapies they were receiving. There were 285 adult patients
enrolled in the study, of which 224 patients in the pralnacasan dose groups and
55 in the control group were available for evaluation. Approximately 80% of
patients were taking concomitant DMARDs and approximately 55% were taking
concomitant corticosteroids at study entry.
"This trial was designed to collect information about the activity of
pralnacasan alone and in combination with other RA treatments in a diverse
patient population," commented Dr. Douglas. "We believe that the study design
and the data will facilitate the design of a focused development plan."
Study Results
A broad range of clinical and pharmacodynamic measures was used to assess
pralnacasan`s anti-inflammatory activity. Results showed that patients receiving
pralnacasan had a strong trend toward a dose-dependent improvement in signs and
symptoms of disease, as measured by ACR20 response rates after 12 weeks
(p=0.076). Patients are considered to have an ACR20 response if they achieve at
least a 20% improvement in their tender-joint and swollen-joint counts in
addition to 20% or more improvement in a minimum of three additional criteria
set by the American College of Rheumatology (ACR). Analyses of patient
sub-groups not receiving concomitant DMARDs, methotrexate or corticosteriods
showed similar strong trends indicative of pralnacasan`s anti- inflammatory
effect. Patients receiving the high dose of pralnacasan in the
intention-to-treat population exhibited ACR20 response rates greater than 40%.
In addition, the high dose of pralnacasan induced significant decreases in the
inflammatory biomarkers C-reactive protein (CRP), erythrocyte sedimentation rate
(ESR), and serum amyloid A (SAA).
In the study, pralnacasan demonstrated good tolerability alone and in
combination with other RA therapies when administered for up to 24 weeks.
Based on the demonstrated anti-inflammatory effects, Aventis and Vertex conclude
that clinical proof-of-mechanism for pralnacasan as a novel, first- in-class
therapy for inflammatory diseases has been obtained.

available inhibitors of ICE. Their intensive design efforts were based on the
three-dimensional molecular structure of ICE, solved by Vertex researchers in
1994. In 1998, pralnacasan became the first ICE inhibitor to enter clinical
development. In September 1999, Vertex and Aventis expanded their original
agreement. Under this agreement, Aventis funds development of pralnacasan and
will pay Vertex up to $62 million for the successful development of pralnacasan
in RA, the first targeted indication. Aventis has retained the rights to develop
pralnacasan for additional indications and Vertex will receive similar
milestones on these indications. Vertex has co- promotion rights in the United
States and Europe and will receive royalties on global sales, as well as a

the interleukin-1 beta converting enzyme inhibitor pralnacasan, based on
clinical proof-of-mechanism data obtained in a recently-completed Phase II
clinical trial in rheumatoid arthritis (RA). Pralnacasan is an oral, small
molecule drug candidate licensed for development by Aventis from Vertex. In this
Phase II clinical trial, proof-of-mechanism for pralnacasan as a novel
anti-inflammatory drug candidate was obtained based on an assessment of the
compound`s clinical activity and tolerability. With this result, pralnacasan has
met Aventis` criteria for expanded clinical development.
The data from the Phase II clinical trial in RA supports further clinical
development of pralnacasan in RA, as well as initial Phase II clinical trials in
osteoarthritis and other potential indications. Aventis will evaluate the timing
of initiating such trials.
"These results demonstrate that pralnacasan, an oral cytokine inhibitor, has
anti-inflammatory effects in rheumatoid arthritis," said Frank Douglas,
executive vice president and head of Drug Innovation & Approval at Aventis. "We
and Vertex look forward to completing our analysis of this trial and designing
additional rheumatoid arthritis studies. At the same time, we will consider
initiating clinical studies in osteoarthritis and other indications, if
appropriate. This will allow us to define fully the clinical applications and
market opportunity for pralnacasan."
Pralnacasan is an orally administered inhibitor of interleukin-1 beta converting
enzyme (ICE), an important enzyme regulating inflammatory processes. ICE
regulates the production of interleukin-1 beta (IL-1 beta) and IL-18, key
proinflammatory cytokines that initiate and sustain the progression of
inflammation. Inhibiting ICE may be a useful strategy for curtailing damaging
inflammatory processes common to a number of acute and chronic conditions.
Study Design
The Phase II clinical trial was designed to evaluate the safety, efficacy and
pharmacokinetics of pralnacasan alone and in combination with other treatments
in patients with active RA. The clinical trial was conducted in Europe and
enrolled adult patients diagnosed with mild to moderate RA at least 6 months
prior to the start of the study. Patients in the study were able to continue
their treatment with anti-inflammatory therapies. Upon study entry, patients
were required to have received either: 1) a fixed dosage regimen of RA therapies
that included one or more disease-modifying anti-rheumatic drugs (DMARDs) and/or
corticosteroids for at least three months, or 2) no additional RA drug therapy.
The clinical trial was a 12-week, double blind, randomized, placebo-controlled
study. After the initial 12 weeks of the trial, patients had the option of
extending treatment for an additional 12 weeks. In the study, patients were
randomized to receive one of two doses of pralnacasan or placebo, in addition to
any other RA therapies they were receiving. There were 285 adult patients
enrolled in the study, of which 224 patients in the pralnacasan dose groups and
55 in the control group were available for evaluation. Approximately 80% of
patients were taking concomitant DMARDs and approximately 55% were taking
concomitant corticosteroids at study entry.
"This trial was designed to collect information about the activity of
pralnacasan alone and in combination with other RA treatments in a diverse
patient population," commented Dr. Douglas. "We believe that the study design
and the data will facilitate the design of a focused development plan."
Study Results
A broad range of clinical and pharmacodynamic measures was used to assess
pralnacasan`s anti-inflammatory activity. Results showed that patients receiving
pralnacasan had a strong trend toward a dose-dependent improvement in signs and
symptoms of disease, as measured by ACR20 response rates after 12 weeks
(p=0.076). Patients are considered to have an ACR20 response if they achieve at
least a 20% improvement in their tender-joint and swollen-joint counts in
addition to 20% or more improvement in a minimum of three additional criteria
set by the American College of Rheumatology (ACR). Analyses of patient
sub-groups not receiving concomitant DMARDs, methotrexate or corticosteriods
showed similar strong trends indicative of pralnacasan`s anti- inflammatory
effect. Patients receiving the high dose of pralnacasan in the
intention-to-treat population exhibited ACR20 response rates greater than 40%.
In addition, the high dose of pralnacasan induced significant decreases in the
inflammatory biomarkers C-reactive protein (CRP), erythrocyte sedimentation rate
(ESR), and serum amyloid A (SAA).
In the study, pralnacasan demonstrated good tolerability alone and in
combination with other RA therapies when administered for up to 24 weeks.
Based on the demonstrated anti-inflammatory effects, Aventis and Vertex conclude
that clinical proof-of-mechanism for pralnacasan as a novel, first- in-class
therapy for inflammatory diseases has been obtained.
"We and Aventis have pioneered the scientific development of ICE inhibitors,
starting with innovative early drug design efforts. This research provided the
framework for demonstrating that oral ICE inhibitors can lead to clinical
benefit in inflammatory diseases like rheumatoid arthritis," said Vicki Sato,
Ph.D., President of Vertex. "Pralnacasan is an example of our focus on unmet
medical needs and providing the next generation of first-in- class therapeutics
for patients seeking safer, more effective treatments."
Final data analysis is continuing, and the companies anticipate that additional
data from this study will be provided during Aventis` R&D Day presentations in
June.
Vertex and Aventis began collaborating in 1993 to discover and develop orally
available inhibitors of ICE. Their intensive design efforts were based on the
three-dimensional molecular structure of ICE, solved by Vertex researchers in
1994. In 1998, pralnacasan became the first ICE inhibitor to enter clinical
development. In September 1999, Vertex and Aventis expanded their original
agreement. Under this agreement, Aventis funds development of pralnacasan and
will pay Vertex up to $62 million for the successful development of pralnacasan
in RA, the first targeted indication. Aventis has retained the rights to develop
pralnacasan for additional indications and Vertex will receive similar
milestones on these indications. Vertex has co- promotion rights in the United
States and Europe and will receive royalties on global sales, as well as a
co-promotion royalty and reimbursement of co- promotion costs.

For the three months ending March 31, 2002, the Company`s loss was $22.1
million, or $0.29 per share, compared to a loss of $11.0 million, or $0.15 per
share, in the first quarter of 2001. The 2001 first quarter loss is stated
before merger related costs and a non-recurring charge.
Total revenues for the three months ending March 31, 2002 were $40.7 million, as
compared to $35.0 million for the first quarter of 2001. Revenues from the
pharmaceutical business for the first quarter of 2002 were $20.6 million,
compared to $18.2 million for the same period last year. For the three months
ended March 31, 2002, discovery tools and services revenues were $20.1 million,
as compared to $16.8 million for the same period last year.
Cost of revenues for the three months ending March 31, 2002 were $8.6 million,
as compared to $10.5 million in the same period last year. This decrease
reflects the discovery tools and services business` focus on the higher margin
product revenues.
Research and development expenses for the three months ending March 31, 2002
were $47.0 million, compared to $32.5 million for the first quarter of 2001. The
increase in research and development expenses in the first quarter of 2002
reflects the Company`s commitment to advancing its broad clinical pipeline of
more than 12 drug candidates targeting major therapeutic areas and continuing to
invest in its productive gene family drug discovery programs.
Sales, general and administrative expenses for the three months ending March 31,

Die Ankündigung von Partner Merck, der europäische Zulassungsantrag für Erbitux würde sich noch verzögern, führte bei Imclone zu einem Einbruch des Aktienkurses um 12%. Im Rahmen der Erwartungen fielen zwar die Q1 Zahlen von Biogen aus (47 cents pro Aktie), das Unternehmen sprach aber gleichzeitig eine Gewinnwarnung für Q2 und das laufende Geschäftsjahr aus. das war natürlich ein schock, nach der ersten "pleite" im vergangenen jahr nun wieder nicht grade ein erfolg. ich denke diesmal leidet das grundsätzliche vertrauen in VRTX.
im vorbörslichen handel gestern hatte die aktie schon über 10% verloren, noch bevor nachrichten zu lesen waren.
was soll man jetzt machen ? nun ja, wie ich in beitrag #12 schon geschrieben habe bin ich vor einigen wochen schon bei 29,50 € ausgestiegen, was sich im nachhinein als glückliche aktion ´rausstellt. das war zwar alles andere als der hochpunkt, aber den trifft man eh nie. wenn ich mir den verlauf nach den letzten schlechten nachrichten ansehe und hier parallelen ziehe, könnte es noch weiter runter gehen, fragt mich bitte nicht wie weit, in dem fall würde ich jetzt die reißleine ziehen und evt. später wieder rein.

ich lese ihr Heft nun im dritten Jahr, seit zwei Jahren als Abo (nun allerdings gekündigt), anfangs, als ich noch überhaupt keine Ahnung von der Börse hatte, war ich von Ihren Empfehlungen äußerst beeindruckt. Leider muß ich seit einigen Monaten feststellen, daß Ihre Tipps mit zunehmenderer Sicherheit immer mehr daneben treffen, dies mittlerweile aber sehr regelmäßig. Das ist nicht weiter schlimm, denn das ist auch ein Tipp, denn wenn Sie eine Aktie in den Himmel loben, muß man nichts anderes tun, als diesen Wert zu verkaufen, um nicht völligen Schiffsbruch zu erleiden. Das beste Beispiel dafür ist die Aktie von Vertex; natürlich konnten Sie diese Entwicklung nicht vorhersehen, es ist allerdings erstaunlich, wie oft diese ZSituation eintritt.
Da ich nicht davon ausgehe, daß dieses Schreiben in Ihrem Heft veröffentlicht wird, bitte ich um Kenntnisnahme der Gedanken eines enttäuschten Lesers.

announced that they will collaborate to discover novel drug targets that may be
implicated in inflammation and other diseases.
Under the terms of the agreement, the parties will utilize Immusol`s Inverse
Genomics(TM) technology and technology developed at Vertex`s Aurora Biosciences
subsidiary to inactivate novel genes that prevent the cellular response to
certain important inflammatory cytokines and growth factors. The collaboration
will make use of Aurora`s beta-lactamase technology, and in particular two of
Aurora`s CellSensor(TM) cell lines. As part of the agreement, Vertex will make
an undisclosed payment to Immusol.
The goal of the collaboration will be to evaluate whether the combination of the
two companies` technologies can identify therapeutically-important protein
targets implicated in various inflammation pathways. Information gained through

ich habe, glaube ich, den fehler gemacht und mich in diesen wert zu sehr verbissen, wobei ich nach wie vor denke, daß der wert lang-lang-langfristig gut ist (wuscher, skaya, und alle anderen......durchhalten!!!!!); leider läuft es aber meistens anders als man denkt, vor allem die letzten 2 1/2 verf...ten jahre !!!!
ich fahre nun volles risiko, zu verfolgen im thread "<<< STEM >>>"
