US panel not convinced spray flu vaccine safe

GAITHERSBURG, Md., July 27 (Reuters) - A U.S. advisory panel said on Friday it was not convinced that an experimental influenza vaccine sprayed into the nose was safe enough to win government approval.

The decision by a Food and Drug Administration committee prompted maker Aviron (NasdaqNM:AVIR - news) to announce it could not bring the vaccine to the market in time for the upcoming flu season.

The product, called FluMist, could offer a pain-free alternative to annual flu shots. The FDA panel ruled that studies in 24,000 people showed the vaccine was effective in preventing the illness in kids as young as one year old and adults up to age 64.

But the panel voted 10-4 that information presented during a two-day meeting was insufficient to show FluMist was safe. Many members said further data might ease their concerns and they urged Aviron to continue development.

``I do think this vaccine will turn out to be safe but from the data we`ve seen ... I do think we need more information,`` said Judith Goldberg, director of biostatistics at New York University School of Medicine.

FluMist is key to the future of Aviron, which is trying to get its first product on the market. Industry analysts predict FluMist, which American Home Products Corp. (NYSE:AHP - news) would co-market, could generate $1 billion in annual sales in a few years if it gets FDA clearance.

Aviron Chairman and Chief Executive Boyd Clarke told reporters after the panel vote that, ``we will not be able to launch this vaccine in time to participate in the 2001-2002 flu season.``

The company had hoped to have limited quantities available for the upcoming flu season, which usually begins in the fall and peaks in the winter.

``Our primary objective still remains securing licensure of this vaccine in order to participate in a full launch for FluMist in 2002,`` Clarke said.

Aviron is continuing to provide data on FluMist`s safety to the FDA, he added.

About 70 million people get flu shots each year to prevent the fever, aches and misery the illness brings. Flu complications kill 20,000 Americans each year and send 100,000 people to the hospital.

FluMist contains live, weakened flu strains that are sprayed up the nose to stimulate immunity against the virus at a place where it enters the body. Flu shots are made with killed flu viruses.

Panel members said they were concerned about a lack of information on how FluMist affected people with asthma or weak immune systems, as well as children who were given other immunizations at the same time.

Aviron said FluMist`s most common side effects were mild, such as a runny nose.

In one clinical trial, FluMist provided children with 95 percent protection from feverish illnesses and 97 percent protection from ear infections, a common flu complication.

Panelists said FluMist`s easy administration made it appealing for use in children, a group hard-hit by the flu, but not commonly vaccinated with shots.

``As a pediatrician, I am really very excited about the possibility of having an effective, relatively safe vaccine with ease of administration,`` Dr. Walter Faggett said.

Studies in adults were less successful, failing to show that FluMist reduced feverish illness during a flu outbreak compared with a placebo spray. Data did show FluMist cut rates of severe illness and upper respiratory infections.

Company studies did not compare FluMist to a flu shot.

Trading of Aviron shares was halted during the panel meeting. Aviron is based in Mountain View, California.

Mir ist absolut unklar wie BB in die investieren konnte, die Abneigung der FDA gegen diese Art der Virusbekämpfung via selber "aktiven Impfungen". Besonders bei Produkten die auch bei Kindern eingesetzt werden sollten. Grosse Hedge-Funds haben seit längerem darauf spekuliert, dies ist bekannt. Und sie haben gewonnen...

Also Scios hatte Avalon auch kurz vor dem FDA Termin auf Sell geschaltet, Natrecor ist aber trotzdem durchgekommen.

Und bei Scheitern von Symlin/Amylin lagen sie mit ihren öffentlich bekanntgegebenen Argumenten völlig falsch, sondern die Geschichte scheiterte an für Anleger völlig neuen Facts (Hypoglykämie). Vielleicht haben die sich auch eine Quelle in der FDA gekauft.
Ich habe der SEC erst einmal einen netten Brief geschrieben.

Es ist schon erstaunlich (und zugleich traurig) mitanzuschauen, wie sich in diesem Board jetzt wieder alle sogenannten Experten gegenseitig hochschaukeln und in kollektive Panik verfallen. Sicherlich, es wird am Montag einen tiefen Einschnitt im Kursverlauf von Aviron geben. Doch nun ist es eh zu spät. Seine Anteile wird man am Montag nur zu Discountpreisen losschlagen können.

Wenn man sich die Kommentare des FDA-Panels anschaut und sich die Mühe macht, ein wenig zwischen den Zeilen zu lesen, so ist es meines Erachtens klüger, das "Tal der Tränen" auszusitzen. Anbei einige Auszüge des FDA-Statements:

- "Most panelists were excited about the possibility of having the first nasal- spray vaccine approved in the U.S."
- "Most panelists who said the safety of FluMist wasn`t established also said the vaccine will likely prove effective once all of the data is fully analyzed and when ongoing studies are completed."
- "It is likely to prove safe, but the data and analysis is not complete."
- "He (Aviron Chairman and Chief Executive Boyd Clarke) added Aviron has much of the data the panel requested and that the company is in the process of analyzing it before submission to the FDA."
- "The efficacy data was really quite strong," said panelist Dr. Nancy Cox chief of the Centers for Disease Control and Prevention`s Influenza Branch. "It would be nice to have additional data on younger children.""
- "... he (Panelist Dr. David S. Stephens) said FluMist is a crucial product. "I do think this is an important vaccine," Dr. Stephens added. "It is an important breakthrough.""
- "Dr. Sharon R. Seiler, an analyst who follows Aviron for Punk, Ziegel & Co., said the panel`s vote was "clearly a setback." But, she said most panelists want more data, and thinks that data will sway them toward approval."

Fazit: Da es sich bei FluMist um ein Medikament mit einem geschätzen Jahresumsatz in Milliardenhöhe handelt und das Unternehmen bei einer derzeitigen Marktkapitalisierung von ca. 1,2 MRD. US-Dollar plant, die Zulassung für FluMist nunmehr rechtzeitig für die Grippesaison 2002/03 zu erhalten, ist sicherlich nicht mit Kurzsturz auf Dauer zu rechnen. Montag wäre meines Erachtens der richtige Zeitpunkt, sein Kontingent an Aviron-Aktien aufzustocken.

Aviron`s Flu Vaccine Fails to Win FDA Panel Backing (Update3)

Gaithersburg, Maryland July 27 (Bloomberg) -- Aviron didn`t prove that its FluMist nasal-spray influenza vaccine was safe, a U.S. Food and Drug Administration expert panel said.

The advisers voted 10-4 to recommend that the FDA reject the drug. The agency usually follows the advice of its expert panels. FluMist would be the company`s first product on the market, as well as the first needle-free flu vaccine.

Panel members expressed concern about the lack of information on a possible link between FluMist and pneumonia. Aviron made no systematic effort to categorize or analyze 37 cases of pneumonia among people who took FluMist, compared with 12 cases in the placebo group, FDA reviewer Chrisanna Mink said.

``I think one big problem is our discomfort with the pneumonia issue,`` said panel member Kathryn Edwards, a pediatrics professor at Vanderbilt University. She voted to recommend rejection.

Aviron Chief Executive Boyd Clarke said the company still plans to meet its primary objective of introducing the vaccine beginning in the 2002 flu season. The vote ensures that Aviron will miss what he called a secondary goal, which was to introduce the product on a small scale in the next flu season.

``We will not be able to launch this vaccine to participate in the 2001-2002 flu season,`` Clarke said.

Months, Not Years

Earlier, the panel voted narrowly to say that the vaccine was effective in preventing the flu in adults and children.

Still, the lack of data on the interaction between children`s vaccines and FluMist appeared to trouble panel members. Children under the age of 5 often receive as many as 15 doses of other vaccines.

Panel members did appear encouraging about the potential for eventual approval of the vaccine, if additional information was supplied.

``I think since we last heard about this vaccine the progress has been marvelous,`` said Robert Daum, a pediatrics professor at the University of Chicago.

Among the information the panel asked for was an analysis of an already-complete study, said Harry Greenberg, Aviron`s chief scientific officer.

``It`s a large amount of work but not a large amount of time,`` Greenberg said. ``My hope would be that component could be measured in months, not in years -- and not in lots of months.``

The decision is a blow to Aviron, which had hoped to manufacture limited quantities of the product for the upcoming flu season. Investors have been anticipating the product since 1998 and analysts expect the drug could yield about $500 million per year in sales if approved.

Mountain View, California-based Aviron is developing the product with American Home Products Corp.

Manufacturing problems at some vaccine makers have contributed to shortages in the flu serum in recent years. About 20,000 people die from complications of the flu each year, according to the U.S. Centers for Disease Control and Prevention.

The experimental vaccine is also novel in that it comes from a live virus. Traditional flu vaccines are made from dead germs.

Die Panikmacher haben eine Kleinigkeit übersehen:
Die Gutachter haben nicht gesagt, dass FluMist NICHT sicher ist. Sie haben nur gesagt,
dass aufgrund der derzeit nur unvollständig ausgewerteten, von Aviron vorgelegten
Sicherheitsdaten, nicht entschieden werden kann, ob der Impfstoff sicher ist.
Die Experten haben eine weitere Auswertung der Daten und eine Vorlage zu einem späteren
Zeitpunkt verlangt.
Sicher ist, dass sich die Zulassung verzögert. Nicht sicher ist, dass FluMist NICHT zugelassen wird.
Ich denke, dass der Kurs bis zur endgültigen Entscheidung im Bereich 25-30$ pendelt.
Nach dem Entscheid durch die FDA wird es heftige Kursausschläge geben. Die Richtung wird von der
FDA-Entscheidung abhängen.

Das ein Zulassungsgremium einfach sagt, dass ein Medikament nicht sicher ist, dies habe ich noch nie erlebt. Wenn schwere eindeutige Sicherheitsbedenken exisitieren, dann wird auch keine Zulassung beantragt.
Dann kommt in der Regel die Bemerkung dazu, dass zusätzliche Daten her müssen.
Vielleicht -ich habe mich nicht kundig gemacht- legen damit die Ärzte genau den Finger auf die Wunde. Wie sieht es mit abgeschwächten Viren aus bei Personen mit sich entwickelnden Immunsystem oder geschwächtem Immunsystem? Vielleicht hat sich Aviron bewusst bisher davor gedrückt, weil sie unangenehme Nachrichten befürchteten.
Für Amylins Symlin, in völlig identischer Lage, ist die Sache jedenfalls noch keinesfalls gegessen.

Aviron hat ja die Zulassung nicht zum ersten mal beantragt. Es ist jedes Jahr das selbe Trauerspiel. Die Wirksamkeit steht ausser Zweifel aber die Sicherheit nicht. Langsam müssten doch die Verantwortlichen die gewünschten Daten liefern können. Oder auch nicht, dann kann man aber FluMist auch vergessen.

Ich finde in der Historie nur Probleme mit der sterilen Produktion der Impfung. Die Zulassung war damals aber deshalb nicht beantragt worden.

Schön das Aviron auch ohne Deiner qualifizierten Bemerkung sich erholen konnte.

Ich denke das die Ergebniss für ein erneutes vorsprechen bei FDA in kürze vorliegen könnten.

"Positiv" ist zu bewerten das es bei dem angeblichen "Konkurenzprodukt" einer Schweizer Firma, herbe Probleme gab.

Ich sehe AVIRON als ein Langfristinvestment.FLUMIST stellt im Moment die Basis für den weiteren Verlauf dieser Firma dar.

Aviron entwickelte sich in den letzten zwei Wochen hervorragend. In einem Interview mit BörseOnline bestätigte der Fondsmanager die guten Produktqualitäten seitens FLUMIST.
Ich gehe von einer Stellungsnahme seitens des Management bezüglich der weiteren Studien für den kommenden Monat aus.
Bei positiver Meldung dürfte sich der Kurs dann endgültig stabilisieren.
Interessant ist auch Aviron zweites Produkt (Eppstein-Barr) welches sich schon in der fortgeschrittenen zweiten Phase befindet.
