Nachdem die Vorbörse bereits 15,25$ gesehen hatte
verlor IR nach insgesamt lustlosen Verlauf, mit
deutlich geringerem Umsatz (1.187,000), 5,7%, auf 13,45$.
Tief war heute 13,25$. Das Verhältnis Sell/Buy
lag bei 10:1. Dies entäuscht und verwundert zugleich.
Diese Konstellation hätte dramatischere Auswirkungen
haben müssen.
Desweiteren ist ein Stimmungsumschwung zu beobachten.
Ungeachtet, neuer positiver Nachrichten, überwiegt an den Boards die negative Einstellung zu IR.
Dies kann sehr wohl auch mit der z.Zt. in Boston
stattfindenden Biotechmesse zu tun haben. Vielleicht
will man abwarten.

Man kann dadurch deutlich machen was die Meldung vom 23.03.00
bedeutet hat, den bei der nachfolgenden Studie wurde kein Durchbruch
erzielt.

Endlich ist die Auswertung der großen Remune-Studie präsentiert worden. Leider brachte sie überhaupt nicht die von vielen
erhofften Ergebnisse. Dr. James Kahn fiel die undankbare Aufgabe zu, diese an 2527 Personen durchgeführte
placebokontrollierte, doppelblinde, randomisierte Phase III-Studie zu präsentieren. Die mediane CD4-Zellzahl der
Studienteilnehmer lag bei Studienbeginn bei 418 Zellen/µl, die mediane Ausgangsviruslast bei 3,1 log. Primärer Endpunkt der
Studie war progressionsfreie Überlebenszeit, sekundäre Endpunkte waren Surrogatmarker. Die Verum-Gruppe erhielt (auf dem
Hintergrund einer stabilen HAART) alle 12 Wochen 10 Mio IU Remune, die Placebo-Gruppe (auf dem Hintergrund einer stabilen
HAART) IFA (das Adjuvant alleine). Der durchschnittliche Beobachtungszeitraum betrug 88,7 Wochen. Die Drop-out-Rate lag
bei 8% pro Jahr.
90 Patienten verschlechterten sich klinisch: 44 in der Verum-Gruppe und 46 in der Placebo-Gruppe (p=0,75). Die
Progressionsrate betrug 2,1 Ereignisse pro 100 Personenjahre des Follow-up. Da die Rate zu gering war und keinen
Unterschied zwischen den Studienarmen zeigte, empfahl das DSMB, die Studie zu beenden.
Die vorläufige Auswertung der VL-Ergebnisse von 1567 der 2527 Patienten ergab keine statistisch signifikanten Unterschiede
zwischen den beiden Armen. Zum Auswertungszeitpunkt Woche 48 zeigte sich eine leichte Überlegenheit der Verum-Gruppe
bezüglich der CD4-Zellzahl (+16 Zellen/µl im Vergleich zur Placebo-Gruppe, p<0,01). Erwartungsgemäß lag die
Lympozytenproliferation auf die Core-Proteine bei der Verum-Gruppe mit 45% bzw. 34% deutlich über der der Placebo-Gruppe
(< 1%) [Kahn J. LB-21, Addendum, S. 18].

Die zellvermittelte Immunantwort auf HIV ist - so wird angenommen - der wichtigste Mechanismus der Kontrolle des HIV
bei
Menschen, die in der Lage sind, die Virusvermehrung ohne ART zu kontrollieren - den sogenannten
Long-Term-Nonprogressoren. Diese Immunantwort ist durch eine starke Helferzellproliferation gegen das HIV-Kernprotein
p24
charakterisiert.
Eine erfolgreiche HAART reduziert die Virusreplikation auf sehr geringe Level und erlaubt eine teilweise Rekonstitution des
Immunsystems - selbst bei Menschen mit weit fortgeschrittenem Immundefekt. Es gibt allerdings bislang nur sehr wenig
Hinweise darauf, daß es - selbst nach langer erfolgreicher HAART - ebenfalls zu einer Rekonstitution der HIV-spezifischen
Immunantwort kommt. Bislang konnte eigentlich eher das Gegenteil nachgewiesen werden: Bei Menschen, bei denen vor
Beginn der Therapie eine HIV-spezifsiche Immunantwort nachweisbar war, verschlechterte sich diese unter Therapie und
schien
bei länger andauernder erfolgreicher Unterdrückung der Virusreplikation vollständig zu verschwinden.
Dieses Verschwinden der Immunantwort auf HIV ist eigentlich nicht überraschend, da die Aufrechterhaltung einer
zellulären
Immunantwort eine andauernde Stimulation in Form der Antigenpräsentation erfordert. Da durch HAART der Virusumsatz
auf
ein sehr niedriges Niveau reduziert wird, können HIV-Antigene nicht mehr präsentiert werden und die spezifische
Immunantwort
verschwindet. Tatsächlich wird die Reduktion aktivierter Lymphozyten als ein Marker für eine erfolgreiche ART gesehen.
Das paradoxe dieser Situation ist, daß eine lang andauernde und ununterbrochene Stimulation durch Antigen, die mit
einer
unkontrollierten HIV-Replikation einhergeht, eben keine wirksame HIV-spezifische Immunantwort hervorruft, sondern
übereinstimmend als die Ursache für deren Versagen gewertet wird. Möglicherweise ist die Hyperaktivierung des
Immunsystems oder die Zerstörung von Helferzellen, die die Antwort einleiten, die Ursache dafür. Denn aktivierte
T-Lymphozyten sind die primären Zielzellen des HIV.
Es wurde überlegt, daß eine suffiziente Stimulierung der HIV-spezifischen zellvermittelten Immunantwort durch eine
Immunisierung mit einer modifizierten Form des HIV (bei Supression der nativen HIV-Infektion mittels HAART) die mit der
chronischen Infektion einhergehende Anergie beheben könnte. Der Hauptkandidat für einen derartigen Versuch ist
Remune, das
von Jonathan Salks und der Immune Response Corporation entwickelt wurde und von Agouron zur Marktreife
weiterentwickelt
werden soll.
Remune ist ein abgetötetes HIV-1 mit einem Stamm A-Enve-lope und einem Stamm G-gag. Die äußeren Hüllproteine
(gp120)
sind depletiert. Die Antigenpräparation ist chemisch und durch Bestrahlung inaktiviert.
In einer Veröffentlichung im Wissenschaftsmagazin Journal of Infectious Diseases berichten Ronald B Moss und Mark
Wal-lace über die Ergebnisse einer kleinen Studie, die die Fähigkeit des Immunsystems nach Behandlung mit Remune in
Kombination mit HAART beurteilen sollte. Die HIV-1-spezifsiche Lymphozytenproliferation wurde bei 15 Patienten vor und
nach
Behandlung untersucht. Die Studienteilnehmer hatten median einen CD4-Zellzahl von 586 Zellen/µl (Bandbreite 223 - 1147)
und
eine Plasmaviruslast von 953 Kopien/ml (Bandbreite <400 - 4625). Eine Zweifach-Nukleosidkombination wurde mit einem
oder
zwei PIs kombiniert. Remune wurde am 1.Tag und dann alle 12 Wochen in der Dosis von 10 IU verabreicht.
Die in vitro Immunfunktionsassays verwendeten das komplette gp120 depletierte Immunogen, gereinigtes p24-Antigen der
Remune-Vakzine (np24), ein rekombinantes p24 (rp24) und ein Candida-Antigen als Kontrolle. Das Erkennen der Antigene
wurde vor und nach der Immunisierung durch Standard-Lymphozytenproliferationsassay (LPA) und Flow-Zytometrie
bewertet.
Mit CD4, CD8 und CD56 (NK-Zellen) depletierten PBMCs wurden zusätzliche Proliferationsassays durchgeführt. Die
Chemokinproduktion auf das Antigen wurde mittels MIP-1-ELISA gemessen.
Nach der ersten Immunisierung wurde eine signifikante Zunahme der Immunantwort auf die Gesamtvakzine, np24 und rp24
beobachtet. Im Gegensatz dazu veränderte sich die Immunantwort auf das Candida-Antigen nicht signifikant. Nach
weiteren
Injektionen wurde die Immunantwort nicht weiter stimuliert, blieb jedoch signifikant über den Ausgangswerten vor
Behandlungsbeginn. Die antigenspezifische Flow-Zytometrie zeigte, daß die CD4, CD8 und NK-Zell-Phänotypen die - in
Reaktion auf das Antigen - hauptsächlich proliferierenden Zellen waren. Es konnte nach Immunisierung ferner eine durch
HIV-Antigen stimulierte Produktion von Beta-Chemokinen (MIP-1 beta) beobachtet werden.
Die Autoren schlossen: "Die Ergebnisse der Studie legen nahe, daß eine mangelhafte HIV-1-spezifische Immunantwort
mit
einer HIV-1-spezifischenen Immuntherapie bei chronischer HIV-Infektion verbessert werden kann."

Die Ergebnisse dieser Studie waren wesentlicher Auslöser für eine große, 2.500 Patienten umfassende Phase III-Studie
zu
Remune plus HAART in den USA. Diese Studie ist beendet. Wie bereits berichtet, hat das DSMB im März die
Auswertung der
Studie bis zur vollständigen Vorlage aller virologischer Untersuchungsergebnisse ausgesetzt. Bis heute ist von diesem
vielversprechenden Ansatz leider nichts mehr zu hören gewesen. Stattdessen schießen überall Therapiepause-Studien
aus
dem Boden, die mit dem gleichen Hintergrund quasi eine Autovakzinationsstrategie fahren - obwohl die Auswertung aller
bisher
zu diesem Ansatz durchgeführten kleinen Studien zeigte, daß das Absetzen der HAART alleine nur zum Wiederanstieg
der
Viruslast führt. Es ist im Interesse aller Beteiligten zu hoffen, daß die Ergebnisse der Phase III-Studie schnellstmöglich
publiziert werden.

zu
Remune plus HAART in den USA. Diese Studie ist beendet. Wie bereits berichtet, hat das DSMB im März die
Auswertung der
Studie bis zur vollständigen Vorlage aller virologischer Untersuchungsergebnisse ausgesetzt. Bis heute ist von diesem
vielversprechenden Ansatz leider nichts mehr zu hören gewesen. Stattdessen schießen überall Therapiepause-Studien
aus
dem Boden, die mit dem gleichen Hintergrund quasi eine Autovakzinationsstrategie fahren - obwohl die Auswertung aller
bisher
zu diesem Ansatz durchgeführten kleinen Studien zeigte, daß das Absetzen der HAART alleine nur zum Wiederanstieg
der
Viruslast führt. Es ist im Interesse aller Beteiligten zu hoffen, daß die Ergebnisse der Phase III-Studie schnellstmöglich

completion of the merger, the current officers and directors of Immune

Meiner Meinung nach ist das eine Konsoldierung aufgrund von
Unschlüssigkeit, d.h. die Studie soll zwar erfolgreich sein
aber es gibt noch keine Ergebnisse die die Meldung bestätigen.
Unterstützung liegt ca. bei 11,4 $! Würde die Chance nutzen und
nachkaufen. Denke ab nächster Woche könnte es wieder aufwärts gehen.
Hoffe ein bißchen geholfen zu haben.

hab mich, wie wahrscheinlich mehrere von euch, etwas zu früh gefreut. da sich für mich jedoch am grundton nichts geändert hat, habe ich nun wieder grund mich zu freuen, ich werd halt heut nochmal einkaufen!
intraday für die nächsten tage bis in den bereich bis 10,5 - 11 wäre nicht tragisch.
schlußkurse sollten allerdings ( bei ca. 11,5-12) drüber liegen.

On March 27, at an AIDS medical conference in Israel, the company presented encouraging data from pre-clinical trials of a potential HIV vaccine consisting of Remune and a novel immunostimulatory sequence of DNA. Remune is the company`s own HIV immunogen, which is supposed to slow
the replication of HIV-1 through increased immunologic control over infected cells. Remune may work with currently available antiviral drugs as a complementary treatment for HIV infection. They believe that while
antiviral drugs interrupt the reproduction process of HIV within infected cells, Remune stimulates the immune systems to destroy HIV-infected cells.
This vaccine appears to strengthen the immune response in animals, including both T-helper and T-killer cell activity that may be involved in the fortification against HIV infection.
Immune Response currently has eight products in their pipeline, but nothing has been commercialized as of yet. Remune, discussed above and currently in Phase III testing, is the furthest along. Autoimmune
products for rheumatoid arthritis, Psoriasis, and Multiple Sclerosis are in Phase I/II and Phase II. Colon cancer and brain cancer therapies are in Phase II testing, and a gene therapy program for Hepatitis and Hemophilia are in pre-clinical stages.
Today`s news is encouraging, but revenues from this venture are a long way off. Remune revenues, however, are potentially much closer, but are not guaranteed to be substantial.
In late February, they announced positive results from a 297 patient, 40 week study, in Thailand. A company licensee, Trinity Medical Group Co., Ltd., administered the trial. During the trial, patients were randomized
to receive Remune or placebo by injection at weeks 0,12, 24, and 36. The primary endpoint was a change in CD4 helper T cell counts, but HIV-specific immune response and viral load were also measured.
The patients treated with Remune experienced an average of an 84 cell increase in CD4 count, which was significantly greater than the placebo group at p<0.05. Secondary endpoints of humoral and cell-mediated immune responses directed at HIV were also higher in Remune patients than in the control arm (p<0.05). However, there was not a statistically significant difference in viral load between the two groups during the 40-week trial, and that is the only FDA acceptable endpoint for US approval. Remune does
not eradicate the virus, but according to this trial, it may control it. Furthermore, it can help to restore the function of the immune system in HIV infected patients.
Remune has been submitted to the Thai Minister of Public Health for commercial approval. There are an estimated 1.4 million HIV-infected people in Thailand. The treatments would cost about $100 for one year.
The larger opportunity for Remune is in the US and Europe. Approval in these markets remain at least two years away and the first Phase III trial data is not expected to be available before mid-2001. Warner-Lambert is administering the trial. In the US, Remune must be used in conjunction with other treatments, but in some countries - such as Thailand - it may be used on its own.
First Security Van Kasper analysts Alan Auerbach rates the company a Strong Buy, and believes that the company`s current market cap does not fully reflect the full breath of the its pipeline. "We continue to expect several announcements this quarter", Auerbach wrote, "that we believe will give the company`s clinical and pre-clinical programs more exposure, and thus raise its profile with investors."
Immune Response Corporation has a market cap of $333 million, and they have $23 million in cash with very little debt. Revenues last year totaled $20.8 million, and institutions control 20% of the float. They
have traded in a 52 week range of 2.56 to 19.75.

Immune Response Corporation (IMNR $13) has been in the news recently because of recent advances in their work in the HIV arena. The California based biopharmaceutical company is developing immune-based therapies to
induce specific T cell responses for the treatment of HIV, autoimmune diseases and cancer. Today, they released a statement announcing their intentions to pursue a preventive HIV vaccine.

Short term I think we can see IMNR (from technicals) reaching back into the high teens. Today biotech index got creamed ...down 6.5% the nasdaq lost 162 and IMNR held on for some time. Shorts are waiting longer in the day to make a decision...things will turn around. I think we all know about the fundamentals... I hate to say be patient...but the public will jump on good information...it just depends on how it is presented...especially if IMNR was smart enough to say how much money they could potentially make from their hard work.
Not sure if anyone looked at time of sales but the market makers knew what was going on with biotechs today...12k buys, 200 shares sold...price drops...another 8k buys, 100 shares...price drops...and on and on...we got fucked by the market makers...
They are collecting positions, because they are getting ready for a run up...its the old philosophy buy low...sell high. The shorts are getting defensive.

Die downpusherei ist zu Ende. Die Meinung ist überwiegend positiv.
Was fehlt sind konkrete Zahlen was IR denn mal eines Tages
mit Remune verdienen wird.
Im generellen sollte man sich darüber im klaren sein, daß
IR eine heiße Spekulation ist und die Biotechs derzeit
verprügelt werden. Das wird sich in Kürze ändern.
IR hat über
20% institutionelle Anleger u.a. American Express.
Das gibt eine gewisse Sicherheit nach unten.
Noch was zur Charttechnik. Halten die 12$ gehts ab nach 21$.
