Biib answered the question straight out, no hesitation. Tysabri did not cause PML.
Followed by the statement that the 3 people were immune surpressed.
But Tysabri was not the cause.
FDA must know this, if BIIB makes the statement at the conference.

As expected, no surprising and/or earthshattering news was presented. However, here are a few positives I picked up from listening in:
1) There was a major emphasis on the ALZ programs. Emphasized the fact that EFFICACY was observed in the AN1792 study. Emphasized that in ALZ, ELN isn`t just working on a program, but a SERIES of programs.
2) After not mentioning drug delivery in last week`s conference, hit the subject hard in this one. The highlights - mentioned that there are "many many" deal discussions ongoing at this time, and in reference to DD deals, specifically said, "The Sky`s the Limit."
3) Mentioned that they are still awaiting word from the FDA on an anticipated AC, and ELN/BIIB will announce to the public as soon as they get word from the FDA. There are "parallel discussions" ongoing with both the FDA (US) and EMEA (Europe) regarding the launch & relaunch of Tysabri. Regarding the potential number of Tysabri patients, KM mentioned that "nothing has changed" since a year ago (think about that for one sec....).
4) "Lots of things" going on in Parkinsons. "Lots of things" going on in the "innovative" labs in South San Francisco.
A couple of personal observations:
- He spoke of things that would be included in Tysabri`s "upcoming new label." This suggests a couple of things: one, KM is still convinced that Tysabri`s return is WHEN not IF; and two, substantial discussions on labelling have already occurred.
- Overall, it certainly at least SEEMED like KM talked about Elan as if it will continue to be an independent entity. There also seemed a sense of confidence that, with Tysabri`s imminent return, increasing nano royalties, a series of ALZ programs, and a number of other "innovative" things coming down the pipe, should ELN choose to remain independent, it could be a pretty danged successful entity.
- For those who are complaining that KM did not orate like Martin Luther King, keep in mind that he`s likely had multiple meetings with numerous investors yesterday and today, and these will continue thru tonight and tomorrow. Importantly, there was a lot of meat in today`s presentation, IMO.
Overall, a good and solid presentation IMO.

1- He will take away most of the financial burden weighing down the co.
2- He will get Tysabri thru the FDA & into the market.
3- He will grow nantech & it will all fall to the bottom line.
4- He will get the Alz. drug on the market as it already has shown efficacy.
5- He will make us all rich.
6- He doesnt need 6 days ...he can do it all in 4 easy steps.
7- With his positive approach to creating a biotech giant who am I to argue the point.

First, the presentation room was SRO. Prob 400 people in attendance. You heard the talk and saw the slides...
Breakout session not webcast. Smaller room, approx 100-120 people. Packed, SRO. About 90% of Q`s were about Tysabri and PML risk. They wanted reassurance about JC Virus, PML risk, etc. Lars mentioned that there are 8 drugs which cause PML...silence...They wanted more info about PML risk...Lars told them, it`s a matter of looking also at the benefits of Tysabri. He said in a population of 100,000 MS`ers, there would be 50,000 fewer relapses if they were using Ty...still silence(not dumbfounded silence, just DUMB silence)...then more Q`s about PML and risk...DUH!!!!
In the end some not very bright "analysts" asking the same dumb Q`s, trying to figure a way to see past the PML risk while not realizing what Lars was telling them.
One person asked if closing the 55 JV`s posed a risk to Elan`s development...she must have been about 3 years late to the Elan story....
A few Q`s about AD program but nothing worth reporting...kinda dumb Q`s and responses were in line...
A Q about "profitability". Shane said that they guided breakeven by Q4 and that by Q3 they were only $5-6M SHORT of breakeven. Shane essentially said, we`ll let you know how we did in a month or so...CLEARLY HE AND KM IMPLIED that they will achieve this objective.
IN all, it was a pretty dull Q session with equally dull people. Too bad. There is such a good story to hear and they just didn`t ask...
KM and JM both made it abundantly clear that an Advisory Committee WILL BE CALLED. Probably early March per KM`s presentation.
Nice close today! Think I`ll stay on vacation!

(AFX UK Focus) 2006-01-11 06:11 GMT:
Biogen: FDA to rule on Tysabri by end of March
Article layout: raw
SAN FRANCISCO (AFX) -- Executives for Biogen Idec said Tuesday that they expect the U.S. Food and Drug Administration to make a decision by the end of March to clear the market return of the company`s recalled drug, Tysabri.
In a presentation before investors at the annual J.P. Morgan Healthcare Conference, Biogen executives said that the FDA is scheduled to rule by March`s end on whether to approve new labeling for Tysabri that would warn the drug has been linked to an extremely rare but deadly brain disease called PML. Tysabri was approved in late November 2004 by the FDA for the treatment of multiple sclerosis.
Biogen and partner Elan Pharmaceuticals voluntarily pulled Tysabri from the market in February 2005 after three cases of PML were reported. Two of the patients died. About 3,000 patients had taken the drug before it was recalled. The drug`s recall surprised both medical experts and Wall Street analysts, many of whom had hailed Tysabri as a wonder drug in the treatment of MS. Industry watchers had predicted that Tysabri, which appeared to be more effective and better tolerated than existing MS treatments, would greatly expand the market for MS drugs. Currently, many MS sufferers eschew drug treatment due to the unpleasant side effects of the drugs. Since that time, Biogen and Elan have been conducting an exhaustive safety review of the drug to determine how the drug can be mostly safely prescribed to MS patients. The companies filed last fall to have the drug approved with a new label that discusses the risks of contracting PML, a disease that generally occurs in patients whose immune systems are severely compromised. According to Biogen Chief Executive Jim Mullen, the new label will dictate that Tysabri should only be used "with great caution" by people whose immune systems are already comprised, such as with immunosuppressant drugs. Mullen reiterated that the company expects to be able to have the drug back on the market by midyear. He added that Biogen was in discussions with health insurers about how the new labeling will affect coverage. Biogen`s executive president for development, Burt Adelman, added that the label also will contain information supporting Tysabri`s unusually high effectiveness in slowing down the progression of MS. In addition, Adelman noted that many of the leading drugs used to treat MS can also cause dangerous side effects. The one great exception, he said, is a group of drugs known as interferons, which includes Biogen`s widely prescribed Avonex. Tysabri had been considered to be superior to interferons because it did not cause the flu-like symptoms associated with those drugs and appeared to be far more effective. Mullen said that the company expects to have the FDA hold a special advisory-panel meeting on the matter. He added that a major topic of the meeting will likely be what sort of language should be included on the label about diagnosing and treating PML in its earliest stages. The FDA has not yet contacted Biogen about scheduling such a meeting, according to Mullen. Biogen`s chief financial officer, Peter Kellogg, declined to speculate on what sales of a newly-launched Tysabri might look like. "Until we all know the new label, we can`t estimate," he said. Prior to its recall, analysts had predicted that Tysabri could have peak sales upward of $2 billion a year. This story was supplied by MarketWatch. For further information see www.marketwatch.com.

I think that KM must have a soft spot for the street. After all of the abuse he and ELN suffered last year from the hands of US research analysts, he continues to telegraph reasons that should allow the anlayst community to do an about face that allows them to save face. He`s been doing this for three months now, and the latest message surrounds the little talked about drug technology business.
It is clear to me that within three years, this will likely grow to a 1 billion $ revenue stream. And the valuation metric on this high margin business is off-the-chart relative to the other businesses. If a conservative metric is to value a drug company at 4-6 times revenues, then the royalty business has to be twice that. As KM said, the "sky is the limit", with 50 billion in potential. If ELN gets 40% of that market (very acheivable based on ELN`s best of breed, proven status), and you assume 5% royalties, the 1 billion target on revenue is met.
So, on the DD business alone, ELN is worth eight billion.
Which analyst will use such logic is unclear. They are so fixated on Ty that they can`t seem to digest that ELN is not a `one trick pony`. But here`s their opportunity to wipe the egg of their faces, and get on with things.

The word has been spread thru an interview with
the Irish Press and now thru the JPMorgan Conference.
Powerful would be a good word to describe it. It has powerful "rewards" for a risk that is being minimized at the present PPS.
Look at these possibilities
Blockbuster status for Tysabri in a Global Market
Commercializing a few products in the Alzheimer arena
Continued explosion in the EDT Business
Expansion of a very lucrative Hospital products Division.
A Parkinson Therapy
Tysabri 2
Collaborations with Labs and Learning Institutions
from around the World.
The number of potential businesses with "powerful"
cash cow implications allows Mngt. a great deal of flexibility in planning its` assailt on the Profit and Loss Statement.
So far, this Mngt. Team has shown it can handle the Balance Sheet rather well. This is the "healthiest"this Corpuration has ever been.
The risk you now have to assess is your trust in this Mngt. Team to execute the Business Plan. Their ability to produce a Profit and Loss Statement that shows growth in both Top Line and Bottom Line. That, ultimately, will determine Elans` PPS .
Powerful enough, in my opinion, to

=DJ Biogen: FDA To Rule On Tysabri By End Of March >BIIB
.
By Val Brickates Kennedy
SAN FRANCISCO (Dow Jones)--Executives for Biogen Idec Inc. (BIIB) said Tuesday that they expect the U.S. Food and Drug Administration to make a decision by the end of March to clear the market return of the company`s recalled drug, Tysabri.
In a presentation before investors at the annual JPMorgan Healthcare Conference, Biogen executives said that the FDA is scheduled to rule by March`s end on whether to approve new labeling for Tysabri that would warn the drug has been linked to an extremely rare but deadly brain disease called PML. Tysabri was approved in late November 2004 by the FDA for the treatment of multiple sclerosis.
Biogen and partner Elan Corp. (ELN) voluntarily pulled Tysabri from the market in February 2005 after three cases of PML were reported.
In addition, Adelman noted that many of the leading drugs used to treat MS also can cause dangerous side effects. The one great exception, he said, is a group of drugs known as interferons, which includes Biogen`s widely prescribed Avonex.
Tysabri had been considered to be superior to interferons because it did not cause the flulike symptoms associated with those drugs and appeared to be far more effective.
Mullen said that the company expects to have the FDA hold a special advisory panel meeting on the matter. He added that a major topic of the meeting likely will be what sort of language should be included on the label about diagnosing and treating PML in its earliest stages. The FDA has not yet contacted Biogen about scheduling such a meeting, according to Mullen.
Biogen`s chief financial officer, Peter Kellogg, declined to speculate on what sales of a newly launched Tysabri might look like. "Until we all know the new label, we can`t estimate," he said.
Prior to its recall, analysts had predicted that Tysabri could have peak sales upward of $2 billion a year.

Depending on the Alz results ELN could really start to go up then

not seen anything yet. ELN was trading much higher previous to our original approval. Today`s action simply suggests we are right in our beliefs. Call it pumping, call it luck, call it puppet speak, does not matter. Many here, and you know whom you are, have enhanced your financial situation tremendously over this past year or so. Those paying attention are running on confidence in the Corporate structure as it stands now. While there is a bet on an AC being convened, or not, it diminishes with each passing day. Any retreat on that news will quickly fill back. KM has spoken. He did Elan Corp. proud. He demonstrated to the world ELN is for real again. That is what the street needed to hear. Management and all employees of ELN Corp. should be proud of what they have accomplished this past year or so. The hurdles before them were large. They leaped them handily. Be satisfied knowing KM and Company are worthy of your continued support. Our hats should be off to them as well.
Our partner in MS, Biogen, while understandably suspiciously motivated, does understand the requirement to fulfill all contractual obligations. It is business for them to protect the Avonex franchise. It is equally as important for them to promote the Tysabri franchise. There is plenty of room for all of us. Let their management prove with deeds their commitment to Tysabri as we go forward. This investor believes they will not dissapoint.
Congrats to all that understand the message and continue to support ELN. These are the the moments in this life to reflect on how fortunate many here really are. We all are guilty of an occasional misgiving with another poster, but at least we seem to mend fences rapidly. Here`s to you and your`s as we celebrate a dandy day at the office. Our plan is coming together, Let`s be prepared for more of the same. Go team ELN!!

I have been around for a long time. I do not post often. We all know many investor make their biggest mistake by selling to soon when they have a winner.
I rode Elan up to $30 and back down to $3 and on paper lost over $1.2 million on/after Black Monday. But you know what, PML could not be seen by anybody, it was a total fluke, and I will get all the $$$ back and more as I now own many more shares.
Think about this. Elan hit $30 when we did not have a claer picture on profits, on the leverage of drug delivery, or more importantly the 4 shots on goal for ALZ programs -- the best ALZ programs in the world -- AAB-001 will work. We also have Parkinsons play now and a robust early stage pipeline that is just coming into the light. When Tysabri comes back and you add on all this great new information, why in the world would you sell now, or at $20 or at $30.
Look at ELN price history and learn. Not too far down the road $30 will be viewed as a very cheap price. I am going to ride this all the way until at least one on the ALZ programs stikes gold.
Elan`s Alzheimer`s drug will dwarf Tysabria`s impact on the price per share. I hope you will think this through and look back at history to see just how high Elan can go before you sell too early. Ride this one to the next peak. The real ride is jusy getting started.
I also owned Elan stock and $65 and that too will be cheap someday within 3 years.
I will hold. Elan will fund my retirement and it will be sweet living! Look back and plan ahead to get ahead now. This one is easy because you already have a blueprint right before you. Elan has never been stronger in it`s history than it is today. Do your own thinking/DD and invest wisely for the long term.
Good luck to all Long!

Approval to restart Phase IIa trial in MS patients
Antisense Therapeutics (AUSTRALIA)
January 12, 2006
Antisense Therapeutics is pleased to report that the Ethics Committee of the University of Essen in Germany has approved the company`s application to restart the Phase IIa trial of its antisense compound, ATL1102, for patients with relapsing remitting multiple sclerosis. The University of Essen is the primary trial site for the Phase IIa clinical trial. The Company now has the requisite approval and regulatory documentation in place to restart the Phase IIa trial at this centre.
Patient enrolment and dosing are expected to commence at the University of Essen in February/March 2006. The other 8 trial centres will, in turn, be initiated in the coming months. The treatment and patient monitoring stages of the 80-patient trial are expected to be completed by the end of 2006 assuming patient recruitment proceeds at the anticipated rate.
The Ethics Committee approval follows the recommendation of a specially convened Medical Advisory Board of independent medical experts to continue the drug`s development.
Antisense CEO Mark Diamond said "We are pleased to have received this approval to progress the further development of our MS drug, which as stated by the Medical Advisory Board, appears to have significant potential as a therapeutic agent in relapsing-remitting MS."
He added that "The Company has been focussed on restarting the Phase IIa trial for ATL1102 in MS patients in an endeavour to restore the shareholder value that has been created over the last 4 years through successful preclinical animal studies and the completion of a Phase 1 clinical trial in humans." ATL1102 is a second-generation antisense inhibitor of an immune system protein called VLA-4 and is designed to block the synthesis of VLA-4 which is known to play a part in both the onset and progression of multiple sclerosis. ATL1102`s mechanism of action in blocking VLA-4 is different to other drugs in development that target VLA-4 (including the monoclonal antibody Tysabri®) and represents a novel therapeutic approach to the treatment of MS.
Multiple sclerosis is a life long chronic disease of the central nervous system which is believed to affect as many as 2.5 million people worldwide. While existing drug sales for this disease were greater than US$4 billion in 2004 there remains a high demand for more effective and better tolerated treatments. The Phase IIa trial of ATL1102 will assess the activity and safety of the drug in MS patients.

5. About Antisense Therapeutics Limited
1. Background re development of ATL1102
After successful results in preclinical animal studies and a Phase I trial in humans, Phase II clinical trials on ATL1102 commenced in December 2004.
This trial was voluntarily halted by Antisense Therapeutics in March 2005 in light of safety issues associated with Biogen Idec and Elan Corporation`s multiple sclerosis drug Tysabri® and in May 2005 the company established and convened an independent Medical Advisory Board (MAB) to consider the potential development paths for ATL1102. Although ATL1102, an antisense inhibitor, is a different drug from Tysabri®, a monoclonal antibody, and thereby works by a different mechanism, both compounds target the same immune system protein (VLA-4).
In August 2005, the MAB unanimously recommended that the company continue the development of ATL1102 in MS and that the Phase IIa trial be restarted with the addition of certain safety parameters to address the potential safety issues related to JC viral activation and the appearance of progressive multifocal leukoencephalopathy (PML) reported in the Tysabri® trials. The directors of Antisense Therapeutics accepted and agreed with this recommendation.
Apart from the addition of the suggested safety parameters, the trial design and clinical assessment objectives remain the same for the Phase IIa trial as reported by the company when this trial was first initiated in December 2004 with the exception that anticipated patient numbers have increased from 60 to 80 as reported in February 2005.
2. Phase IIa study design summary
This study is a multi-centre, randomized, double-blinded, placebo-controlled clinical trial, in approximately 80 patients with relapsing-remitting MS. Patients will receive ATL1102 or placebo over eight weeks. ATL1102 will be delivered by subcutaneous injection on a twice-a-week dosing schedule at a dose of 400 mg per week. The goal of the Phase 2a trial is to obtain preliminary evidence of the drug`s effectiveness which will be evaluated using MRI (magnetic resonance imaging) indices. MRI`s will be conducted at monthly intervals over the 8 week dosing period and at monthly intervals during the 8 week period following completion of dosing.
The trial will be conducted at 9 sites across Germany, which includes the primary trial centre, for which Ethics Committee approval has been received. Applications to initiate these other sites have been filed with the Institutional Review Boards and Ethics Committees of the respective participating centres. Enrolment and dosing will commence at each site as the requisite approvals are received.
3. Update on Tysabri®
Since announcing that they had voluntarily suspended Tysabri from the U.S. market and all ongoing clinical trials based on reports of PML, Biogen Idec (Biogen) and Elan Corporation (Elan) have reported that they have "completed a comprehensive safety evaluation of more than 3,000 Tysabri patients in collaboration with leading experts in PML and neurology. The results of the safety evaluation yielded no new confirmed cases of PML beyond the three previously reported" (17 November 2005 Biogen news release).
On November 17, 2005 Biogen and Elan announced that "the supplemental Biologics License Application (sBLA) for Tysabri® (natalizumab) for the treatment of multiple sclerosis has been accepted and designated for Priority Review by the U.S. Food and Drug Administration (FDA). The FDA grants Priority Review status to products that are considered to be potentially significant therapeutic advancements over existing therapies that address an unmet medical need. Based on the FDA`s designation of Priority Review for Tysabri® in MS, the companies anticipate action by the Agency approximately six months from the submission date [September 26, 2005], rather than 10 months for a standard review."
4. About ATL1102 for MS
ATL1102 is a second generation antisense inhibitor of CD49d, a subunit of VLA-4 (Very Late Antigen-4), and is currently in development as a treatment for MS. In inflammation, white blood cells (leukocytes) move out of the bloodstream into the inflamed tissue, for example, the CNS in MS, and the lung airways in asthma. The inhibition of VLA-4 may prevent white blood cells from entering sites of inflammation, thereby halting progression of the disease. Antisense inhibition of VLA-4 has demonstrated positive effects in a number of animal models of inflammatory disease including MS. While the current market for MS drugs is large (in excess of US$4 Billion per annum) there continues to be a high demand for improved MS therapies. The directors of Antisense Therapeutics believe ATL1102 could have significant commercial potential should clinical investigations show the compound to be suitably safe and effective.

5. About Antisense Therapeutics Limited (ASX:ANP)

ANP`s mission is to create, develop and commercialise novel antisense pharmaceuticals for large unmet markets. The company`s product pipeline includes ATL1102 for Multiple Sclerosis, and ATL1101 for Psoriasis, both of which have advanced to the stage of human clinical trials and ATL1103, a potential treatment for growth (acromegaly) and sight disorders (diabetic retinopathy and wet-are related macular degeneration), which is in pre-clinical development.

Was denkt Ihr, wo würde der Kurs ohne TY stehen ? Würde dieser wieder auf 3$ fallen ?
Macht Ihr Euch keine Gedanken ?

Mein Portfolio: 99% Elan, 1% Mologen

We believe Elan’s share price momentum over the past six weeks has been generated by a
combination of (i) increasing optimism that Tysabri will be approved by the FDA by late March
and back on the market in Q2’06; and (ii) a greater awareness of the progress being made in
Elan’s Alzheimer’s pipeline. On the latter, late 2006 could see AAB-001 move into Phase III on
interim analysis of Phase II data and ACC-001 has just started Phase I. A further string to the
pipeline is a gamma secretase drug trial being conducted by Eli Lilly, which is currently in Phase
II. Elan has the option to co-promote the drug and will receive royalties from the marketed
product. Adjusting our sum of the parts valuation for the company, to reflect increased confidence
in an Alzheimer’s drug reaching the market, gives a value for Elan ex-Tysabri of $5.62 per share.
This implies that the market is currently valuing Elan’s share of Tysabri at $10.57 per share or
$4.4bn (total Tysabri value $8.8bn). This equates to c.4.6x peak sales in our current model. New
blockbuster drugs can trade at 4.5x to 5.0x sales. If the market continues to believe in the
inherent value of Tysabri, backed by a solid Alzheimer’s drug in the pipeline, then there is still
room for some price appreciation and that there will be support at least up to $17.10 (Tysabri
valued at 5x our peak sales forecast of $1.9bn). However, we would advise caution at these
levels, unless there is significant newsflow in the near term that can support the current degree
of optimism. Given that our valuation is less than 15% ahead of the current price, we are
changing our recommendation on the stock to Add with a price target of $17.10, at the upper end
of our price range.

The Market`s Most Overvalued Stocks
Friday January 13, 6:00 am ET
By Mitchell P. Corwin, CFA, CPA
Equity valuations as a whole barely budged in 2005, but the market remains a bit on the pricey side when compared to Morningstar`s collective fair value estimates (To see more on Morningstar`s perspective on market valuations, check out our Market Valuation Graph). The median stock in our coverage universe of 1,700 stocks is now trading at nearly a 10% premium to our estimate of its fair value. However, within our overall coverage universe, there remain plenty of stocks that are trading at significant discounts or premiums to our fair value estimates.
For today`s Stock Strategist, we`ll take a look at some stocks at the high end of the range: those trading at prices more than 2 times our estimate of the company`s underlying fair value. Nearly 100 stocks were pricey enough to pass this screen as of Jan. 11, 2006. All of these stocks are expensive enough that we`d consider selling them, but we narrowed the list a bit further by screening for stocks lacking an economic moat and showing an above-average level of business risk.
A total of 80 stocks made the final cut. Over one fifth of those stocks were in the industrial materials sector and nearly another fifth are classified as energy firms. The common thread running through many of these high-priced stocks is the influence of high energy prices and insatiable demand by investors for anything remotely correlated with the booming price of gold.
In addition to energy and industrials, many other sectors are represented in this list, including business services, financial services, hardware, health care, and media. I thought I`d highlight a few of these larger companies that we think are overvalued for reasons other than having to do with the price of commodities:
Sirius Satellite Radio (NasdaqNM:SIRI - News)
Price/fair value ratio: 302%
From the Analyst Report: "With all of the substitutes to satellite radio available, each installed satellite radio unit will not translate into a self-paying subscriber. Not all consumers will want to spend $150 per year for satellite radio when terrestrial radio, still available through satellite radio hardware, costs nothing. In our opinion, digital music on MP3 players, wireless networks capable of streaming Internet radio into the car, and content through cell phones loom as potential threats, too."
US Airways Group (NYSE:LCC - News)
Price/fair value ratio: 330%
From the Analyst Report: "Let`s get one thing straight: Newlywed US Airways is not a low-cost carrier, no matter what its ticker suggests. Despite its newfound financial stability and some favorable near-term industry trends, US Airways cannot escape its poor business fundamentals. This is not a buy-and-hold stock."
MEMC Electronic Materials (NYSE:WFR - News)
Price/fair value ratio: 328%
From the Analyst Report: "MEMC Electronic Materials is one of many suppliers of silicon wafers, the disks on which semiconductors are fabricated. Despite the firm`s impressive comeback, the industry`s commodity nature and MEMC`s capital intensity still concern us."
LSI Logic (NYSE:LSI - News)
Price/fair value ratio: 204%
From the Analyst Report: "LSI has also struggled to keep pace within its own ASIC market. The firm has steadily been losing share, as its manufacturing capabilities have fallen behind those of rivals like IBM (NYSE:IBM - News). Larger ASIC suppliers, by streamlining their leading-edge manufacturing with the design process, offer a much higher value proposition to customers than LSI, in our view."
Elan ADR (NYSE:ELN - News)
Price/fair value ratio: 533%
From the Analyst Report: "Elan is a broken company desperately trying to fix itself. Its success requires Tysabri`s return to the market, but even then, the company faces an uphill path to become cash flow positive (let alone stay that way). We have incorporated some of the risks into our fair value estimate, but we would still require a hefty margin of safety before investing."

Tysabri 0.01% mortality v Aspirin 0.02%
by: clovus (50/M/Ft.Lauderdale)
Long-Term Sentiment: Strong Buy 01/15/06 11:30 am
Msg: 882830 of 882848
There is a 0.1% risk of contracting PML if you use Tysabri in combination with other immunosuppressive drugs, but so far, no cases of PML have turned up in patients who took Tysabri alone. To put this risk in perspective, a recent study showed that the mortality risk for aspirin is 0.2%. So an MS patient who is comfortable with the risks of taking aspirin, ought to be comfortable with the risk of taking Tysabri.

Saw this article in their weekly online newsletter:

Wall Street Surprise?
"We look for stocks that could double in two to three years," Ken Kam,
who oversees the ground-breaking Marketocracy investor Web site.
Although his top pick for 2006, Elan, has already doubled since he
first recommended it, he still feels that could happen again.
"My top pick for 2006 is Elan (ELN NYSE). In my strategy, the secret
to good stock research is to frame the right questions and then ask
the people best qualified to answer them. At Marketocracy, our m100
members—those with the 100 best investment track records— help us pick
stocks, identify the issues, and frame the right questions. Recently,
I asked the broader Marketocracy community if they could help me
understand the investment puzzle on Elan and Tysabri. I heard back
from members that were physicians, multiple-sclerosis patients,
nurses, lab technicians, industry executives, scientists, etc.
Together, they made me comfortable making Elan the largest position in
the portfolio."
"As background for those who are new to this story, Elan and its
partner, Biogen Idec, had pulled their drug, Tysabri from the market
last February after two patients treated with Tysabri in combination
with Biogen`s Avonex in clinical trials contracted PML, a rare and
frequently fatal disease. ELN fell from $27 to $7 and when a third
case of PML was discovered, it dropped to $3. Since then, an extensive
review of nearly all of the patients showed no additional cases, and
no cases of PML have turned up in patients who took Tysabri alone. In
November, the FDA accepted their application and agreed to give
Tysabri a priority review.
"There are 400,000 MS patients in the US and 300,000 are using
competing drugs. But, those drugs have already failed to help another
100,000 who have stopped using them. For the 100,000 that are not on
any MS medication right now the choice boils down to this: Tysabri can
cut the risk of a relapse by 50%, but there is a 0.1% risk of
contracting PML if using Tysabri in combination with other
immuno-suppressive drugs. To put this risk in perspective, a recent
study showed the mortality risk for aspirin is 0.2%. So an MS patient,
who is comfortable with the risks of taking aspirin, ought to be
comfortable with the risk of taking Tysabri. If 25% of this group
starts to use Tysabri, they would generate over $600 million a year of
sales, 50% of which would go to Elan.
"Citibank, Bank of America, and Merrill Lynch reiterated sell ratings
on Elan recently. All three cited a belief that even if the FDA
reapproves Tysabri in March, MS patients and doctors will not rush to
use the drug until more is known about the risk of PML. But I think
Wall Street is underestimating the potential by forgetting to look at
the other 300,000 MS patients who are now using other drugs. One of
the common side effects of competing drugs is constant flu-like
symptoms. Tysabri does not seem to give MS patients these same
flu-like symptoms.
"For this reason alone, even if Tysabri were no more effective than
other drugs, some of the 300,000 MS patients may switch. MS patients
who are on another drug may have to go off their meds for several
months so their bodies will be clear of the old drug before they can
start on Tysabri. So, one early indicator that Tysabri may be bigger
than Wall Street expects is if you start to hear of MS patients going
off their existing drugs.
"And, that is exactly what I am starting to hear about; MS patients
are going off their meds—against their doctors advice— so that they
will be able to start on Tysabri as soon as its available again. This
tells me that Tysabri sales are going to ramp faster than Wall Street
expects. If 10% of these patients switch to Tysabri, it would mean the
potential US market may be more than twice as big as Wall Street
expects. Good things happen to stocks that surprise Wall Street to the
upside. If I`m right in this assessment, then Wall Street is in for a
big upside surprise."

Elan $16.02 BUY Alzheimer’s Disease Research Gathers Pace
• In a Reuters interview on Friday Wyeth’s head of Neurology discovery said that the
company intends to expand on the number of Alzheimer’s disease products in clinical
development with Elan co-developing one of the Alzheimer’s products expected to
enter the clinic this year.
• Elan currently has two Alzheimer’s disease products in clinical trials with Wyeth as a
co-development partner. AAB-001 (a monoclonal antibody) and ACC-001 (an
immunoconjugate) are in Phase II and Phase I clinical trials respectively with Wyeth
expecting AAB-001 to receive FDA approval “as soon as 2009”. This would appear an
ambitious timeline with Phase II efficacy and safety data on AAB-001 not expected
until later in 2006/early 2007.
• To date no clinical data is available for AAB-001 and we have not factored in a material
contribution from this drug to our current assessment. Should data emerge that this
product can slow Alzheimer`s disease progression, there is scope for a very significant
uplift in valuation.

1. ELN and WYETH holding up conference call the same day on Jan. 31.
2. Is this a coincidence or something more? Especially after WYE, forecasting a product for ALZ on market in 2009?
3. So far we did not hear about scheduled AC for Tysabri. With a final approval due on, or before March 26, 2006, are you really under the belief that an AC will be held?
4. with 68% reduction of relapse episodes and related decrease in disability (every relapse increase disability); the virtually no, or minor adverse events and side effects, are you really expecting that there will still a market for B-INF’s?
5. How do you judge Serono’s selling of the company. Just a capital gain or fear for the future?
6. How a stock like ELN will react when in 2006 in rapid succession you will hear of a “spotless” approval of TY in USA, Europe, Canada and Australasia?
7. What about approval in Crohn?
8. And finally, what about hearing on Jan, 31 that ELN is profitable in 2005-ex Tysabri?
Imagine the future. The future is spectacular.

with this FDA process? I told you last month that if FDA had genuinely classified Tysabri reapproval as priority review (Which is what Dr Banks told LJD) then they could have completed their review by the end of last year.Further, if an AC was deemed necessary I believe the Agency had a responsibility to all Mser`s to make a notification prior to this. You have Mser`s going off other treatments in readiness for taking Tysabri. In essence the Agency is putting these individuals at more risk with their failure to communicate the need or lack there of an AC. It has been eleven months people and a miracle drug is still off the market. How many people have to regress to wheel chairs before the FDA takes action? The problem now days is we are left with very few options when protesting a system that does not work effectively. We have written letters to Congress people, FDA personnel and committee chairs but no one listens. I still say the best way to get their attention is through the media ie a well orchestrated march with the local media present. When I see the needless suffering from FDA`s inaction it makes me sick and ashamed to say I once worked for this organization.Steal, Lindsey et al it is Time!!!! jt- it`s hard for me to fathom this sad Tysabri saga, from its beginning to now.
I could easily say that I`m disillusioned, for sure.
But I can`t say with whom I`m disillusioned, because I don`t have access to all the details of who do what, and to whom, beginning last February.
To be quite honest, I`m much more likely to believe that the culprit is Biogen, and not the FDA.
I believe that Biogen made the decision to withdraw Tysabri from the trials and from the market, not the FDA (and I believe that Elan was not in the room when that decision was made).
And I suspect that the current delay in getting Tysabri back in trials and on the market is due to Biogen`s continued attempts to maximize and prolong sales of Avonex- as stupid as that is, for them for Elan, and most importantly of all, for MS sufferers.
I haven`t trusted Biogen right from my first exposure to them, at a 2004 conference in Boston. Their overly-slick canned presentations, followed by their overly-duplicitious and pathetic handling of only moderately-difficult questions in the Q+A, was so transparent as to be almost comical.
This subject has been discussed and debated ad nauseum on this board, and I hope no one replies to this post.
One of Kelly Martin`s greatest accomplishments, IMHO, has been in dealing with these so-called partners in Cambridge.
He inherited a very difficult legacy from Geaney and his Boys, and the Biogen partnership was a part of it. I`ll never know what Geaney`s marketing options for Antegren might have been, and perhaps this was the lesser of a bunch of evils- but I am sure that it`s a tough assignment.
And it`s probably no easier for the FDA to deal with Biogen than it is for Elan.
I`m convinced that Mullen will be on the outside looking in one of these days. He`s done BIIB shareholders no favors during the past few years, that`s for sure.

I keep saying this over and over again. Do you expect Boob to "sacrafice" a billion dollar drug (Avonex) of which they get 100% revenue for Ty priority of which they share 50% with Elan knowing that Ty is vastly superior to A? I have been in pharma sales for 30 years and have done numerous co-marketing campaigns with other companies and sold their drugs and if it was a drug of "the other company" and it was similar to ours with the same indication I spent 80% on my companys drug because my bonus was signficantly more with MY companys drug. Now remember Boob has total marketing control for TY. See the dilema!!How will Boob sell Ty and Avonex.Use Avonex first then if patient fails regimen switch to Ty?? Remember combo therapy is not a consideration. I would feel a lot better if Elan was in there selling Ty along with Boob.

Ireland`s Elan has altered its licensing agreement with Roche to give the Swiss firm expanded access to a technology designed to improve the performance of drugs.
Elan`s NanoCrystal technology - one of the few elements in the business to be retained as the Irish firm sold off assets to get rid of massive debts - is already winning partnerships among top tier drug companies. The new deal gives Roche the right to apply it to a number of proprietary drug candidates and, in return, Elan will receive development milestones and royalties on sales of any product incorporating the use of technology.
NanoCrystal is designed to improve the solubility of drugs by processing them into tiny nanoparticles - typically less than 1000 nanometers (nm) in diameter, which are produced by milling the drug substance using a proprietary wet milling technique.
These are stabilised against agglomeration by surface adsorption of selected GRAS (Generally Regarded As Safe) stabilisers - with the result that the drug present a much greater surface area for dissolution. The result is an aqueous dispersion of the drug substance that behaves like a solution - a NanoCrystal colloidal dispersion which can be processed into finished dosage forms for all routes of administration. Paul Breen, executive vice president, global services & operations at Elan, said the agreement: "highlights the robustness of the technology and the interest of pharmaceutical companies to use this technology.
Elan already has agreements in place with Johnson & Johnson for the technology, which formed the basis of a new solid-dose formulation of J&J`s immunosuppressant drug Rapamune (sirolimus) in 2003. Rapamune was previously available only as an oral solution in bottles or sachets. The oral solution requires refrigeration storage, and must be mixed with water or orange juice prior to administration. Since then, the NanoCrystal technology has also reached the market in two dosage forms of Merck & Co`s Emend (aprepitant), a drug used for the treatment of nausea and vomiting caused by cancer chemotherapy.

Also flow_solver...Ball bitte flach halten

Imagine this. You`re God forbid, an MSer. You are already totally blind in one eye. You begin having visual acuity problems in your other eye. You are scared to death and immediately ask a caregiver to drive you in to see your specialist. After explaining your new symptom to the specialist they tell you ruh roh, oh my. Oh crap. Boy oh boy. Damn. Oh my. Now it seems you`re sufferring from bilateral optic neuritis! Doc, that means MS in both my eyes now? Yep, sorry but here`s a prescription for that. You tell the doc, thanks. That might might help a little right now but it surely won`t do me much frikken good in the long run will it? So doc can you please get me back on the Tysabri when it comes back?
EDSS getting worse? Yep, it`s already bad enough that many of us don`t have the mobility we once had. When some of us were just hoping it couldn`t get any worse after losing most of our mobility and then suddenly one day you can`t even see where the phuck you are rolling to?
Yep, it sucks being an MSer and many of us do complain about it. The good news is a few of us want our Tysabri back right now, damnit!

ELN is still way undervalued.
Ty will outpace the initial sales estimates which drove ELN over $29 and the Alz program has progressed well past the point where it drove ELN over $60.
As SOON as Ty is back ELN will be stronger than it was when it was in the high $60s.
So....even though ELN has done well lately, IMO it has lots of PPS to make up before it gets near its true value.

Notice period on advisory panel on Tysabri could be imminent
Previous close: 1661c
Analyst: jack.gorman@davy.ie
On the assumption that a 45-day notice period is provided by the FDA to Elan/BiogenIdec for an advisory panel meeting on Tysabri, we would anticipate that this could be provided in the next few days.
The FDA has tentatively scheduled an advisory panel meeting on peripheral and central nervous system drugs for March 7th—there is now only one day set aside for this. Another tentative date of May 17th is also in the FDA calendar, but this would extend past the expected Tysabri timeline under priority review. The March 7th date is the most likely and we understand that the companies have been preparing on the basis of this assumption as well.
The FDA is scheduled to have reviewed and decided on the sBLA for Tysabri by March 26th at the latest. In the interim, fullyear results should provide 2006 guidance and could also provide more detail on the Alzheimers Disease programme (we note with interest that Wyeth has scheduled its own results release for the same day, January 31st).

In the interim, fullyear results should provide 2006 guidance and could also provide more detail on the Alzheimers Disease programme (we note with interest that Wyeth has scheduled its own results release for the same day, January 31st).
